    Notes: Summary Fingerprint (FP) profiles in vacuolated lymphocytes of mucopolysaccharidoses I-H, II, III-A, and III-B are a numerically rare, but possibly consistent finding as they have not been seen in vacuolated lymphocytes of other non-neuronal lipofuscinosis (NCL) lysosomal diseases. Their nosologic significance is not clear, but they may be as non-specific as tubular inclusions in lymphocytes and they are identical to those FP profiles seen in juvenile NCL.

    Notes: Summary The histochemical absence of alkaline phosphatase activity from intramuscular, intrahepatic and intrasplenic vessel walls in a fetus afflicted with biochemically and radiographically proven congenital lethal hypophosphatasia (CLH) indicates that in this disease alkaline phosphatase activity is absent both biochemically and histochemically. It is conceivable that intrafetal skin or muscle biopsies in suspected CLH may be tested for histochemical activity of alkaline phosphatase activity.

    Description / Table of Contents: Summary Rabbit kidney slices of outer zone exhibited a significant decrease of oxygen consumption under the influence of furosemide in a concentration of 1 or 100 εg/ml (Na-content of medium 150 or 350 mval/l), while cortex slices were hardly depressed. This is in favour of the opinion, that furosemide acts by inhibiting active Na reabsorption in the ascending Henle's loop.
    Notes: Zusammenfassung An Kaninchennierenschnitten wurde eine signifikante Abnahme des Sauerstoffverbrauchs der Außenzone bei Furosemid-Konzentration von 1 und 100 εg/ml (Natrium-Konzentration 150 oder 350 mval/l) gemessen, während der Abfall bei Rindenschnitten gering war. Dies wird auf eine Hemmung des aktiven Natriumtransportes durch Furosemid im aufsteigenden Teil der Henleschen Schleife bezogen.

    Description / Table of Contents: Summary A practical, exact and cheep method for the determination of the haematocrit by means of dilution analysis with the aid of51Cr-EDTA is being described. The mean error of ten determinations is not greater than 0.6%. In comparison to haematocrit values obtained by capillary centrifugation these haematocrit values are 5.5% lower. This method may be easily used in cases when haematocrit results ought to be exact for 0.1 vol.-%.
    Notes: Zusammenfassung Mit Hilfe von51Cr-EDTA läßt sich der Hämatokrit durch Isotopenverdünnungsanalyse leicht, billig und exakt bestimmen. Der Fehler des Mittelwertes aus 10 Bestimmungen beträgt 1,5%. Bei Vergleich mit Hämatokritwerten, die durch Zentrifugieren mit einer Capillar-Mikromethode ermittelt worden waren, lag der herkömmliche Hämatokritwert im Mittel um 5,7%, im Einzelfall um 14,9% zu hoch. Da sich mit51Cr-EDTA ein Hämatokritwert cinfach, schnell und in etwa 1,5 ml Blut ermitteln läßt, eignet sich die beschriebene Methode für alle Fragestellungen, bei denen der Hämatokrit auf 0,1 Vol.-% genau bestimmt werden soll.

    Description / Table of Contents: Zusammenfassung 6 gesunde Versuchspersonen nahmen während 3 Tagen je 0,6 mg β-Methyl-Digoxin oral und anschließend während 5 Tagen je 0,3 mg. Nach den ersten 3 Tagen war mit dem Anstieg des Digoxin-Spiegels auf 1,5 ng/ml (1,2·10−9 M) der Na-Gehalt in den Erythrocyten von 4,8 auf 6,6 mval/kg signifikant angestiegen. Gleichzeitig fiel das Mg von 4,1 auf 3,7 mval/kg signifikant ab. Der K-, Ca- und Cl-Gehalt blieben konstant. Unter der Erhaltungsdosis fiel der Digoxin-Spiegel auf 1,2ng/ml (1·10−9 M) ab. Dennoch nahm der Na-Gehalt weiter bis auf 8,3 mval/kg zu, wobei außer Mg jetzt auch K von 84,6 auf 82,0 mval/kg signifikant abfiel. Die Versuche bestätigen die Auffassung von Hoffman, daß das Na-Transpertsystem aus 2 Komponenten besteht, die durch ihre unterschiedliche Glykosidempfindlichkeit differenziert werden können. Sie legen klinisch nahe, daß sich die Erythrocyten-Elektrolyte zur Überwachung einer Digitalistherapie eignen.
    Notes: Summary 6 healthy controls received daily doses of 0.6 mg β-methyl-digoxin for 3 days and further 0.3 mg for the following 5 days each. After the first 3 days sodium content in the red cells increased from 4.8 to 6.6 meq/kg significantly in parallel to the glycoside level of 1.5 ng/ml (1.2×10−9 M). At the same time magnesium in the cells fell significantly from 4.1 to 3.7 meq/kg. There was no change in the concentration of potassium, calcium or chloride at this time. While the glycoside level dropped during the following 5 days under 0.3 mg of methyl-digoxin to 1.2 ng/ml (1×10−9 M), sodium content in the cells increased further up to 8.3 meq/kg and was paralleled by a decrease in potassium content from 84.6 to 82.0 meq/kg. There were no changes in plasma electrolyte concentrations during the experiments. Our results confirm the concept of Hoffman that there are two transport pathways in human red cells which differ basically in their dependencies of the sodium and potassium composition of the external medium and the concentration of glycoside needed to inhibit the transport. Our data evidence that red cell electrolyte contents reflect levels of digoxin. They might be useful to follow up the therapeutical effect of cardiac glycosides.

    Notes: Zusammenfassung Nach der Inkubation von überlebendem Pankreasgewebe der Ratte in einem Medium, dem Alloxan und IPTD beigegeben ist, läßt sich histologisch keine für eine bestimmte Zellart spezifische Schädigung erzielen. Wird dagegen überlebendes Gewebe der Bauchspeicheldrüse in einem Medium mit D 860 inkubiert, dann ist im histologischen Schnitt die Masse der B-Zellen zusammengesintert und zerstört. Diese Versuche, die den Verhältnissen am Lebenden ganz unähnlich sind, sind ungeeignet, eine gezielte betacytotrope Alloxanwirkung darzustellen, zeigen aber andererseits eindeutig die Affinität der Sulfonylharnstoffe für die B-Zellen.
