    Notes: Stable silk fibroin (SF) membranes were prepared by modifying SF with polyethylene glycol diglycidyl ether (PEGDE) for use as enzyme immobilizing matrix. Morphology, structure, and water solubility of the modified silk fibroin (MSF) membrane were investigated. SEM images revealed greater porosity in the MSF membranes. IR spectra confirmed the predominant β-sheet conformation when the PEGDE was greater than 4%. In addition, the MSF membranes were highly insoluble within the pH range 4 10. An amperometric glucose biosensor was initially constructed using glucose oxidase (GOx) immobilized on the MSF membrane, coupled to a Prussian Blue (PB) deposited Au electrode. The response characteristics of the biosensors based on the GOx immobilized onto the 4% and 8% PEGDE modified SF membranes (4P-MSF and 8P-MSF) and the methanol treated SF membrane (MT-SF) were investigated. Amperometric measurements at the applied potential of 0.0 V demonstrated that the glucose biosensor based on the GOx immobilized onto the 4P-MSF membrane exhibited high sensitivity to glucose with a short response time, less than 3 s. Moreover, the stability of the biosensors based on the 4P-MSF membranes was better than those based on the MT-SF membrane after 2 weeks of storage in a phosphate buffer of pH 7.0 at 4 °C. These fundamental electroanalytical features of the sensor are therefore expected to be useful in biosensor application

    Notes: Genetic exchange by natural transformation in Streptococcus pneumoniae occurs in a cell-density dependent process and is initiated by a small extracellular signalling molecule, the competence-stimulating peptide (CSP). comC, the gene for this peptide, has previously been identified and encodes a 44 amino acid pre-peptide that is apparently processed to an active molecule that consists of the C-terminal 17 amino acids. We have sequenced the region adjacent to comC and shown that it is the first gene of an operon, com, consisting of two downstream elements, comD and comE, which encode members of the two-component family of sensor regulators. Null mutants with defects in either comC or comD were transformation deficient and failed to respond to exogenous CSP. A comC mutant did not exhibit any detectable CSP activity, while a comD mutant that contained an intact comC produced minimal CSP activity. In mixed-culture experiments consisting of isogenic pairs of pneumococci (Csp+ and Csp−), we showed that induction of competence by quorum sensing was independent of CSP. Northern analysis showed that com was transcribed as a single polycistronic message, while analysis of strains with transcriptional fusions showed that com was constitutively expressed under conditions that both promoted or repressed the development of competence. Finally, we showed genetically and biochemically a CSP-dependent transcription of rec, a competence-induced locus, and that ComD and ComE are required for this CSP-dependent expression.

    Notes: Low-temperature scanning electron microscopy (LTSEM) is a promising measuring technique for probing the spatial distribution of the superconducting properties of high Tc thin films. In the presented article we have given some experimental observations on the spatial variation of the critical current density of two kinds of high Tc thin films using a LTSEM. The results show that in the polycrystalline TlBaCaCuO film prepared by dc magnetron sputtering, the maximum local critical current density is about 3–4 times greater than the minimum local critical current density. The microstructure analysis implies that the strong inhomogeneous critical current density distribution in the TlBaCaCuO film is due to its granular structures and the existence of several structural phases. For c-axis oriented epitaxial YBaCuO thin film deposited in situ by laser ablation, the results indicate a less inhomogeneous critical current density distribution. The maximum local critical current density is less than 1.5 times the minimum local critical current density in the epitaxial thin film.

    Notes: NdFe10+xMo2−2xTix and NdFe10+xMo2−x (0≤x≤1.0) have been synthesized by using a reduction-diffusion process with superfine precursor particles as starting materials. The easy direction of magnetization of the NdFe10+xMo2−2xTix system has been determined by ac susceptibility and x-ray diffraction of the aligned samples. By comparison with NdFe12+xMo2−x, a magnetic phase diagram of the NdFe10+xMo2−2xTix system has been proposed. © 1998 American Institute of Physics.

    Notes: Using a high Tc superconducting grain boundary Josephson junction, harmonic mixing experiments in the mm waveband were carried out, aiming at as large a harmonic number and as high a signal frequency as possible. The dependencies of intermediate frequency output on dc bias, harmonic number, frequency of local oscillator (LO), and other parameters were carefully studied. Until now, our best result was the mixing between the signal at 95 GHz and the 105th harmonic of LO at about 900 MHz. Preliminary experiments using a high Tc harmonic mixer and phase-locking loop were tried to stabilize the frequency of a mm wave source.

    Notes: We have measured the magnetic hysteresis loops of an epitaxial YBa2Cu3O7−δ thin film using our recently developed device which can provide the field magnitude in the range of 0–1000 Oe and the field sweep rate up to 107 Oe/s. The shape of the hysteresis loop measured changes with the field-sweep rate up to the critical sweep rate; and over the critical sweep rate the ac magnetization reaches its real critical state where magnetization does not change even with a further increase in the field-sweep rate. The critical sweep rate is about 106 Oe/s at 77 K. With the hysteresis loops and Bean model, we have calculated the magnetization critical current density (Jcac) which is consistent with that obtained by I–V measurements. We have also studied flux motion and activation energy under the high sweep rate magnetic field. At temperature 77 K, the velocity of the flux motion is of the order 10 m/s and the pinning energy U0/k is about 339 K which is much smaller than the magnetization decay measurement. © 1997 American Institute of Physics.

    Notes: A hysteresis loop tracer to measure the ac magnetization of high-Tc superconductors in the frequency range of 30 Hz–10 kHz is described, with driving field amplitudes up to 0.1 T. Due to the high frequency of the magnetic field, the hysteresis loops could be displayed on an oscilloscope and their details could be observed. The demagnetization factor, the stability, and the frequency characteristics of the tracer were tested. The ac magnetization of granular high-Tc superconductors was studied as a function of field amplitude and frequency. © 1997 American Institute of Physics.

    Notes: The space-charge neutralization lens (S-C neutralization lens) is investigated from the theoretical point of view. The working parameters and the focal length formulas of the lens are given. According to this article, a S-C neutralization lens has been designed and used as a focusing and transmission unit in the intense beam facility at the Institute of Nuclear Research of Lanzhou University. The experiments show that it has an excellent operating characteristic for focusing intense beams and adjusting optical-path matching.

    Notes: The kinetics of the γδ T-cell response was analysed in the context of the overall haematological response in subjects experimentally infected with sporozoites of Plasmodium falciparum. Numbers of γδ and αβ T cells and NK cells declined markedly during infection to reach minimum values 12–13 days post-infection when the patients were ill. This decline commenced from the beginning of the erythrocytic cycle and well before parasites could be detected microscopically and clinical symptoms developed. Platelet numbers also declined. In vivo activation of γδ T cells was evident with sequential up-regulation of the activation markers CD69 and HLA-DR. γδ T cell numbers were highest after treatment with the majority being CD4−CD8−, HLA-DR+ and showing reduced CD45RA expression. Contrary to some published observations γδ T-cell percentages remained within the normal range. Little evidence of up-regulation of activation or memory markers was observed in the αβ T-cell population. In vitro proliferative responses to malaria antigen which involve γδ T cells were lost as the infection progressed and the lymphocyte count declined but these could be restored with the addition of exogenous IL-2 to cultures. The authors findings are consistent with a protective and/or immunomodulatory role for γδ T cells in malaria.
