    Notes: Summary Characterization of 13 isolates of luminescent bacteria from specimens of Cleidopus gloriamaris has revealed differences between these organisms and previously described species of Photobacterium. Further studies on the biochemical and serological attributes of these and other luminescent bacteria are essential, before ecological studies on the fish-microbe symbiosis are possible.

    Notes: Collisional–radiative (C–R) calculations based on the generalized multithermal equilibrium (GMTE) model are used to find the effects of radiative transfer and diffusion on excited-state populations in a hydrogen plasma. Radiation absorption factors are introduced to study the nonlocal aspects of radiative transfer. Unlike escape factors, the absorption factor can represent the absorption of more radiation than that emitted, and hence provides C–R analysis in strong gradients such as the outer edge of arcs or plasma jets. The Boltzmann temperatures of excited atoms are found not necessarily equal to the electron kinetic temperature, contrary to the usual assumption.

    Notes: In order to better understand the role of antioxidant enzymes in plant stress protection mechanisms, transgenic tobacco (Nicotiana tabacum cv. Xanthi) plants were developed that overexpress both superoxide dismutase (SOD) and ascorbate peroxidase (APX) in chloroplasts. These plants were evaluated for protection against methyl viologen (MV, paraquat)-mediated oxidative damage both in leaf discs and whole plants. Transgenic plants that express either chloroplast-targeted CuZnSOD (C) or MnSOD (M) and APX (A) were developed (referred to as CA plants and AM plants, respectively). These plant lines were crossed to produce plants that express all three transgenes (CMA plants and AMC plants). These plants had higher total APX and SOD activities than non-transgenic (NT) plants and exhibit novel APX and SOD isoenzymes not detected in NT plants. As expected, transgenic plants that expressed single SODs showed levels of protection from MV that were only slightly improved compared to NT plants. The expression of either SOD isoform along with APX led to increased protection while expression of both SODs and APX provided the highest levels of protection against membrane damage in leaf discs and visual symptoms in whole plants.

    Notes: Abstract A spectrophotometric method of determining extractable lipids was compared with a gravimetric method to estimate intracellular polar and non-polar lipids in the oleaginous yeast, Candida curvata. The methods correlated well but the spectrophotometric method was simpler, required less sample, and its accuracy was not affected by non-lipid components.

    Notes: Abstract Vegetable oils promoted mycelial growth ofVolvariella volvacea. Ethyl esters of major components of saponified fatty acids (palmitic, stearic, oleic and linoleic acid) from vegetable oils were stimulatory. The stimulatory effect of these fatty acids varied with concentration and degree of unsaturation; relatively high concentrations being inhibitory. Mycelial growth appears to be promoted by low concentrations of fatty acids. Supplementation of growth medium with sunflower oil altered membrane permeability and this resulted in an increased uptake of glucose. The total mycelial lipids accounted for only 30% of consumed lipids, the remainder being metabolized. The failure of the fungus to adjust the degree of unsaturation in membrane lipids when it was transferred to 0°C may partially explain its susceptibility to chilling injury.

    Notes: Summary A method for the cultivation of Pleurotus sajorcaju on a relatively large scale using cotton waste as substrate has been developed, and the mushroom so obtained has higher protein content than and comparable carbohydrate content to Agaricus bisporus, Volvariella volvacea, Lentinus edodes and Pleurotus ostreatus. The crude fats, ash, energy value, vitamin and mineral contents are lower and yet the differences are not great. The biological efficiency from cotton waste compost is lower than that from straw compost, however, the former has the advantage of giving rather even yield over successive flushes. This mushroom has a high potential to be produced economically on a large scale in Hong Kong.

    Notes: Summary A new general method has been developed for the specific histochemical identification ofO-acyl sugars in any epithelial glycoprotein. These sugars include hexose, 6-deoxyhexose andN-acetylhexosamine with an ester substituenent(s) located on a potentialvicinal diol(s). In the procedure reported [the periodic acid-borohydride reduction-saponification-selective periodate oxidation-borohydride reduction-periodic acid-Schiff (PA-Bh-KOH-PA-Bh*-Bh-PAS) method] the initial PA-Bh treatment rendersvicinal diols located on either sialic acid or neutral sugars PAS unreactive. In the subsequent steps ester substituents are removed from bothO-acyl sugars andO-acyl sialic acids by saponification (KOH), sialic acidvicinal diols are selectively removed by the PA*-Bh sequence andO-acyl sugars are stained with the PAS technique. This method has the advantage that the results are obtained with a single section and the results are either positive or negative. Consequently, it is superior to the three indirect methods investigated because it does not require an observer to compare the intensity or the shade of the staining obtained with serial sections. Using the PA-Bh-KOH-PA*-Bh-PAS method we have demonstrated, for the first time, thatO-acyl sugars occur in the epithelial goblet cell glycoproteins of adult human colon. The effect of the presence ofO-acyl sugars on the interpretation of a number of other methods for the histochemical investigation of glycoproteins is discussed. It is recommended that the results obtained with the PA-Bh-KOH-PA*-Bh-PAS method be evaluated before histochemical procedures for the investigation of neutral sugars andO-acyl sialic acids are selected.
