    Beschreibung / Inhaltsverzeichnis: Summary An in vitro system was developed to test the propensity of rat erythrocytes (RBC) toward phagocytosis by homologous peritoneal macrophages after in vivo aging or after in vitro surface modification. “Old” RBC obtained from erythrocyte populations separated into fractions on the basis of their density were found to be phagocytosed to a significantly greater extent than cells obtained from “young” cell fractions. Neuraminidase treatment of RBC resulted in extensive phagocytosis in comparison to control cells representing the whole population or “old” cells. Galactose oxidase treatment of neuraminidase treated RBC afforded no protection from uptake by macrophages. In contrast, treatment of RBC with neuraminidase immobilized on Sepharose 4B, leading to removal of up to ∼25% of the total membrane sialic acid, did not lead to phagocytosis above control values. These results indicate that extreme caution is necessary in the interpretation of experiments in which neuraminidase is utilized to produce simulated “aged” RBC.
    Notizen: Zusammenfassung Es wurde ein in vitro-System entwickelt, in dem die Tendenz von Rattenerythrozyten (RE) gemessen wurde, entweder nach einer in vivo-Alterung oder einer in vitro-Oberflächenveränderung durch homologe peritoneale Makrophagen phagozytiert zu werden. Wurden „alte“, dichtere RE in Fraktionen auf Basis ihrer Dichte aufgetrennt, so war die Phagozytose von „alten“ RE wesentlich ausgeprägter als jene von „jüngeren“ Zellen. Die Neuraminidase-Behandlung von RE führte zu einer hochgradigen Phagozytose im Vergleich zu den Normalkontrollen der gesamten Zellpopulation oder „alten“ Zellen. Eine Galaktose-Oxydase-Behandlung von neuraminidasebehandelten RE brachte keinen Schutz gegenüber den Makrophagen. Im Gegensatz dazu führte die Behandlung von RE mit Neuraminidase, die an Sepharose-4B festgebunden ist, zwar zu einer Entfernung bis zu ∼25% der gesamten membrangebundenen Neuraminsäure, ergab jedochkeine über den Kontrollwerten liegende Phagozytose. Daraus folgt, da\ man bei der Interpretation von Experimenten mit Neuraminidase sehr vorsichtig sein mu\, wenn das Enzym dafür verwendet werden soll, künstlich „gealterte“ RE zu erzeugen.

    Notizen: Abstract Ulcerative colitis is associated with abnormalities of mucin synthesis and secretion, features that may also be associated with malignant change. It has been shown that South Asians in Britain have a high incidence of ulcerative colitis but a low incidence of colorectal carcinoma compared with their European counterparts. Previous studies have demonstrated changes in colonic mucin sialylation and sulphation in both South Asian and European cases with ulcerative colitis. This was related to disease severity, but changes were also found in quiescent disease. The aim of the present study was to determine glycoconjugate expression in the colon from South Asian cases and to compare results with those from a group of affected Europeans. Glycans were identified in formalin-fixed, paraffin-embedded tissue from 17 South Asian patients with ulcerative colitis and from 11 European patients with a similar degree of colitis, by the application of 10 biotinylated lectins. These were directed against a range of sialyl, fucosyl and 2-deoxy, 2-acetamido-galactosyl sequences, using an avidin--peroxidase revealing system and semiquantitative assessment. The South Asian group showed a reduction in the binding of agglutinins from Sambucus nigra in the apical-membranous region of enterocytes, and a decrease in apical Maackia amurensis agglutinin binding. These results suggest that South Asians with ulcerative colitis show a different distribution of terminal N-acetyl neuraminyl residues, either in their α-2,6 or α-2,3 linkage, compared with their European counterparts. The changes in sialylation observed in European cases compared with normal disease-free control subjects were present in quiescent disease, but were also related to disease activity. Their absence in Asians with ulcerative colitis may imply an inherent, genetically determined variation in this group, which may also play a part in their reduced risk of subsequent malignancy

    Notizen: Summary Two new histochemical procedures for detecting sulphated and non-sulphated sialomucin in colonic mucosa were assessed: the saponification—Alcian Blue pH 1—periodic acid—phenylhydrazine—Schiff method (KOH—AB pH 1—PAPS) and the mild periodic acid modification of this (KOH—AB pH 1—mPAS). Using normal colonic mucosa obtained from 11 non-cancer patients, the mPAS and PAPS techniques were tested for specificity and reproducibility for staining sialic acid, either alone or in combination with Alcian Blue. A spectrophotometric method was devised to quantify the uptake of both Schiff and Alcian Blue stain by sections. At low temperature and pH5.5, the mPAS procedure had improved specificity over the PAPS procedure, and after saponification it could be used to stainO-acetyl-substituted sialic acid. When used in combination with Alcian Blue at pH 1, however, underestimation of the sialic acid content occurred owing to interference between Alcian Blue and Schiff dyes. Interference was even greater with KOH—AB pH1—PAPS procedure for both sialic acid and sulphate components. We conclude that caution must be exercised in interpretation of the staining results obtained with these new combination methods and that more accurate information on the sialic acid and sulphate content of colonic mucin is obtained by staining serial sections with the mPAS technique and Alcian Blue pH 1 alone.

    Notizen: Summary The increasing knowledge of the structure and function of macromolecular carbohydrate-protein compounds (glycoproteins) has led to an increase in interest in these molecules in biology and especially in medicine in recent years. In saliva the largest part of the glycoproteins is constituted by the mucins. Salivary mucins exhibit very high molecular weight, have a high percentage of carbohydrate (approx. 70%) in the form of fucose, N-acetylglucosamine, N-acetylgalactosamine, galactose mannose and N-acetylneuraminic acid. They have a negative charge due to N-acetylneuraminic acid and sulphate residues, which together with the high molecular weight of these glycoproteins leads to characteristic viscous properties. The most important mucin properties which play a role in the upper respiratory and alimentory tract will be presented, especially the viscous character with respect to lubricative functions, the protection of mucous surfaces against microorganisms and other agents. A few physiological and pathological mucin analysis of human saliva and its method of analysis will be presented.
