    Notes: Summary and Conclusions The changes in hematological parameters and physicochemical composition of red cells which accompany their aging in vivo and the results of recent experiments in which the peripheral zone of density fractionated erythrocytes were labeled with positively charged colloids or treated with relatively high molecular weight polycations suggest a reordering or restructuring of membrane components of red cells, rather than a change in their mean surface charge density. Age-related alterations in the two-dimensional arrangement of integral membrane glycoproteins may arise as a result of membrane loss. These types of perturbations may also be responsible for other altered properties of aged cells as seen, for example, in their behavior in two-phase aqueous polymer systems [30], their agglutinability in the presence of certain antisera [4], their permeability to ions [17], and mechanical properties of the membrane [32].

    Description / Table of Contents: Summary An in vitro system was developed to test the propensity of rat erythrocytes (RBC) toward phagocytosis by homologous peritoneal macrophages after in vivo aging or after in vitro surface modification. “Old” RBC obtained from erythrocyte populations separated into fractions on the basis of their density were found to be phagocytosed to a significantly greater extent than cells obtained from “young” cell fractions. Neuraminidase treatment of RBC resulted in extensive phagocytosis in comparison to control cells representing the whole population or “old” cells. Galactose oxidase treatment of neuraminidase treated RBC afforded no protection from uptake by macrophages. In contrast, treatment of RBC with neuraminidase immobilized on Sepharose 4B, leading to removal of up to ∼25% of the total membrane sialic acid, did not lead to phagocytosis above control values. These results indicate that extreme caution is necessary in the interpretation of experiments in which neuraminidase is utilized to produce simulated “aged” RBC.
    Notes: Zusammenfassung Es wurde ein in vitro-System entwickelt, in dem die Tendenz von Rattenerythrozyten (RE) gemessen wurde, entweder nach einer in vivo-Alterung oder einer in vitro-Oberflächenveränderung durch homologe peritoneale Makrophagen phagozytiert zu werden. Wurden „alte“, dichtere RE in Fraktionen auf Basis ihrer Dichte aufgetrennt, so war die Phagozytose von „alten“ RE wesentlich ausgeprägter als jene von „jüngeren“ Zellen. Die Neuraminidase-Behandlung von RE führte zu einer hochgradigen Phagozytose im Vergleich zu den Normalkontrollen der gesamten Zellpopulation oder „alten“ Zellen. Eine Galaktose-Oxydase-Behandlung von neuraminidasebehandelten RE brachte keinen Schutz gegenüber den Makrophagen. Im Gegensatz dazu führte die Behandlung von RE mit Neuraminidase, die an Sepharose-4B festgebunden ist, zwar zu einer Entfernung bis zu ∼25% der gesamten membrangebundenen Neuraminsäure, ergab jedochkeine über den Kontrollwerten liegende Phagozytose. Daraus folgt, da\ man bei der Interpretation von Experimenten mit Neuraminidase sehr vorsichtig sein mu\, wenn das Enzym dafür verwendet werden soll, künstlich „gealterte“ RE zu erzeugen.

    Notes: Desialylation of human red blood cells (RBC) by Vibrio cholerae neuraminidase (VCN) was found to produce cells with electrophoretic properties which were inconsistent with the view of simple loss of N-acetyl-neuraminic acid (NANA) as the sole effect of VCN treatment. Modification of human RBC with 50-350 U VCN/1010 RBC for one hour at 37°C releases 90-100% of the NANA and produces a progressive decrease towards zero in their electrophoretic mobilities when measured in 0.15 M NaCl (pH 7.2) at 25°C. The appearance of positive groups on the desialylated cells was indicated by the VCN-treated cells displaying positive mobilities below ∼ pH 5.5 and increased negative mobilities at ∼ pH 9 as well as substantial increases in their mobility at neutral pH following treatment with formaldehyde. Adsorption of about 95% of the VCN activity at 0°C to the RBC did not produce any significant change in their electrophoretic mobilities thus indicating that the observed changes in the electrophoretic properties of the RBC following VCN treatment could not be attributable to adsorption of VCN.These studies indicate that the cationic charge groups which appear at the electrophoretic surface of the RBC after VCN treatment are probably of endogenous origin. It is suggested that this alteration rather than simple NANA release may operate to shorten the in vivo survival time of desialylated red cells.
