    Notes: A pilot-scale process for the isolation of an aliphatic, amidase from Pseudomonas aeruginosa has been developed. A constitutive, partially irrepressible mutant was employed to give a high initial enzyme concentration. An existing laboratory isolation procedure has been scaled up and modified particularly by substitution of polyethylene glycol for ammonium sulfate precipitation as the first stage in the conversion of the fractionation to continuous operation. Full recovery of activity was achieved with the modification. The recovery of enzyme from a subsequent chromatographic stage was 85% and the maximum overall purification was 28-fold.

    Notes: A pilot-plant process has been developed for the continuous extraction and partial purification of prolyl-tRNA synthetase from mung bean. The bean slurry was wet ground in a hammer mill, clarified by two-stage centrifugation, and the protein in the effluent fractionated by precipitation at pH values of 5.2 and 4.2. The throughput was 13 kg dry bean/hr. The improved extraction process and reduced processing time resulted in an enzyme product with a specific activity 16 × that previously obtained in the batch process. The yield was also 50-60 times higher.

    Notes: Release of protein from a suspension of bakers' yeast (Saccharomyces cerevisiae) by disruption in an industrial agitator mill has been studied. Protein release on disruption in the mill is a first-order rate process. The rate constant is dependent on at least six parameters. Increased disruption efficiency was obtained at higher agitator speeds, greater loading of bead attritive elements and lower rates of upward recycle of yeast suspension through the mill. An increase in temperature from 5 to 42°C was accompanied by a reduction in disruption efficiency of approximately 20%. With optimal values of the parameters examined the throughput of the mill is 5.32 kg/hr of soluble protein for 90% disruption.
