    Notes: A pilot-scale process for the isolation of an aliphatic, amidase from Pseudomonas aeruginosa has been developed. A constitutive, partially irrepressible mutant was employed to give a high initial enzyme concentration. An existing laboratory isolation procedure has been scaled up and modified particularly by substitution of polyethylene glycol for ammonium sulfate precipitation as the first stage in the conversion of the fractionation to continuous operation. Full recovery of activity was achieved with the modification. The recovery of enzyme from a subsequent chromatographic stage was 85% and the maximum overall purification was 28-fold.

    Notes: A laboratory process for the isolation of type B acid phosphatase from human erythrocytes has been scaled up 80 times. The enzyme was purified 1300-fold by adsorption on calcium phosphate gel, ammonium sulfate precipitation, and gel filtration. The yield was 27%. Results for the type A enzyme are also presented.

    Notes: Some of the problems involved in the scale-up of enzyme isolation procedures have been examined. In particular the isolation of prolyl-tRNA synthetase has been investigated. Using a modified procedure (500 × laboratory scale) the enzyme has been separated from 25 kg. of Phaseolus aureus (mung bean) and Partially purified by acid precipitation followed by fractional precipitation with ammonium sulfate. The yield of enzyme was 42 g. (10.5% recovery).
