Arndt-Eistert Reactions in Carbohydrate Chemistry. - Syntheses of Biologically Important Carbohydrtes, 2. Total Synthesis of RhodinoseThe rearrangement of the α-diazoketones from peracetylated aldonoyl chlorides takes place with elimination of the β-acetoxy groups. This reaction is generally applicable to syntheses of the 2,3,6-trideoxyhexoses from streptomyces antibiotics. - 2,3,6-Trideoxy-L-threo-hexose (rhodinose) (17) has been synthesized in 13 steps from L-xylose (4). The key step is the rearrangement of 3,4,5,6-tetra-O-acetyl-1-deoxy-1-diazo-L-xylo-2-hexulose (7) with formation of methyl 4,5,6-tri-O-acetyl-2,3-dideoxy-L-threo-hex-2-enoate (9). Hydrogenation of 9 yields the 2,3-dideoxyhexonic acid ester 11. Sponification of 11 and ring closure to the lactone 13 gives the rhodinonolactone 16 after introduction of the 6-deoxy group. The antibiotic sugar 17 is formed by reduction of the lactone 16 with diisobutylaluminium hydride as a mixture of anomeric furanoses and pyranoses.

Die Umlagerung der α-Diazoketone aus peracetylierten Aldonoylchloriden, die unter Eliminierung der β-Acetoxygruppen verläuft, ist ein allgemeines Syntheseprinzip für 2,3,6-Tridesoxyhexosen der Streptomces-Antibiotika. - Schlüsselreaktion einer dreizehnstufigen Synthese der 2,3,6-Tridesoxy-L-threo-hexose (Rhodinose) (17) aus L-Xylose (4) ist die Umlagerung der 3,4,5,6-Tetra-O-acetyl-1-desoxy-1-diazo-L-xylo-hexulose(7)zum 4,5,6-Tri-O-acetyl-2,3-didesoxy-L-threo-hex-2-enonsäure-methylester (9). Hydrierung von 9 zum 2,3-Didesoxyhexonsäureester 11, Verseifung von 11 und Ringschluß führen zum Lacton 13, aus dem durch Einführung der 6-Desoxy-Gruppe das Rhodinon-säurelacton 16 entsteht. 16 liefert nach Reduktion mit Diisobutylaluminiumhydrid den Antibiotikum-Zucker 17 als Gemisch anomerer Furanosen und Pyranosen.
