    Notizen: The complete synthetic medium for the growth of Tetrahymena pyriformis E was subjected to gamma-radiation from cobalt60. A dose-response curve indicated the range of radiation which damaged the medium so that growth of Tetrahymena was inhibited.The essential vitamins and amino acids which comprise the medium were individually irradiated in solution. Media were prepared with single irradiated components and tested for their ability to support growth of the protozoa.Thiamine, riboflavin, pantothenate, pyridoxine, folic acid, and thioctic acid were destroyed by less than 1 × 106 rep, while 2 × 106 rep was required to inactivate nicotinic acid. Most amino acids proved to be relatively radiation-resistant. At the high radiation levels of 1 × 107 rep and 2.3 × 107 rep respectively, only serine and methionine were damaged. All other amino acids remained biologically active even after receiving 2.3 × 107 rep, the highest level of radiation employed. Organoleptic observations, however, showed radiation-caused color and odor changes in many of those amino acids whose biological activity for Tetrahymena was unaffected. The nucleotide, guanylic acid, was also inactivated at 2.3 × 107 rep.

    Notizen: SUMMARY. Vegetative clones of Tetrahymena pyriformis, variety 2, mating type II, were the recipients of various dosages of X-rays from 10,000 to 400,000r. One clone from the 400,000 level (UM7X), when mated with non-irradiated cells of various mating types in variety 2, failed to produce a migratory nucleus. A one-way transfer from the normal to irradiated mate occurred, resulting in the production of haploid exconjugants.When haploids were crossed with diploids the former failed to produce a migratory nucleus. A one-way nuclear transfer from the diploid to the haploid occurred resulting again in the production of haploid exconjugants. When two haploids were crossed a clone resulted which showed 80 to 90 percent amicronucleate forms.The mating type of the derived haploid is always that of the diploid parent. All derived haploids manifest physiological and morphological characteristics similar to those of the diploid except that the haploids possess a smaller micronucleus.

    Notizen: Abstract. Neopterin is a biochemical marker for the activation of the cell-mediated immune system. We measured neopterin, β2-microglobulin, and acute phase proteins in 31 HIV-seropositive and -seronegative Zambian patients with tuberculosis, using stored sera that had been obtained at the beginning and at end of antituberculosis treatment. In both HIV-seropositive and -seronegative patients neopterin and acute phase proteins were elevated when tuberculosis was initially diagnosed and fell during treatment. In contrast, the mean β2-microglobulin level increased during antituberculous therapy in the HIV-seropositive group. Serum neopterin levels at diagnosis were correlated with other parameters of disease activity (fever, anemia, and weight loss). In both groups, patients with persistently elevated neopterin levels at the end of treatment were more likely to suffer relapse of tuberculosis or other adverse health events in the subsequent follow-up period. Neopterin can be used to monitor the response to antituberculous therapy in both HIV-seropositive and -seronegative patients and may have a prognostic value for the patients' wellbeing in the follow-up period.

    Notizen: Abstract Nafion® is a commercially available perfluorosulphonate cation exchange membrane commonly used as a perm-selective separator in chlor-alkali electrolysers and as the electrolyte in solid polymer fuel cells. This usage arises because of its high mechanical, thermal and chemical stability coupled with its high conductivity and ionic selectivity, which depend strongly on the water content. The membrane was therefore studied in different states of hydration with two complementary techniques: atomic force microscopy (AFM) and small angle X-ray scattering (SAXS) combined with a maximum entropy (MaxEnt) reconstruction. Tapping mode phase imaging was successfully used to identify the hydrophobic and hydrophilic regions of Nafion. The images support the MaxEnt interpretation of a cluster model of ionic aggregation, with spacings between individual clusters ranging from 3 to 5 nm, aggregating to form cluster agglomerates with sizes from 5 to 30 nm. Both techniques indicate that the number density of ionic clusters changes as a function of water content, and this explains why the bulk volumetric swelling in water is observed to be significantly less than the swelling inferred from scattering measurements.
