    Notes: Summary Cell-free translation of salivary gland RNA or of purified Balbiani ring RNA (75S) in a reticulocyte lysate system gives rise to high molecular weight translational products (HMTP). In addition to a common size (approx. 1×106 daltons) HMTP share imunogenic determinants with the giant secretory proteins of salivary glands. This suggests that HMTP correspond to in vivo secreted proteins and thus, corroborates the notion that 75S-RNA is the messenger for these proteins. The time course of HMTP synthesis and the lack of appearance of lower molecular weight components as translational products of 75S-RNA indicate that the synthesis of HMTP (and of secretory proteins) occurs in one piece by an uninterrupted process. HMTP are regarded the largest polypeptides so far synthesized in a cell-free system.

    Notes: Zusammenfassung Durch Abwandlung der elektrophoretischen Auftrennungsmethode wird das Vorkommen einer neuen Isoenzymbande der Kreatinkinase, das ursprünglich auf die Vogelordnung Passeriformes beschränkt zu sein schien, allgemein in verschiedenen Geweben von Vögeln nachgewiesen. Eine weitere neue Isoenzymbande wird in bestimmten Entwicklungsstadien der Skelettmuskulatur beobachtet. Es wird diskutiert, dass diese neuen Isoenzyme alternative thermodynamisch mögliche Konformere der Gehirn-Kreatinkinase (BB-Enzym) oder ihrer Monomere (B) sein könnten. Anhand der Untersuchungsergebnisse wird aufgezeigt, dass diesen vermutlich durch Konformationsunterschiede bedingten Isoenzymen physiologische Bedeutung zukommen könnte.

    Notes: Summary Unequivocal identification of M-CK in cell extracts from fused cells of myogenic cell lines is difficult due to almost identical behaviour of the M-CK and a contaminating enzyme activity in electrophoresis. If CK dimers present in cell extracts were subjected to dissociation and reassociation in the presence of exogenous B-CK subunits, the formation of easily identifiable MB-CK was demonstrated, indicating the presence of M-CK in the myogenic rat cell lines.

    Notes: Summary By combining enzyme histochemistry for fiber typing with immunohistochemistry for slow and fast myosin a correlation between fiber type and myosin type was sought in human skeletal muscle. Fiber typing was done by staining for myofibrillar ATPases after preincubation at discriminating pH values. Myosin types were discriminated using type specific anti-rabbit myosin antibodies shown to cross-react with human myosin and were visualized by a protein A-peroxidase method. Type I fibers were shown to contain slow myosin only, type IIA and IIB fibers fast myosin only, and type IIC fibers both myosins in various proportions. When muscle biopsies from well-trained athletes were investigated essentially the same staining pattern was observed. However, rarely occurring type I fibers with high glycolytic activity were detected containing additional small amounts of fast myosin and occasional type IIA fibers had small amounts of slow myosin. Based on the observation of various fiber types in which slow and fast myosin coexist we propose a dynamic continuum of fibers encompassing all fiber types.

    Notes: Summary The distribution of three myofibrillar M-band proteins, myomesin, M-protein and the muscle isoform of creatine kinase, was investigated with immunocytochemical techniques in skeletal muscles of embryonic, fetal, newborn and four-week-old rats. Furthermore, muscles of newborn rats were denervated and examined at four weeks of age. In embryos, myomesin was present in all myotome muscle fibres of the somites, whereas M-protein was detected only in a small proportion of the myotome muscle fibres and muscle creatine kinase was not detected at all. In fetal and newborn muscles, all fibres contained all three M-band proteins. At four weeks of age, when fibre types (type 1 or slow twitch fibres and type 2 or fast twitch fibres) were clearly discernable, the pattern was changed. Myomesin and muscle creatine kinase were still observed in all fibres, whereas M-protein was present only in type 2 fibres. On the other hand, in muscle fibres denervated at birth all three M-band proteins were still detected. Our results suggest 1) that during the initial stages of myofibrillogenesis expression and incorporation of myomesin into the M-band precede that of M-protein and muscle creatine kinase; 2) that expression and incorporation of all three M-band proteins during fetal development is nerve independent and non coordinated to the expression of different forms of myosin heavy chains, and 3) that the suppression of M-protein synthesis during postnatal development is nerve dependent and reflects the maturation of slow twitch motor units.

    Notes: Summary Chicken muscle cells secrete characteristic proteins when grown in the serum-free and hormonally defined culture medium ‘DMN’. The most prominent band detected by gel electrophoresis represents a protein of mol.wt 22,000. Fibroblasts released a mol.wt 16,000 protein and fibronectin (mol.wt 220,000) into the medium. The mol.wt 22,000 protein band resolved in 2 dimensional gels into 2 spots which migrated to the same positions as small heat shock proteins as well as butyrate-inducible proteins (BIP) which can be demonstrated in whole cell extracts after butyrate treatment in the presence of serum. The synthesis and release of the mol.wt 22,000 protein is repressed by supplementing the culture medium with serum but not with chick embryo extract.
