    Notes: Abstract Aims/hypothesis. The pre-clinical phase of diabetic nephropathy is characterised by increased glomerular filtration rate and episodes of microalbuminuria. The cause of the microalbuminuria has been variably ascribed to alterations of the size or charge selective barriers of the glomerulus or both or as a consequence of the haemodynamic changes. Our aim was to investigate very early albumin permeability alterations in isolated glomeruli which were not subject to perfusion pressure.¶Methods. Isolated glomeruli were studied from 120 male Wistar rats, divided into three groups: streptozotocin-treated, streptozotocin-treated with insulin pellet implants, and controls. From each group ten animals were killed at 7, 14, 28, and 56 days after induction. Study variables included blood pressure, proteinuria, iopamidol clearance, albumin permeability and glomerular area. Subsequently, albumin permeability, proteinuria, and iopamidol clearance were determined in an additional group of 40 diabetic animals studied at 24, 72, 96, and 120 h after induction.¶Results. Albumin permeability increased steadily from induction in streptozotocin-treated animals, reaching a plateau at approximately 120 h. Glomerular filtration rate was shown to increase significantly at approximately 7 days and proteinuria correlated with it. Glomerular hypertrophy was observed both in streptozotocin-treated animals and in streptozotocin-treated rats with insulin pellet implants. Strict blood glucose control delayed the appearance of the permeability defect in isolated glomeruli and inhibited the increase in glomerular filtration in intact animals. It did not prevent glomerular hypertrophy.¶Conclusion/interpretation. An albumin permeability defect exists early in isolated non-perfused glomeruli from streptozotocin-treated rats and seems to be independent of glomerular filtration rate alterations.     Notes: Summary The effect of chronic treatment with indapamide on blood pressure (BP), baroreceptor sensitivity (BRS) and vascular reactivity (VR) were investigated in 10 patients with essential hypertension. After 3 months of therapy with indapamide 2.5 mg/d the mean arterial pressure (MAP) had decreased from 135±6 to 112±2 mmHg (p〈0.001); the heart rate (HR) had not changed, VR had decreased from 6.1±1.2 to 4.8±1.8 (pg·min·kg)−1 (p〈0.05), and BRS had increased from 8.3±3.7 to 12.2±5.3 ms/mmHg (p〈0.005), with a leftshift of the relationship between BP and heart period. An inverse correlation was found between the pre-treatment systolic blood pressure and the change in baroreceptor sensitivity after indapamide (r=0.59; p〈0.05). In conclusion, chronic treatment with indapamide enhances BRS and resets the reflex. The resetting may account for the lack of tachycardia at rest observed after treatment with indapamide. The mechanism by which indapamide interferes with the baroreceptor reflex requires further investigations.

Cardiac angiotensin-converting enzyme (ACE) was estimated by radioligand binding in myocardial homogenates and by autoradiography studies of radioligand binding to ACE in heart sections.3. Radioligand binding studies demonstrated an increase in binding sites in animals with myocardial infarction, compared with sham operated animals. Mean increases were 457% in right atrium, 295% in left atrium, 326% in right ventricle, 187% in left ventricle and 530% in the region of the left ventricular infarct, compared with the sham left ventricle.4. Autoradiography studies confirmed tissue homogenate binding studies, demonstrating (i) a marked increase in ligand binding in the infarct area and (ii) an increase in binding density in the hypertrophied myocardium of right and left atrium, and right and left ventricle.5. The induction of cardiac ACE in the myocardium of rats with chronic heart failure may participate in the pathophysiology of cardiac hypertrophy.

    Notes: 1. Cardiac angiotensin-converting enzyme (ACE) is localized in high concentration in cardiac valves, coronary vessels, right and left atrium and right and left ventricle.2. Cardiac ACE is functionally active in converting angiotension I to angiotensin II.3. The level of cardiac ACE measured by radioinhibitor binding or by quantitative in vitro autoradiography was greatly increased after experimental myocardial infarction in the rat. The increase was greatest in the fibrous scar tissue of the free left ventricular wall infarct, but there were also significant increases in the ACE concentration in the four chambers of the heart.4. Treatment with enalapril for 4 weeks following coronary ligation inhibited cardiac ACE, including the high levels found in the scar in the left ventricular free wall.5. There was a close relationship between the systolic blood pressure and left ventricular mass in several models of experimental hypertension, despite varying degrees of activation of the renin–angiotensin system. However no relationship between the degree of left ventricular hypertrophy and changes in cardiac ACE could be determined.6. Inhibition of cardiac ACE may contribute to the beneficial effect of ACE inhibitors in cardiac hypertrophy and remodelling, and may play a part in the cardioprotective role of ACE inhibitor.

    Notes: Abstract Domain growth of bulk partially melted Nd1+xBa2−xCu3O7−δ in air was found to be severely limited by liquid-phase segregation and losses. The use of a reduced oxygen pressure greatly improved the stoichiometry control, leading to the formation of well-developed domains and enhanced superconducting properties. Direct nucleation of Nd1+xBa2−xCu3O7−δ from the liquid phase by dissolution of Nd2BaCuO5 and a spiral growth mechanism in three perpendicular directions of the platelets stacked in the domains, are suggested.

    Notes: Abstract Large domains with platelets almost parallel to each other were obtained in bulk YBa2Cu3O7−δ by a single-step partial melting procedure. The mechanisms of nucleation and growth of platelets are discussed. The nucleation of peritectic material from the liquid phase is favoured by heterogeneities in the melt. Experimental evidence of spiral growth of the nuclei in the [0 0 1] direction is given. Furthermore, structures of growth, which could also be an indication of spiral growth in the [0 1 0]/[1 0 0] directions, are shown. The final morphology of the domains can be explained on the basis of the periodic bond chain (PBC) theory if the growth rates of flat (F) faces of the platelets are dominated by kinetic coefficients which differ between them. The morphology of the as-grown (0 0 1) surface is explained in the framework of the PBC theory with the shape of the steps of macrospirals governed by the transition from roughness to smoothness of the liquid-solid interface. An account of large step heights is given by the model of giant screw dislocations caused by an impurity-induced lattice-constant gradient. Even higher step heights are correlated to the presence of obstacles and lack of liquid phase.
