    Notes: Pituitary binding sites for gonadotrophin-releasing hormone (GnRH) were examined in Booroola ewes on Days 4 and 12 of the oestrous cycle and more than 12 months after ovariectomy. No significant differences were observed in the affinity of the binding sites for [125 I]D Ala6, des Gly10 GnRH ethylamide, in non-carriers (++) or homozygotes of the fecundity gene (FF), in intact or ovariectomized animals (overall Ka= (1.8 ± 0.1) × 109 M−1; mean ± SEM, n = 67). In ++ ewes, the number of GnRH binding sites on Day 4 of the oestrous cycle (0.199 ± 0.025 fmol/mg pituitary, n = 8) was significantly greater (P〈0.05) than on Day 12 (0.107 ± 0.012 fmol/mg pituitary, n = 12). In contrast in FF ewes the number of binding sites on Days 4 and 12 were similar (Day 4: 0.119 ± 0.030, n = 8; Day 12: 0.092 ± 0.014, n = 10). On Day 4, but not Day 12, the binding site numbers were significantly higher in ++ compared with FF pituitaries (P〈0.05). Likewise on Day 4, but not Day 12, the plasma concentrations of follicle-stimulating hormone were significantly higher (P〈0.005) in FF (1.60 ± 0.15 ng/ml) than ++ ewes (0.96 ± 0.10 ng/ml). After ovariectomy, no gene-specific differences in binding site numbers were noted despite significantly higher plasma follicle-stimulating hormone concentrations in FF animals. No gene-specific differences in plasma luteinizing hormone concentrations were observed in these animals on Days 4 or 12, or after ovariectomy. Moreover, luteal phase plasma progesterone concentrations were also similar between ++ and FF ewes. We conclude that the gene-specific differences in pituitary responsiveness to GnRH and in pituitary gonadotrophin output are unlikely to be due to differences in GnRH receptor binding affinities.

    Notes: Summary Paracetamol absorption and 99Tc-DTPA measurements have been used to determine the influence of levodopa on gastric emptying in 8 healthy elderly volunteers. In the absence of levodopa 7 subjects showed a rapid gastric emptying pattern by gamma-camera and a single major peak in the plasma concentration-time curve of paracetamol. One subject showed two rapid phases of gastric emptying separated by a period of negligible emptying and had 2 separate peaks in the paracetamol plasma concentration-time curve. In the presence of levodopa, the gamma-camera data for 6 subjects showed a pattern of gastric emptying consisting of 2 rapid phases separated by a plateau. In each case secondary peaks in the plasma concentration-time curve of paracetamol occurred about 30 min after the end of the plateau. The time to 90% emptying on the gamma scan was increased significantly from 40 min to 65 min in the presence of levodopa. Comparison of the present data with those reported previously indicates that levodopa affects gastric emptying in the both elderly and young volunteers to a similar extent.

    Notes: Abstract. Objective: Abnormal right ventricular function in patients with pulmonary heart disease (PHD) may contribute to their reduced exercise capacity. We have evaluated the effect of subacute dosing with xamoterol, a beta-1 adrenoreceptor partial agonist with inotropic properties, on right ventricular function in patients with PHD. Patients: Twelve patients with advanced chronic obstructive pulmonary disease and clinical evidence of PHD received xamoterol 200 mg twice daily or placebo for 7 days in a randomised double-blind crossover study. Right heart krypton-81m radionuclide ventriculography was used to derive right ventricular ejection fraction (RVEF) both at rest and during submaximal exercise, and indices of right ventricular systolic and diastolic function at rest. Results: During treatment with placebo, mean RVEF was 0.53 at rest and was unchanged during exercise. After xamoterol, mean RVEF was 0.55 at rest and increased to 0.59 during exercise. Xamoterol increased right ventricular peak ejection rate from 3.04 to 3.45 EDV⋅s–1 and mean early diastolic filling rate from 1.00 to 1.20 EDV⋅s–1. Conclusion: Subacute treatment with xamoterol in patients with PHD improves right ventricular systolic and diastolic function at rest and results in a favourable augmentation in right ventricular function during submaximal exercise.

    Notes: Abstract The absorption of 75Se-23-selena-25-homotaurocholate (SeHCAT) was compared with vitamin-B12 absorption and conventional radiography in 44 patients with inflammatory bowel disease. The retention of SeHCAT was normal in 11 patients with ulcerative colitis but was abnormally low in 9 patients with terminal-ileal resection, 9 out of 14 patients with small-bowel Crohn's disease and in 2 out of 10 patients with Crohn's colitis. The 5 patients with small-bowel Crohn's disease and normal retention had either inactive disease or no radiological evidence of terminalileal involvement. Measurements of the absorption of vitamin B12 did not discriminate between these groups, and there was very poor correlation between B12 and SeHCAT absorption (r=0.506, P〈0.05). There was extremely good correlation of SeHCAT retention measured using a whole-body counter with that measured using an uncollimated gamma camera (r=0.96, P〈0.001). The results suggest that SeHCAT retention may prove complementary to conventional methods of assessing small-bowel disease in patients with inflammatory bowel disease. As measurement by gamma camera is feasible, this test can be used in most departments of nuclear medicine.

    Notes: Abstract A theoretical description of the relationship between the count rate detected in the cardiac region of interest and the arterial organ input function in dynamic scintigraphy is presented. It is shown that, provided the time-activity curve for the heart is corrected for extra-cardiac and unlabelled activity, it is proportional to the arterial organ input over both the first-pass and equilibrium phases of the passage of an intravenously injected radiopharmaceutical. A practical example demonstrating the value and validity of correcting the cardiac curve in dynamic radio colloid scintigraphy is described. Estimates of the colloid clearance rate to reticuloendothelial sites using the cardiac curve without correction were significantly lower (mean 0.085/min) than those derived from the liver uptake curve (mean 0.213/ min). However, corrected cardiac curve clearance rates (mean 0.225/min) were not significantly different from the liver ones. Also, the corrected cardiac curve clearance values correlated linearly with liver curve values (correlation coefficient 0.89, standard error of the estimate 0.021/ min), whereas the uncorrected values showed no significant correlation. Thus, correction of the cardiac curve gave clearance rate values that were both more accurate and more precise than those obtained without correction.

    Notes: Abstract Simultaneous measurements of the clearance rates of technetium 99m diethylenetriaminepentaacetic acid (99mTc-DTPA) and chronium 51 ethylenediaminetetraacetic acid (51Cr-EDTA) were performed in 30 patients with a range of renal function (glomerular filtration rates between 9 and 120 ml/min). Using multiple blood samples, the two clearance values correlated well (r=0.991, standard error 3.9 ml/min), but DTPA clearance was systematically higher by 7.6%. For each radiopharmaceutical, an equation was derived to correct clearance values obtained using only plasma samples taken at 2 and 4 h for the systematic error inherent in this technique compared with analysis of the complete plasma concentration-time curve. The root mean square error remaining after application of these equations was 1.9 ml/min for both the EDTA and DTPA data. The corresponding errors obtained using the equation derived by Brochner-Mortensen for EDTA plasma clearance were 2.2 ml/min and 1.9 ml/min, respectively, these values were not significantly different from those obtained using the equations derived in this study.

The theory of the technique is presented and the validity of the assumptions involved is discussed. The sources of error are considered and the corresponding error magnitudes assessed.
