Summary 1. A method for the determination of bile acids is described combining the separation by thin layer chromatography with the spectrofluorometric measurement after treating the bile acids for one hour at 60° C in sulfuric acid (65 Vol.-%). 2. The fluorescence of the derivatives of trihydroxycholic acid was most intensive at 420 mµ, when excited at 385 mµ, for derivatives of dihydroxycholic acid at 450 mµ, when excited at 410 mµ. 3. The limit of determination of cholic acid and its taurine and glycine conjugates as well as of the conjugates of desoxycholic acid is reached at a concentration of 0,02 µg/ml of the final volume. The limit of determination of desoxycholic and chenodesoxycholic acid is 0,06 µg/ml, because of the weaker specific fluorescence. The concentration of bile acids varies linearly with the inensity of fluorescence. The method is also suitable for the determination of taurochenodesoxycholic acid and lithocholic acid. 4. This method, already successfully employed in the analysis of bile acids in rat and human bile, was used for the determination of several bile acids in human serum. Free desoxycholic acid was never found in serum even under pathologic conditions. Free cholic acid was detected in the serum of three patients out of seventeen with catarrhal and obstructive jaundice. 5. The concentration of glycocholic acid (1,5 µMol/100 ml serum) exceeds that of taurocholic acid (0,6 µMol/100 ml serum) in human serum as well as in human bile. In obstructive jaundice the concentration of glycocholic and taurocholic acid was 11,4 µMol/100 ml and 5,7 µMol/100 ml, respectively (three cases). In catarrhal jaundice an elevated concentration was found mainly of glycocholic acid (15,7 µMol/100 ml) but also of taurocholic acid (3,2 µMol/100 ml) in four patients out of fourteen.

Zusammenfassung 1. Es wird eine Methode beschrieben, mit der nach Auftrennung eines Gallensäurengemisches auf der Dünnschichtplatte die einzelnen Gallensäuren anhand der Fluorescenz, die sie nach Inkubation in 65%iger H2SO4 (Vol-%) entwickeln, bestimmt werden können. 2. Für die Derivate der Trihydroxycholansäure wurde die höchste Fluorescenzintensität mit einer Anregungswellenlänge von 385 mµ und einer Wellenlänge der Sekundärstrahlung von 420 mµ erzielt. Das Maximum der Wellenlänge der Sekundärstrahlung wurde für dic Derivate der Dihydroxycholansäuren bei 450 mµ mit einer optimalen Anregungswellenlänge von 410 mµ bestimmt. 3. Die Bestimmbarkeitsgrenze für Cholsäure, deren Taurin- und Glycinkonjugate sowie der Taurin- und Glycinkonjugate der Desoxycholsäure liegt bei 0,02 µg/ml Cuvettenfüllung. Aufgrund der schwächeren Fluorescenz der freien Desoxycholsäure und der Chenodesoxycholsäure liegt deren Bestimmbarkeitsgrenze bei 0,06 µg/ml Cuvettenfüllung. Bis zu einer Konzentration von 2 µg/ml bestand eine direkte Proportionalität zwischen Gallensäurenkonzentration und Fluorescenzintensität. Neben der Bestimmung der bereits genannten Gallensäuren wurde die Methode auch zur Messung der Taurochenodesoxycholsäure und der Lithocholsäure benutzt. 4. Die Methode, die sich bereits bei der Bestimmung der Gallensäurenzusammensetzung in Ratten- und Menschengalle bewährt hat, wurde auch zur Bestimmung der Gallensäurenzusammensetzung in Humanserum herangezogen. Freie Desoxycholsäure konnte im Serum auch unter pathologischen Bedingungen nicht nachgewiesen werden. In 3 von 17 Seren, die von Patienten mit Hepatitis epidemica bzw. Verschlußikterus stammten, konnte freie Cholsäure nachgewiesen werden. 5. Im Serum des Menschen überwog, wie auch in der Galle, die Glykocholsäure-Konzentration (1,5µMol/100 ml) die der Taurocholsäure (0,6 µMol/100 ml). Im Serum von Patienten mit Verschlußikterus war die Konzentration der beiden Gallensäuren angestiegen. Ähnliche Verhältnisse wurden im ersten Stadium der Erkrankung bei Patienten mit Hepatitis epidemica gefunden.
