    Notes: An indirect immunoperoxidase method was used to study the expression of tissue polypeptide antigen (TPA) and cancer antigen 125 (CA 125) in 47 benign and malignant ovarian tumours. Tissue polypeptide antigen and CA 125 antigen were expressed respectively in 22 (73%) and 16 (53%) of the 30 adenocarcinomas and in five (29%) and four (23%) of the 17 benign tumours. Co-expression of TPA and CA 125 antigen occurred in 12 (40%) malignant and four (23%) benign tumours. Ultrastructurally, TPA and CA 125 antigens were located at the cell surface and microvillous surfaces.Evaluation of combined TPA and CA 125 antigen results revealed a remarkable improvement in the positivity rate and a significant decrease (P〈0.05) in the negativity rate of ovarian carcinomas as compared with the result of each one separately. These findings provide complementary evidence for the previous results on the plasma levels of TPA and CA 125 antigen and suggest that specific combinations of tumour markers may be more effective for the diagnosis and monitoring of ovarian carcinomas, than the use of any single marker.

    Notes: Variations in the number of silver-stained nucleolar organizer region-associated proteins (AgNORs) were studied in paraffin sections of 42 benign prostatic lesions, comprising four cases of granulomatous prostatitis, five of squamous or transitional metaplasia, eight of atypical and 25 of regular hyperplasia, and 37 of prostatic adenocarcinoma, with their metastases. There was a significant difference between the mean AgNOR counts of the benign and malignant prostatic lesions (1.58±0.26 v. 4.34±1.53; P〈0.01). The mean AgNOR counts significantly increased with increasing Gleason's grade (P〈0.01) and clinical stage (P〈0.05) of the tumours. AgNOR counting may contribute to the conventional diagnostic and prognostic indices of cancer of the prostate.

    Notes: Aim: When surgery with postoperative superficial electron irradiation is applied, the recurrence rate of keloid lesion has been found to be 〈30%. In this study, we assessed the molecular changes underlying the effect of electron irradiation by differential global gene expression analyses of both cultured keloid and normal skin fibroblasts. Materials and Methods: Primary cultured fibroblasts from 4 active keloids and their adjacent normal dermal tissues were irradiated at a calibrated dosage of 15 Gy with 6 MeV electron beam generated by a linear accelerator. Corresponding paired non-irradiated cells were used as control. RNA isolated from the collected cells was labeled with 33P, hybridized to the cDNA microarray gene filters and analyzed. Results: After irradiation, the gene expression profiles of keloid and normal skin fibroblasts were closely similar. Electron irradiated keloid fibroblasts showed suppressed levels of collagen typeI (alpha2), collagen typeVI (alpha1), matrix metalloproteinase 2, fibronectin 1, insulin-like growth factor binding protein 3, alpha-1-antichymotorypsin and heparan glucosaminyl 1 as compared with their non–irradiated counterparts. Conclusions: Downregulation of matrix synthesis and upregulation of protease inhibitors and apoptosis promoting genes by electron irradiation may inhibit keloid development. This mode of therapy appears to exert a positive effect toward lowering the recurrence rate of keloid formation.

    Notes: Aim: Keloid lesions develope as a result of abnormal growth of dermal fibroblasts after the injury. On the other hand, cell division cycle (Cdc) 25 is a family of phosphatases that activate the cell cycle regulating cyclin-dependent kinases. The three members of this family, Cdc25A, Cdc25B, and Cdc25C act in different phases of the cell cycle. In this study, we examined the dgree of expression of these phosphatases in keloid fibroblasts. Methods: Primary cultures of keloid and adjacent normal dermal fibroblasts (n = 4) as well as frozen and paraffin-embedded keloid and normal dermal tissues (n = 12 and n = 17 respectively) were examined by Western blot and immunohistochemical analyses for the expressions of Cdc25A, Cdc25B, Cdc25C and phosphorylated Cdk2. Results: Cdc25A protein levels were frequently increased in keloid fibroblasts as compared to the adjacent normal-appearing fibroblasts or different normal dermal fibroblasts. In contrast, Cdc25B and Cdc25C were none or rarely expressed. The incrased levels of Cdc25A were associated with lower levels of its substrate, Cdk2, in a phosphorylated or inactive form. Conclusions: Taken together, our data show that Cdc25A protein levels increase in keloid fibroblasts and this increase may be sufficient to activate its substrate, Cdk2, and accelerate the cell cycling. These results may have implication for the development of strategies to silence Cdc25A activity after the wound healing as a therapeutic modality for the keloid lesions.

    Notes: Summary Carcinoma of the urachus is not as uncommon as previously considered. 157 cases diagnosed to have urachal carcinoma collected from the Japanese literature were reviewed and the findings were compared with those reported in the English literature. The incidence of the disease ranges from 0.55 to 1.2% of bladder tumours in Japan in contrast with 0.07 to 0.70% in the Western countries. Males accounted for 72% of the patients. The highest age incidence occurred between fifth and sixth decades. The commonest presenting symptom was haematuria (71%). Adenocarcinoma accounted for 88% of the tumours most being mucous producing. Various treatment modalities were used, however, and the prognosis remained uniformly poor. An analysis of 66 patients with known outcome revealed an overall 5-year survival rate of 6%. The authors conclude that the disease tends to have a relatively higher incidence in Japan (Far East). When comparing the parameters of sex, age, presenting symptom, histopathology, treatment and prognosis, urachal carcinoma appears to have the same characteristics in the Far East as in Western countries.

    Notes: Summary The distribution of A, B, O(H) blood group antigens was studied in paraffin sections of 4 and 11 specimens, respectively, of normal skin and squamous cell carcinoma of the penis using specific red cell adherence (SRCA) test. In the normal skin of the penis the antigens appeared on the uppermost cornifying layers, while basal and malpigian layers were devoid of antigens. In squamous cell carcinoma of the penis the antigens were either absent or confined only to the epithelial pearl formations. Each of the 4 normal skins and 3 of the 11 carcinomas were antigen positive. As the blood group antigens were frequently absent in patients with carcinoma of the penis regardless of the tumor grade and stage as well as patient's condition, they lack the power to discriminate invasive potential.

    Notes: Abstract In order to optimize collagen antigen unmasking in paraffin-embedded tissue sections, the effects of various fixatives and duration of fixation in relation to enzyme pretreatment and microwave irradiation for collagen antigen unmasking were studied. A streptavidin--biotin-- peroxidase complex method was used for the immunolocalization of type III and IV collagen antigens. Fixatives and fixation time had significant adverse effects on the immunoreactivity of the antigens. Enzyme pretreatment was found to be superior to microwave irradiation for collagen antigen unmasking. Fixation with paraformaldehyde required shorter enzyme pretreatment and yielded a more enhanced reaction than treatment with formalin and Bouin's fluid. The optimum conditions for type III and IV collagen unmasking were found to be fixation with 4% paraformaldehyde in 0.01 m phosphate-buffered saline, pH 7.4, for up to 3 weeks followed by enzyme pretreatment with 1 mg ml−1 pepsin in 0.01 n hydrochloric acid, pH 2.0, for 30 min (human tissues) or 60 min (rat tissues) at 37°C. It is concluded that collagen antigen unmasking by enzyme pretreatment in tissue sections fixed for a long period of time can be successful if appropriate enzyme(s) and incubation time(s) are employed with regard to the antigen under study and fixative and fixation time used for tissue preparation
