    Notes: Abstract Renal function at the brush border membrane level has been studied using characteristic enzymes, such as alkaline phosphatase, leucine-aminopeptidase and gamma-glutamyl transpeptidase. Urinary enzyme studies were performed using leprosy patients, classified on the basis of bacteriological index (BI〉3; n=20,BI〈3; n=12, BI-ve; n=10) and compared with control subjects (n=10). The role of enzymuria in monitoring WHO-recommended multidrug therapy (MDT) has been evaluated in these patients. A significant increase in the enzyme activities (p〈0.01), as well as significant (p〈0.01) proteinurea in 24-hour urine samples of both the smear positive groups (BI〉3,BI〈3) prior to therapy compared to control subjects, indicates proximal tubular functional impairment at brush border membrane level. In the smear negative (BI-ve) grou, no significant difference was observed in enzyme activities as compared with the control group. In a follow-up study (BI〉3; n=13, BI〈3; n=4) the activities of all the enzymes decreased significantly in all the groups when compared to a corresponding untreated group. The follow-up study was not carried out on the smear negative group. The surprising finding was the differential behaviour of r-glutamyl transpeptidase, whose activity increased significantly (p〈0.01) even after therapy inBI〉3 group when compared with untreated patients. However in a detailed work-up including hepatic and renal function tests, the serum biochemistry was found to be normal both before and after therapy. Urinary excretion of brush border enzymes seems to be related to bacterial load, and their potential in studying the effect of MDT remains unclear.

    Notes: Abstract Both humoral and cell-mediated immune responses were studied in reactivated toxoplasma infection in mice. The infection was established by immunosuppressing chronically infected animals by thymectomy followed by irradiation. The reactivated infection was confirmed by reappearance of toxoplasma tachyzoites in peritoneal exudate. The animals survived until the 8th day post thymectomy. The percentage of splenic T lymphocytes was depressed after 4th day post thymectomy in both the uninfected thymectomised and infected immunosuppressed animals as compared to the intact uninfected controls and intact chronically infected animals. However, there was no significant change (P〉0.05) between the uninfected immunosuppressed control and infected immunosuppressed mice. When the infection progressed to more acute phase there was significant depression (P 〈 0.01) in the percentage of T lymphocytes in comparison to the uninfected thymectomised mice. However, there was no change in the percentage of B lymphocytes (P〉0.05). The function of lymphocytes as assessed by blast transformation was depressed in the presence of nonspecific mitogens, i. e. phytohemagglutinin and concanavalin A until the end of the study. The response to toxoplasma antigen was increased during early stages in the infected immunosuppressed mice (stimulation index = 4.15±0.1 and 4.5±0.15 on 4th and 6th day post thymectomy). On the 8th day post thymectomy, there was a sudden fall in the stimulation index (3.5±0.11). The antitoxoplasma antibodies as detected by indirect hemagglitinin and fluorescence analysis and enzyme-linked immunosorbent assay (ELISA) tests and toxoplasma-specific IgM antibodies as detected by ELISA test were present throughout the study period though at low levels. The significance of these findings is discussed.

    Notes: Abstract Administration of nifedipine to mice over a period of six months caused a significant (p〈0.05) decrease in neutrophilic functions viz superoxide generation, coupled to NADPH oxidase activity as well as NADPH production by HMP shunt. Properties like chemotaxis and phagocytosis showed a similar decrease. From this study, it is seen that nifedipine causes neutrophil functional abrogation which is therefore an apparent concern for the prolonged usage of the drug. However, relevance of the mouse model to clinical situation needs further investigation.

    Notes: Abstract The ability of an iron chelator, desferrioxamine, to inhibit the infarct size in in vivo rat heart was assessed. Anaesthetised rats were subjected to coronary artery ligation (CAL) for 72 hr and infarct size was measured macroscopically using TTC staining. Systolic blood pressure and ECG were monitored. Desferrioxamine (10 mg/kg and 20 mg/kg i.v.) administered half an hour after CAL markedly reduced the infarct size. However, drug treatment did not alter the systolic blood pressure of animals. In addition, desferrioxamine in vitro and in vivo demonstrated an inhibition of rat PMN-evoked and luminol-enhanced chemiluminiscence. The capacity of desferrioxamine to impair the generation or to scavenge directly oxygen free radicals may be responsible for its beneficial effect on myocardial infarct size in rats.

    Notes: Abstract The exposure of cardiac cells to OFR generated artificially, showed a marked decrease (p 〈 0.01) in cellular utilization of glucose alongwith a significant decrease in calcium uptake (p 〈 0.05). We have also provided evidence for a direct relationship of neutrophil OFR production with the extent of myocardial ischemia in patients of myocardial infarction. Our data provides evidence for implication of OFR in myocardial injury and the pivotal role played by modulators like calcium, ECGF and prostaglandins in potentiating damage to the myocardium.

    Notes: Abstract Membrane abnormalities in essential hypertensives (EH) are well known. The respiratory burst enzyme, NADPH oxidase is located in the cell membrane of the neutrophil (PMNLs) and its activity is important in generation of oxygen derived free radical (OFR). Recently OFR have been implicated in vascular changes in variety of conditions. An attempt was made to delineate the status of OFR and antioxidants in EH. Ten, age and sex-matched, healthy controls (GpI) and 26 untreated EH (Gp IIA mild-8, Gp IIB Moderate-8, Gp IIC Severe-10) were studied. After clinical examination and basic laboratory evaluation of subjects, neutrophils isolated from their blood were studied. Chemiluminescence (CL) emitted by PMNLs after stimulation was measured (counts/min) in a luminometer and was taken as measure of OFR production and thereby of NADPH oxidase activity. The levels of antioxidants, super oxide dismutase (SOD) and reduced glutathione (GSH), were also estimated. Chemiluminescence was increased significantly (p 〈 0.01) in Gp IIC (243.04 ± 24.9 × 103 counts per minute) as compared to Gp IIA (2.80 ± 1.87), Gp IIB (34.54 ± 30.24) and Gp I (0.52 ± 0.15) and SOD was reduced significantly (p 〈 0.05) in all EH (Gp IIA 3.9 ± 0.3 units per mg protein, Gp IIB 3.5 ± 0.3 and Gp IIC 3.12 ± 0.3) as compared to controls (4.1 ± 0.2). Similarly GSH was reduced (p 〈 0.05) in EH (Gp IIA 11.2 ± 1.7 mg per gm protein, Gp IIB 8.5 ± 1.1 and Gp IIC 6.6 ± 0.3) as compared to Gp I (13.5 ± 2.5). In essential hypertensives a curvilinear positive correlation was obtained between CL and both systolic (r = 0.7077, p 〈 0.01) and diastolic (r = 0.7965, p 〈 0.01) blood pressure. A significant inverse correlation (p 〈 0.05) was obtained between systolic and diastolic blood pressure on one hand and GSH and SOD on the other. Thus PMNLs of EH have increased emission of CL and depletion of antioxidants. The results indicate that in essential hypertension increased membrane NADPH oxidase activity is present.

    Notes: Abstract When the outer membrane proteins (OMP) of Bacteroides fragilis ATCC 23745 grown under seven different culture conditions were analysed by SDS-PAGE, five of the seven preparations showed similar protein profiles. When the three different OMP preparations were subsequently analysed by Western blotting, that from B. fragilis grown in brain heart infusion broth supplemented with haemin and menadione alone showed more immunodominant bands than those grown with additional cholesterol and methionine or cholesterol and cysteine. Only the former showed immunodominant proteins around 45.6 kDa and 66.2 kDa throughout the course of infection in mice. A protein corresponding to 97.4 kDa was found under all culture conditions. The results emphasise the importance of culture conditions in the expression of immunologically reactive antigens and also the possible use of such antigens in the serodiagnosis of B. fragilis infection.

    Notes: Abstract A crude outer-membrane protein (OMP) preparation from a strain of Bacteroides fragilis, grown in supplemented brain-heart infusion broth, was tested for its protective effect against subcutaneous infection in mice. Immunization with six doses, each of 100, 150 or 200 μg OMP preparation, gave some protection: abscesses completely disappeared 15 to 22 days after immunization. In non-immunized animals and animals immunized with doses of 10, 20, 40 or 80 μg each, well demarcated abscesses were seen beyond day 22 post-immunization. Although crude OMP elicited good antibody response, with maximum titres on day 4 post-immunization, high titres could not be associated with healing of the abscesses.
