Notes: Summary 1. The physical and chemical properties of substance P (extracted from bovine brain) and physalaemin were investigated by several methods. 2. During paper electrophoresis at pH 4.95 substance P migrates towards the cathode, physalaemin remains at the starting point. 3. During carrier-free glucose gradient electrophoresis substance P moves to the cathode whereas physalaemin remains at the starting point. 4. Electrofocusing of physalaemin reveals an isoelectric point of pH 7.0 whereas substance P has an isoelectric point of pH 10.0 to 10.5. 5. The biological activity of physalaemin was reduced by 70% after treatment with diazotized p-nitraniline, but that of substance P and eledoisin was not reduced. All three peptides were inactivated by treatment with phenylisocyanate. It was concluded therefore, that phenolic hydroxyl groups are not essential for the biological activity of substance P.

Description / Table of Contents: Zusammenfassung 1. Substanz P, Physalämin und Eledoisin bewirken beim Huhn Speichelsekretion. 2.
Notes: Summary 1. Substance P, physalaemin and eledoisin cause salivary secretion in the hen. 2. Atropine, phentolamine, propranolol and antazoline did not inhibit the peptide induced salivation. The peptide-induced rise in blood pressure was, however, blocked by large doses of phentolamine. 3. In contrast to the substance P preparation (containing only fraction Fa), Zetler's fraction Fc was devoid of sialogogic and hypertensive action. 4. Bovine parotid gland contains a peptide fraction with activity on smooth muscle but different from substance P. 5. Sustance P and eledoisin can clearly be discriminated by their relative activity on salivation in the chicken and the rat and on the guinea pig ileum. The discrimination between substance P and physalaemin in these tests is much less marked and near the limit of significance.
