    Notes: Abstract Experiments on the effect of cyclic AMP on Na+ and Cl− transport pathways were performed on isolated, perfused preparations of the posterior, osmoregulating gills of the euryhaline Chinese crabEriocheir sinensis. The crabs were caught during the autumn of 1987 in freshwater lakes near Emden (Federal Republic of Germany). N6-2′-O-dibutyryl adenosine 3′:5′-cyclic monophosphate (db-cAMP), 3-isobutyl-1-methylxanthine (IBMX), and 7β-acetoxy-8,13-epoxy-1α, 6β, 9α-trihydroxy-labd-14-ene-11-one (forskolin) induce an increase in the influx of both Na+ and Cl− in the gills. They also induce significant hyperpolarization. This last result indicates that the increase in NaCl uptake is related to an activation of both the Na+/K+ pump and the Cl−channels located at the serosal side of the gill epithelium. The possibility that cAMP acts as a second messenger, mediating the effects of different neuroendocrine factors involved in ionic and osmotic regulation in crustaceans is discussed.

    Notes: Abstract The possible involvement of protein kinase C in control of ion transport was investigated on a preparation of isolated, perfused posterior gills of the Chinese crabsEriocheir sinensis (collected in 1989 from lakes near Emden, northern Germany) acclimated to fresh water. 1-oleyl-2-acetyl-sn-glycerol (OAG) and phorbol 12-myristate 13-acetate (PMA), two activators of protein kinase C, when added to the perfusion saline, induced depolarisation of the transepithelial potential difference (PD) and an increase in transepithelial Na+ influx. The observed increase was proportional to OAG concentration up to 250µM, with a 2.5× accelerated Na+ influx. OAG and PMA remained without effect on Cl− fluxes. The observed effects were in agreement with an activation, via protein kinase C, of the Na+/K+ ATPase located on the serosal side of the epithelium.

    Notes: Abstract The possible involvement of calmodulin-dependent processes in the control of Na+ and Cl- transport pathways has been investigated on isolated, perfused preparations of salt-transporting posterior gills of the euryhaline Chinese crab Eriocheir sinensis (collected near Emden, Germany in autumn 1990). The anti-calmodulin phenothiazine drugs Chlorpromazine and Trifluoperazine induced depolarization of the transepithelial potential only when added to the serosal bathing saline (socalled “in”). This effect is best interpreted by assuming a disturbance of the conductive Cl- pathways located at the baso-lateral side of the epithelium. In agreement with that conclusion is the fact that Trifluoperazine inhibits the Cl- transepithelial influx. Trifluoperazine also induces inhibition of the Na+ influx when added either to the incubation (out) or to the perfusion (in) medium. These results indicate inhibitory effects of the anticalmodulin drug on both the Na+/K+ pump and leak system located at the serosal side and on the Na+/H+ exchange located at the apical side of the epithelium.

    Notes: Summary Electrical breakdown of cell membranes is interpreted in terms of an electro-mechanical model. It postulates for certain finite membrane areas that the actual membrane thickness depends on the voltage across the membrane and the applied pressure. The magnitude of the membrane compression depends both on the dielectric constant and the compressive, elastic modulus transverse to the membrane plane. The theory predicts the existence of a critical absolute hydrostatic pressure at which the intrinsic membrane potential is sufficiently high to induce “mechanical” breakdown of the membrane. The theoretically expected value for the critical pressure depends on the assumption made both for the pressure-dependence of the elastic modulus of the membrane and of the intrinsic membrane potential. It is shown that the critical pressure is expected at about 65 MPa. The prediction of a critical pressure could be verified by subjecting human erythrocytes to high pressures (up to 100 MPa) in a hyperbaric chamber. The net potassium efflux in dependence on pressure was used as an criterion for breakdown. Whereas the potassium net efflux was linearly dependent on presure up to 60 MPa, a significant increase in potassium permeability was observed towards higher pressure in agreement with the theory. The increase as indicated by measurements in which the same erythrocyte sample was subjected to several consecutive pressure pulses. Temperature changes in the erythrocyte suspension during compression and decompression were so small (less than 2°C) that they could not account for the observed effects.

    Notes: Abstract A total of 22 women with fibroadenomas had preoperative dynamic MR study (T1-weighted images every 47 s after injection of Gd-DOTA). Their age, hormonal status, breast MR studies and histopathological slides were retrospectively reviewed. Eleven pre- (n = 2) or post-menopausal (n = 9) women showed no early contrast enhancement. The absence of early contrast enhancement correlated with hyalin stromal component. Eleven pre- (n = 7) or post-menopausal (n = 4) women showed focal (n = 9) or diffuse (n = 2) early contrast enhancement. Early focal contrast enhancement correlated with myxoid (n = 9), mixed hyalin/myxoid (n = 1) or hyalin (n = 1) fibroadenomas. Early diffuse contrast enhancement of the breast correlated with myxoid (n = 1) or hyalin (n = 1) stromal component associated with proliferative fibrocystic disease of the breast parenchyma. The presence of contrast enhancement correlated with myxoid fibroadenomas, whereas absence of contrast enhancement correlated with hyalin fibroadenomas. As hyalin fibroadenomas occurs in post-menopausal women, the diagnostic accuracy of dynamic MRI may be improved in this age group.

    Notes: Abstract The purpose of our work was to describe the clinical and mammographic patterns of breast cancer in women aged 35 years old or less. From 1980 to 1985, 92 women 35 years old or younger were treated in our institution for breast cancer. Their mammograms were reviewed to determine the breast density and to describe the mammographic lesion. The clinical examination found a palpable breast mass (n = 84), nipple discharge (n = 2), axillary lymph node involvement (n = 3), metastasis (n = 1) or was normal (n = 2). The mammograms exhibited dense (n = 59), mixed dense/fatty (n = 29) or fatty (n = 4) parenchymal breast. The mammograms showed opacity with well-defined (n = 13) or ill-defined borders (n = 34), spiculated opacity (n = 11), architectural distortion (n = 9), isolated clusters of microcalcifications (n = 13) or were normal (n = 18). The diagnosis of breast cancer in young womensis difficult, because mammographic accuracy is less reliable and benign disease is far more prevalent.
