    Notes: Zusammenfassung Durch vitalmikroskopische Beobachtungen wurde das Verhalten von Fluorescenzfarbstoffen in der Backentasche des Goldhamsters untersucht. Sowohl nach lokaler wie nach intravenöser Zufuhr verschiedener Fluorochrome stellte sich ein Stofftransport über Lymphgefäße nicht dar. Proteingebundene Fluorochrome blieben im Backentaschengewebe lange Zeit liegen. Das Fehlen von Lymphgefäßen erklärt die immunologische Besonderheit der Hamsterbackentasche.

    Notes: Abstract  LL2 is an anti-CD22 pan-B-cell monoclonal antibody which, when radiolabeled, has a high sensitivity for detecting B-cell, non-Hodgkin’s lymphoma (NHL), as well as an antitumor efficacy in therapeutic applications. The aim of this study was to determine whether intracellularly retained radiolabels have an advantage in the diagnosis and therapy of lymphoma with LL2. In vitro studies showed that iodinated LL2 is intracellularly catabolized, with a rapid release of the radioiodine from the cell. In contrast, residualizing radiolabels, such as radioactive metals, are retained intracellularly for substantially longer. In vivo studies were performed using LL2-labeled with radioiodine by a non-residualizing (chloramine-T) or a residualizing method (dilactitol-tyramine, DLT), or with a radioactive metal (111In). The biodistribution of a mixture of 125I (non-residualizing chloramine-T compared to residualizing DLT), 111In-labeled LL2 murine IgG2a or its fragments [F(ab′)2, Fab′], as well as its humanized, CDR-grafted form, was studied in nude mice bearing the RL human B-cell NHL cell line. Radiation doses were calculated from the biodistribution data according to the Medical International Radiation Dose scheme to assess the potential advantage for therapeutic applications. At all assay times, tumor uptake was higher with the residualizing labels (i.e., 111In and DLT-125I) than with the non-residualizing iodine label. For example, tumor/blood ratios of 111In-labeled IgG were 3.2-, 3.5- and 2.8-fold higher than for non-residualizing iodinated IgG on days 3, 7 and 14, respectively. Similar results were obtained for DLT-labeled IgG and fragments with residualized radiolabels. Tumor/organ ratios also were higher with residualizing labels. No significant differences in tumor, blood and organ uptake were observed between murine and humanized LL2. The conventionally iodinated anti-CD20 antibody, 1F5, had tumor uptake values comparable to those of iodinated LL2, the uptake of both antibodies being strongly dependent on tumor size. These data suggest that, with internalizing antibodies such as LL2, labeling with intracellularly retained isotopes has an advantage over released ones, which justifies further clinical trials with residualizing 111In-labeled LL2 for diagnosis, and residualizing 131I and 90Y labels for therapy.

    Description / Table of Contents: Summary Two golden hamsters and two rabbits from a larger group of animals transplanted intracerebrally with a human mucin secreting carcinoma of the colon maintained in the hamster cheek pouch were found to have foreign-body reactions in their brains resembling tuberculoid granulomas. It is suggested that this inflammatory reaction was induced by regressive changes in the tumor and by mucin it secreted, whereby not only was the foreign-body effect of the tumor significant but also its breakdown products, as phospholipid-containing substances. Hence, these reactions are by and large tumor-specific.
    Notes: Zusammenfassung Bei je zwei Goldhamstern und Kaninchen, denen zusammen mit einer größeren Zahl weiterer derartiger Tiere Suspensionen menschlicher Darmcarcinome, die zuvor in der Hamsterbackentasche gezüchtet und hier als Gallertcarcinom gewachsen waren, ins Gehirn transplantiert wurden, zeigten sich gegen das im Gehirn proliferierte Geschwulstgewebe Abwehrreaktionen vom Typ tuberkuloider Granulationen. Es wird angenommen, daß diese durch regressive, möglicherweise vor allem am Tumorschleim aufgetretene und damit weitgehend tumorspezifische Veränderungen des Geschwulstgewebes ausgelöst wurden, und daß dabei sowohl eine Fremdkörperwirkung des regressiven Geschwulstgewebes als auch Zerfallprodukte desselben in Form phospholipoidhaltiger Substanzen eine Rolle spielten.

    Description / Table of Contents: Zusammenfassung Transplantationen menschlicher Hirntumoren in Goldhamstergehirne führten in diesen regelmäßig zu Abwehrreaktionen in Form einer chronisch-proliferativen Entzündung im leptomeningealen Bindegewebe. Im Hirngewebe waren derartige proliferative Entzündungserscheinungen inkonstant und in der Regel nur gering entwickelt. Bei keinem der 23 transplantierten Tumoren konnte ein dauerndes Wachstum beobachtet werden. Lediglich bei 5 Transplantattumoren (3 Glioblastome, 1 Oligodendrogliom und 1 amelanotisches Melanoblastom) fanden sich in den ersten Tagen nach der Transplantation Tumorzellproliferate im Bereich der Hirnkammern. In Übereinstimmung mit Murhpy u. Sturm (1923) sowie Greene (1951, 1953) wird angenommen, daß Angehen und Wachstum der Transplantate wesentlich davon abhängig ist, ob diese im Hirngewebe oder im Liquor angesiedelt werden, da sich im Hirngewebe mesenchymale Abwehrreaktionen nur in geringerem Grade zu entwickeln vermögen als im Liquor. Andererseits wird es jedoch für wahrscheinlich gehalten, daß der Liquor sich primär besser als Wachstumsmilieu eignet als das Hirngewebe.
    Notes: Summary The transplantation of human brain tumors in the brain of the golden hamster regularly resulted in foreign-body reactions as a chronic, proliferative, inflammation in the leptomeningeal connective tissue. In the brain tissue itself, however, such inflammatory phenomena developed seldomly and only weakly. No continuous heterologous growth could be found in any of the 23 tumors transplanted intracerebrally. In only five cases (3 glioblastomas, 1 oligodendroglioma, and 1 amelanotic melanoma) could tumor cell proliferation be documented in the cerebral ventricles during the first days after implantation. In agreement with the findings of Murphy and Sturm (1923), and Greene (1951, 1953), the success of such transplantations depends mainly on whether the xenogeneic tumor cells come to rest in the brain tissue proper or in a cerebrospinal fluid-containing space, since the mesenchymal foreign-body reaction of the latter localization is certainly the more pronounced of the two. On the other hand, it is proposed that the cerebrospinal fluid is a more suitable milieu for tumor proliferation than brain tissue.

    Description / Table of Contents: Zusammenfassung Durch die lokale Injektion von Fluorescenzfarbstoffen in das lockere Bindegewebe der hervorgestülpten Hamsterbackentasche wurden Ausbreitung und Abtransport im Gewebe vitalmikroskopisch untersucht. Das frei wasserlösliche Fluorescein breitet sich in Gestalt perivasaler Streifen schnell in Richtung zur Basis der Backentasche hin aus, ohne dabei in das Blut überzutreten. Lymphgefäße gelangen nicht zur Darstellung. Das sich in vivo an Protein bindende Brillantsulfoflavin zeigt den streifenförmigen Abstrom verzögert und spärlich. Das in vitro an Plasmaproteine fest gekoppelte Fluorescein-Isothiocyanat bleibt während einer Beobachtungszeit von mehr als 30 min ohne Ausbreitung und Abstrom im Gewebe liegen. Der Stofftransport wasserlöslicher Substanzen im interstitiellen Gewebe der Hamsterbackentasche ist somit gerichtet und perivasal lokalisiert. Ein Abstrom über Lymphgefäße fehlt. Plasmaproteine bleiben im extravasalen Raum lange Zeit liegen.
    Notes: Summary The spread and flow of fluorescent dyes injected into the wall of the hamster cheek pouch was examined by vital microscopic means. Water-soluble fluorescein spreads very quickly in the form of perivascular streaks in the direction of the base of the cheek pouch, without entering into the blood stream. Lymph vessels, however, could not be identified. Brillant-sulfoflavine which binds proteinin vivo showed much slower and reduced flow phenomena. Fluorescein isothiocyanate coupled to plasma proteinsin vitro remained, during an observation period of more than 30 min, stationary in the cheek pouch tissue without any evidence of transport to be seen. The transport of water-soluble substances in the cheek pouch tissue is thus perivascular in nature. Lymph flow is not to be documented by this technique. This is further confirmed by the observation that fluorescent-labelled plasma proteins remain unmoved in the extravascular space of the cheek pouch tissue for a long period of time.

    Notes: Zusammenfassung Die Aktivität der Glukose-6-phosphat-Dehydrogenase (G-6-PD) wurde in Extrakten des heterotransplantierten Humantumors GW-39 nach cytostatischer Therapie mit Actinomycin C untersucht. Es kam zu einer signifikanten Hemmung der G-6-PD-Aktivität in den Tumoren, während die Enzymaktivität in der Leber der tumortragenden Wirtstiere (Hamster) unbeeinflusst blieb. Diese Ergebnisse entsprechen unseren, an einem histopathologisch ähnlichen Humantumorsystem (H.Ad. No. 1) früher erhobenen Befunden und bestätigen damit den tumorspezifischen Enzymeffekt der Behandlung mit Actinomycin C.
