    Notes: Summary The effects on tentacle regeneration inHydra of two DNA-antimetabolites, bromouracil and bromodeoxyuridine, have been studied. Bromouracil inhibits tentacle regeneration inH. vulgaris andH. viridis; bromodeoxynridine inhibits tentacle regeneration inH. vulgaris but notH. viridis over the range of concentrations studied. Thus these two species appear to differ in their susceptibilities to these drugs, and the drugs may have different efficacies in this system.

    Notes: Summary We have cloned part of theETS 1 proto-oncogene and demonstrated the presence of two polymorphic Sst I restriction sites. A probe derived from one of our clones revealed the presence of 8.3 kb, 9.5 kb and/or 11.5 kb fragments on Southern blots of human DNA samples. The relative frequencies of these alleles appear to be significantly different between Saudi and Western populations, but there are no apparent differences in these frequencies between Saudi non-leukemic and leukemic individuals.

    Notes: Abstract 2D-gel electrophoresis was used to investigate protein synthesis in leukaemic cells from a series of 15 chronic lymphocytic leukaemia (CLL) patients, and in non-malignant B-cell populations from different sources. The protein synthesis profiles of CD5+ B-cells from umbilical cord blood and from tonsil were determined, and the levels of expression of their proteins were observed to be similar to the CLL cells. The CD5-cells from cord blood resembled peripheral blood B-lymphocytes, and the protein synthesis profile of CD5-cells from tonsils was very complex. One protein was also identified which consistently appeared to be synthesised at a low level in CD5+ B-cells from tonsil but which was always more prominant in CLL cells and other non-malignant B-lymphocytes. On the basis of these data it is possible that the closest non-malignant counterpart to CLL is the CD5+ B-lymphocyte from cord blood.

    Notes: Summary Polyamine accumulation was related to RNA biosynthesis and content during the cell cycle of CHO-K1 and HeLa cells. The highest correlations were observed between polyamine accumulation and RNA content. Feeding, serumstarved cultures of embryonic chick fibroblasts, which exhibit a 3-fold increase in RNA content, demonstrated that RNA content was most closely paralleled by spermidine content.

    Notes: Summary Synchronized cultures of mitotic HeLa cells were obtained by different protocols and the polyamine content of these cells determined. It was found that the method of synchronization can significantly change the polyamine content of the mitotic cells, and can also alter the time course of polyamine accumulation during the subsequent cell cycle.

    Notes: Summary We have transfected high-molecular-weight DNA from human thyroid carcinomas into murine 3T3 cells. As a result we identified several foci of morphologically distinct transformed cells in each of the tumour DNA transfected cultures. After a total of three rounds of transfection, the transformed cells were shown to form tumours in nude mice. Southern blot analysis of DNA prepared from third-round transfectants demonstrated the presence of human Alu repetitive sequences and, after hybridization with probes for known oncogenes, indicated the presence of the humanH-RAS oncogene in 3T3 cells transfected with three out of four anaplastic carcinoma DNA samples. It appears therefore that activation ofRAS genes may be an important event in the development of the anaplastic thyroid tumours.

    Notes: Abstract We have used the polymerase chain reaction (PCR)-based technique of differential display to analyse changes in gene expression during ageing of the rat brain. In this approach we have compared three young adult (6 months) with three old adult (20 months) animals. RNA preparations from the homogenised brains were subjected to reverse transcriptase (RT)-PCR using 36 different combinations of primer pairs. Any PCR product which was consistently found to be more prominent in the three young brains compared to the three old brains, and vice versa, was scored as potentially representing a gene which was differentially expressed during the ageing of this tissue. Out of a possible 2000+PCR products we identified 44 that might represent genes that exhibit differential expression during ageing of the rat brain. An initial screen of these fragments, by Southern-blotting the PCR products and hybridising them with cDNA probes derived from either young or old brain RNA preparations, indicated that 40% of them represented genes that were differentially expressed. This approach is likely to prove invaluable for identifying cohorts of genes that show differential expression during the ageing process.
