    Notes: The dielectric behavior of the aqueous solutions of three widely differing macromolecules has been investigated: myoglobin, polyvinylpyrrolidone (PVP), and human serum low-density lipoprotein (LDL). It was not possible to interpret unambiguously the dielectric properties of the PVP solution in terms of water structure. The best interpretation of the dielectric data on the myoglobin and LDL solutions was that, in both cases, the macromolecule attracts a layer of water of hydration one or two water molecules in width. For LDL, this corresponds to a hydration factor of only 0.05 g/g, whereas for myoglobin the figure is nearer 0.6 g/g. With myoglobin, part of the water of hydration exhibits its dispersion at frequencies of a few GHz, and the rest disperses at lower frequencies, perhaps as low as 10-12 MHz. The approximate constancy of the width of the hydration shell for two molecules as dissimilar in size as LDL and myoglobin confirms that the proportion of water existing as water of hydration in a biological solution depends critically on the size of the macromolecules as well as on their concentration.

    Notes: Measurements have been made of the permittivity and conductivity of normal and wounded human skin material over the frequency range 10 MHz-10 GHz. The permittivity of the wounded tissue was found to be about 12% higher than that of the normal tissue. A similar percentage increase was observed for the conductivity. These differences are attributed to the presence of a higher proportion of bulk water in the wounded material.

    Notes: Three independent dielectric methods for the measurement of water of hydration (bound water) in a biological material are described and discussed comparatively. For well-defined aqueous solutions of biological molecules, hydration can be obtained from direct observations made on the δ dispersion or from measurement of the dielectric values of the β dispersion. For whole tissue, however, neither of these two methods is applicable, and to deduce the hydration, it is necessary to use the third technique in which the volume of the hydrated biological particle is obtained by measuring the effect of it on the known dielectric properties of pure water. The hydration can then be calculated by deducting the volume of the anhydrous particle from the experimentally determined volume of the hydrated particle. Owing to possible systemmatic errors the uncertainty in the absolute hydration value associated with this technique is rather larger than that obtained with the other two dielectric methods. For studying the differences between hydration in similar tissues, however, this objection disappears.

    Notes: The dielectric properties of developing rabbit brain were measured at 37°C between 10 MHz and 18 GHz using time domain and frequency domain systems. The results show a variation with age of the dielectric properties of brain. An analysis of the data suggests that the water dispersion in the brain of newly born animals can be represented by a Debye equation. This dispersion increases in complexity with age, and there is evidence of a smaller additional relaxation process centered around 1 GHz. It is concluded that the principal contribution to this subsidiary dispersion region arises from water of hydration.

    Notes: Equations are derived which account for the effect of an applied electric field on the fluctuation of protons associated with a macromolecule. The contribution of this proton polarization to the complex permittivity of the macromolecules is evaluated in terms of its effect on both the dispersion due to proton fluctuation and the dispersion caused by dipolar rotation. An expression for the fluctuation correlation time τiδ is derived in terms of the mean lifetimes of the ionised and unionised state τ0 and τ+. If τiδ is very much less than the dipolar correlation time τi, the fluctuation dispersion will occur at much higher frequencies than the dispersion due to orientation polarization; hence the dispersions will be well separated. If τiδ » τi the two regions would overlap and would be indistinguishable as separate entities. At present insufficient data are available to test rigorously these conclusions but the potentialities of the theory in relation to small globular proteins is shown.

    Notes: Recent advances in dielectric theory are applied to two models representing an aqueous solution of dipolar macromolecules. In one model the water is treated as a dielectric continuum and the macromolecule as a finite-sized sphere; in the other both components are represented as point dipoles suspended in a background dielectric. The predicted frequency dependences of the complex permittivity in these two cases agree and the validity of the dielectric technique for estimating macromolecular size and shape is established. The model in which water is treated as a dielectric continuum predicts a larger dielectric dispersion in the radio frequency region, which is consistent with the experimental data available for myoglobin. The validity of the Debye formula for relaxation time and the effect of “dielectric friction” in macromolecular solutions are also discussed.

    Notes: The relative permittivity of aqueous solutions of human serum low density lipoprotein (LDL) and partially trypsin digested lipoprotein (T-LDL) has been determined for various concentrations at 20°C over the frequency range 0.15-100 MHz. Comparison of the dielectric dispersion curves for the digested lipoprotein with those for the native preparation revealed a larger low-frequency dielectric increment, which may be attributed to an increase in the number of counterions moving over the surface of the molecule. An explanation of this observation is an elevation of 70% in the net negative charge on the surface of the trypsin-treated particle as compared to its native counterpart.
