    Notes: Abstract A study of the sorption of d-metal ions by titanium phosphate on a platinum electrode shows that the potential of the platinum electrode significantly influences the uptake of cations by the ion-exchanger. Cathodic polarization leads to a significant increase of the sorption whereas anodic polarization is accompanied by desorption. Therefore it is possible to perform cycles of sorption and desorption simply by switching the potential. The results obtained can be explained by the fact that the layer of titanium phosphate acts as a phase which extracts protons or hydroxide ions. The electrochemical generation of hydroxide ions at negative potentials can, especially at less acidic pH, lead to the formation of metal hydroxides in the ion-exchanger phase. This interference is less important at a lower solution pH.

    Notes: Abstract Interactions of Bovin Serum Albumin and Hemoglobin with an hydroxyapatite gel (HA-Ultrogel, Sepracor), have been studied separately in batch experiments. The adsorption isotherms are of the Langmuir type and can be used directly to scale column operations. For adsorption of hemoglobin alone, in column at pH 6.8 (equal to its isoelectric point) we notice that a classical intraparticle transfer model, based on a constant effective diffusion coefficient represents perfectly the symmetrical breakthrough curve. For acid pH values (pH 5.8), Langmuir isotherms of BSA and hemoglobin adsorptions showed a strong curvature, sign of a quite irreversible adsorption and breakthrough curves obtained under these conditions, exhibit a high dissymmetrical shape for both proteins. In that case, a model of diffusion based on the adsorption on two types of independent sites, with two intraparticle transfer coefficients, gives a good representation of the breakthrough for adsorption of both proteins separately. Binary mixtures of these components were prepared and injected in columns packed with the same support. Competitive Langmuir equation, based on the results obtained in monocomponent batch experiments, give a very good fit to our system. The intraparticle transfer in that case seems to be facilitated, and one effective coefficient alone is enough to predict the breakthrough curves obtained. This behaviour may be the result of an increase of the solution ionic strength, and of the smaller irreversibility feature of the adsorption when proteins are in competition.

    Notes: The limit regimes of equilibrium staged batch parametric pumps are studied. The half-cycle comprises several equilibrations at each temperature, each followed by a discrete transfer of fluid. The concentration distribution of the effluent of each half-cycle is conserved by a cascade of reservoirs. The concepts, methods, and McCabe-Thiele representation are first introduced on the example of the (one-stage, two transfer) pump. The generalization to the (one stage, n transfer) pump leads to a limit regime represented by a staircase construction between the two isotherms, the number of steps of the staircase being equal to the number of transfer steps. The limit regime of the general (N stage, n transfer) pump is then analyzed in terms of the properties of the (one stage, n transfer) pump considered above and of the (N stage, one transfer) pump discussed in the preceding paper of this series. An analytical solution for the separation factor is given in the case of linear isotherms. The importance of conserving the effluent concentration distribution is illustrated by graphical and numerical comparisons with pumps where this is not done.

    Notes: The theory of equilibrium staged parametric pumps with discrete fluid transfers has been presented in previous papers for total reflux. Here, it is extended to open, fractionating systems, operating in cyclic steady state. The cases of and enriching section, a stripping section, and a complete fractionating apparatus are successively considered. Providing adequate variables are defined, the result is a diagram which plays the same role as the McCabe-Thiele diagram in distillation. The optimal feed stage location, the minimum reflux, and the separation factor for given operating conditions are determined analytically and graphically in the case of linear equilibrium isotherms. The method is then generalized to non-linear equilibria, particularly of constant relative volatility type.

    Notes: Several techniques for protein extraction were tested for recovering penicillin acylase from a recombinant strain of Escherichia coli. These techniques include chemical [guanidine hydrochloride, Triton X-100, ethylenediaminetetraacetic acid (EDTA), ethanol/toluene], physical (sonication, freeze-and-thawing), and enzymatic (lysozyme) treatments. Best results were obtained with the combined use of guanidine and EDTA. This extraction procedure was optimized, and it was found that 95% of the enzyme was extracted after a 10 m/M EDTA plus 10 mM guanidine treatment at room temperature for 10 h. The purification factor was 25 when compared to disruption by sonication. This extraction method could avoid purification steps for particular applications. © 1994 John Wiley & Sons, Inc.
