    Notes: Summary The spinal cord and the thoracic and lumbar posterior root ganglia (PRGs) of 14 HIV-positive men and 7 age- and sex-matched controls were studied by routine histology, morphometric analysis of the number of nodules of Nageotte (nN) and the diameters of sensory ganglion cells, immunohistochemistry and in situ hybridization. In 7 patients (2 of whom had evidence of cytomegalovirus ganglionitis) there were increased numbers of nN and diffuse, mild infiltration with CD45R+ T lymphocytes; no B lymphocytes were observed. Macrophages were increased in number in all cases. Whenever more than one ganglion was examined from the same patient, the appearances were similar in all. There was no alteration in the distribution of ganglion cell diameters. Changes in the spinal cord included vacuolar myelopathy (5 cases), HIV myelitis (1 case), microglial nodules (3 cases) and pallor of the gracile tracts (GTP) in 7 cases, in 6 of whom it co-existed with increased numbers of nN. Seven cases had no abnormalities, except the increase in number of macrophages in PRGs. In spite of a correlation between sensory nerve cell loss and GTP our findings suggest that other mechanisms, such as ‘dying back’ may contribute to the pathogenesis of GTP. Moreover, sensory disturbances were found most commonly in association with nerve cell loss; however, loss of sensory ganglion cells was not necessarily associated with evidence of sensory impairment.

    Notes: Abstract  A cDNA clone (Gv1) was isolated from a Medicago truncatula/Glomus versiforme root cDNA library by a differential screening technique. Gv1 represents a full-length G. versiforme mRNA encoding a 99-amino-acid polypeptide. This shares regions of identity with HMP1, a non-histone, cruciform DNA-binding protein from Ustilago maydis. Northern blot analysis indicated that Gv1 transcripts were present in mycorrhizal roots of M. truncatula and M. sativa (alfalfa), but absent in uncolonized roots of both species. A reverse transcription/polymerase chain reaction (RT-PCR) technique detected transcripts in the roots of the M. sativa non-mycorrhizal genotype MN NN-1008 colonized with G. versiforme, as well as in external hyphae collected from the roots of M. sativa colonized with G. versiforme. A genomic copy of Gv1 was detected by PCR in DNA isolated from the roots of M. truncatula colonized with G. versiforme and in DNA isolated from spores of G. versiforme. The PCR product identified in the mycorrhizal DNA preparation was identical in sequence to the cDNA, except for the presence of two introns. A homologue of Gv1 was detected in a second arbuscular mycorrhizal fungus, Glomus intraradices.

    Notes: Abstract. The aim of this study was to develop methods of visualising optical fibres on MRI scans for monitoring interstitial laser therapy. Scans were performed on a specialised MRI extremity scanner at 0.17 T. Optical fibres of 0.4 mm diameter used for delivering laser energy were coated with iron particles from a superferromagnetic contrast agent. MR images of the fibres were acquired using gradient echo sequences (TR/TE = 300/10, 1 mm in-plane, 3 mm slice) and assessed for fibre visibility. Coated fibres could be resolved as lines 2 ± 1 mm wide using the gradient echo sequence. Uncoated fibres were invisible on the sequences used for in vivo therapy monitoring due to partial volume averaging. It is concluded that optical fibre visualisation by MRI may be improved by coating with ferromagnetic particles. Biocompatibility requires further assessment, but direct coating appears to be a promising method for fibre visualisation in MR-guided laser therapy.

    Notes: Malignant hyperthermia susceptibility is genetically heterogeneous. The ryanodine receptor gene on the long arm of chromosome 19 represents an important candidate gene but not all families with malignant hyperthermia demonstrate ryanodine receptor mutations or linkage to this region of 19q. Linkage to chromosome 17 in the region of the adult muscle sodium channel α subunit gene has been suggested in some families; others are not linked to either of these loci. For most families the in vitro muscle contracture test remains the only reliable method of predicting susceptibility to malignant hyperthermia. We have performed linkage analysis in a large family group with malignant hyperthermia in which the in vitro muscle contracture test had been carried out using the procedure standardised by the European Malignant Hyperthermia Group. None of the published ryanodine receptor gene mutations associated with malignant hyperthermia susceptibility were detected in affected individuals but linkage to intragenic ryanodine receptor markers strongly suggest that this gene is involved in malignant hyperthermia susceptibility in this family. This enabled accurate predictive testing by DNA analysis in 11 untested subjects at 50% risk.

    Notes: Physiological homeostatic mechanisms and interventions by anaesthetists attempt to moderate excessive change in many biological variables during anaesthesia. These mechanisms may have fast or slow response times. This study describes how mean arterial blood pressure changes with time and how the change is dependent upon the pre-existing blood pressure. The results demonstrate the ‘regression towards the mean’ concept; low arterial blood pressures increase and high pressures decrease. The data are the result of all interactions and have been used to produce an ‘envelope’ into which 80% of all changes fall. Alarm systems using this envelope could warn of excessive changes that occur within short time intervals.
