    Notes: Abstract:  Pemphigus foliaceus (PF) is a severe autoimmune bullous disorder, characterized by autoantibodies (autoAb) against desmoglein 1 (Dsg1). As T cells may be critical in the pathology of PF, the aim of the present study was to identify and characterize autoaggressive T-helper cells reactive to Dsg1 in PF patients and healthy individuals. Eight patients with the clinical diagnosis of PF and six HLA class II-matched healthy individuals were examined. By magnetic cell-sorting (MACS) cytokine-secretion assay, Dsg1-responsive T-helper (Th) 1 and Th2 cells were isolated and cloned by limiting dilution. The generated T-cell clones (TCC) were characterized regarding proliferative response, TCR Vβ-chain usage, and cytokine profile upon in vitro stimulation with Dsg1. Both Dsg1-reactive Th1 and Th2 cells were detected in PF patients and controls at similar frequencies. A total of 15 Th1 and Th2 clones were isolated from patients and 27 TCC from healthy controls. Analysis of TCR Vβ-chain usage of autoreactive T cells from both groups revealed no predominance of a specific Vβ chain. Noteworthy, the isolated TCC showed a polarized Th1- or Th2-like phenotype upon in vitro culture and stable expression of Th1 or Th2 cytokines during long-term in vitro culture. In summary, our data demonstrate that T-cell autoreactivity against Dsg1 is not restricted to patients with PF. Moreover, both Th1 and Th2 cells were present in patients and healthy donors, suggesting that the loss of B-cell tolerance against Dsg1 in PF is not exclusively determined by the presence of autoaggressive T cells.

    Notes: Two-photon laser-induced fluorescence spectroscopy is used to detect ground-state atomic hydrogen in highly diluted SiH4–H2 radio-frequency discharges currently used for microcrystalline silicon thin film deposition. Along with the fluorescence coming from the hydrogen atoms created in the discharge, additional atoms arising from photodissociation of silicon containing species at 205 nm are observed. This parasitic effect is identified as resulting mainly from the disilane molecules formed in the plasma. Hydrogen atoms created in the plasma and originating from the photodissociation are discriminated by a spectral analysis of the laser-induced fluorescence line profile and relative densities are deduced. Some results on the hydrogen atom and disilane concentrations as functions of the silane dilution, radio-frequency power, gas pressure, and gas temperature are given to illustrate the potentiality of such a diagnostic in SiH4–H2 discharges containing up to 6% of silane. © 2002 American Institute of Physics.

    Notes: Metal ions such as Ni2+, Co2+, Cu2+, or Cr3+ are haptens with a high immunogenic potential, as contact dermatitis caused by ionic metals occurs in about 10–15% of the human population. Since alloys containing Ni2+, Co2+, and Cr3+ are components of implants in replacement surgery, dentures, orthodontic wires, and various other devices, adverse reactions to metal ions create serious problems in practical medicine as incompatibility reactions to metal-containing biomaterials. On the other hand, contact dermatitis to metal ions such as Ni2+ is a well-established model for studying the molecular mechanisms involved in the recognition of haptens by the immune system. Although many investigations have been performed to elucidate the molecular interactions causing contact hypersensitivity in man, many aspects remain to be clarified. This review will focus on the experimental data accumulated so far on the immunologic mechanisms responsible for the recognition of metal ions by T cells and eliciting adverse immune reactions causing contact dermatitis.

    Description / Table of Contents: Abstract. The report deals with the clinical features of a 26-year-old man with the extraabdominal form of deep fibromatosis, which became manifest at the age of 18 years as acute ileus caused by diffuse intraabdominal fibromatous tumours. In the later course of the disease extensive fibromatous lesions developed, mainly at the skin. In addition, multiple keloids occurred after excision or after negligible traumas. The pathogenetic events of the clinical picture described may be linked to the activation of mesenchymal cells leading to the generation of either fibromatous tumours or posttraumatic keloids.
    Notes: Zusammenfassung. Vorgestellt wird ein 26jähriger Mann mit der extraabdominellen Form einer tiefen Fibromatose, die sich erstmalig im 18. Lebensjahr als akuter Ileus aufgrund ausgeprägter intraabdomineller Bindegewebstumoren manifestierte. Später traten flächenhafte, fibromatöse Bindegewebstumoren bevorzugt an der Haut auf. Neben diesen spontan entstandenen kutan und subkutan gelegenen Hauttumoren fanden sich auch Keloide nach Schnittverletzungen oder Bagatelltraumata an der Haut. Diesem Krankheitsbild liegt möglicherweise eine Aktivierung mesenchymaler Zellen zugrunde, die bei entsprechender Disposition zu fibromatösen Tumoren oder posttraumatischen Keloiden führt.

Cytochemische Untersuchungen an Liquorzellen ergaben beieitriger Meningitis (Meningokokken, Pneumokokken) in Segmentkernigen erhebliche Glykogeneinlagerung und Verminderung des Lipoidgehaltes der Granula, beiseröser Meningitis (Mumps, Poliomyelitis) in Segmentkernigen Lipoidverminderung, in mononucleären Zellen vermehrten Gehalt an basischen Proteinen und Glykogeneinlagerung, beituberkulöser Meningitis in Segmentkernigen Glykogenvermehrung, Lipoidverminderung in den Granula unter leichter paraplasmatischer Sudanophilie, in mononucleären Zellen starke Lipoidbeladung des Cytoplasmas. 2. Ergebnisse an Segmentkernigen wurden im Vergleich mit segmentkernigen, neutrophilen Granulocyten im Blut des erkrankten Kindes und gesunder Individuen ermittelt. Zur Beurteilung mononucleärer Liquorzellen wurden Lymphocyten und Monocyten herangezogen, einerseits da sie morphologisch nächstverwandte Zellen darstellen, andererseits da mittels cytochemischer Kriterien die Herkunft der mononucleären Liquorzellen geprüft werden sollte; in letzterem Zusammenhang ergaben sich viele Gemeinsamkeiten insbesondere mit Lymphocyten. Aber auch diese Ergebnisse erlauben keine definitive Einordnung. 3. Bei den untersuchten Infektionskrankheiten ergeben sich an Leukocyten des peripheren Blutes keine Veränderungen gegenüber der Norm. 4. Sogenannte Jodophilie ist auch an Liquorzellen festzustellen. Cytochemische Methoden beweisen, daß die durch Jod markierte Substanz Glykogen ist.

    Notes: Zusammenfassung 1. Der Nucleolus ist eine chemisch tätige Kernorganelle. Er ist schwammoder knäuelartig aufgebaut, enthält basische und andere Eiweißkörper, ferner RNS. Er ist Feulgen-negativ. Die in ihm gebildeten Substanzen sammeln sich auf seiner Oberfläche. Sie sind ein Nukleolipoproteid und verhalten sich Feulgenpositiv. Für den Stoffabstrom zur Kernmembran stehen mit großer Wahrscheinlichkeit Leitröhren zur Verfügung. Durchtritt durch die Kernmembran erfolgt in der Art eines „Durchschmelzens“. Der Nukleolarapparat der untersuchten Zellen besteht aus dem Nucleolus und den Feulgen-positiven Kerneinschlüssen der verschiedensten Lokalisation. 2. Eine vergleichende Wertung der Befunde der einzelnen Autoren ist nur in beschränktem Umfange möglich, da Material, Methoden und Gesichtspunkte der einzelnen Untersucher erheblich divergieren. Daraus folgt auch, daß eine allgemein gültige Definition des Nucleolus noch nicht gegeben werden kann. Für eine Übergangszeit wird vorgeschlagen, einerseits vonnukleären Blasen — beim Vorliegen unstrukturierter Depots mit Feulgen-positiver Randschale —, andererseits vonNukleolen in engerem Sinne — beim Vorliegen einer strukturierten Kernorganelle — zu sprechen.
