    Notes: Summary The karyotypes of the domestic cat and the tiger are presented. The diploid number for both species is 38. Autoradiographic studies on the cat chromosomes show that in females one X-chromosome is late replicating and in males the Y is late replicating. Other chromosomes bear late replicating segments.

    Notes: Abstract The regularly occurring secondary constrictions on metaphase chromosomes of mammalian cells prove to be nucleolus organizers as expected. The expression of nucleolus organizers as secondary constrictions, however, varies from cell to cell and from tissue to tissue, including cultivation in vitro. Electron micrographs of the organizer region show that the nucleolus organizer at metaphase is not a constriction. The width of the organizer area is the same as the condensed chromosomal arms; but the filaments, which are the major components of this region, show a diameter of 50–70 Å. The condensed chromosome arms consist of filaments 150–200 Å in diameter. In some mammalian species, structures similar to the nucleolus organizer are located at the end of chromosomes. These may be terminal nucleolus organizers.

    Notes: Abstract The constitutive heterochromatin, as demonstrated by the C band technique, may be subdivided into a number of categories when other characteristics are considered. The responses to fluorochromes QM and 33258 Hoechst, the behavior following G band staining, the repetitive DNA content, and many other criteria are useful for the classification of heterochromatin. The heterochromatin patterns of three mammalian species are presented to demonstrate that within each karyotype there may be several different types of C bands. In general, a correlation may also be made between GC-rich satellite DNA and dull (or negative) Q fluorescence, and between AT-rich satellite DNA and bright Q, fluorescence.

    Notes: Abstract The DNA replication patterns of the terminal S phase of three species of Mus were analyzed by tritiated thymidine autoradiography. The centromeric heterochromatin of M. fulvidiventris is the latest component to finish DNA synthesis. The Y chromosome finishes replication earlier than the centromeric heterochromatin. The centromeric heterochromatin of M. musculus, on the other hand, is not the latest component to finish DNA synthesis. At the very late S phase, grains are found in the euchromatic arms instead of the heterochromatic areas. The “hot X” and the “hot Y” can be identified in the majority of, but not all, cases. The heterochromatic short arms of the autosomes in M. dunni finish DNA replication earlier than many areas in the euchromatic long arms and the heterochromatin of the sex chromosomes. This indicates that in M. dunni there are at least two types of heterochromatin. The late-replicating zones in the euchromatic long arms are distinctly banded. This banded grain pattern can be seen in all Mus species observed, but in M. dunni it is most exaggerated. Late-replicating chromosome segments can be demonstrated also by 2+ cycles of BUdR incorporation and Giemsa staining.

    Notes: Abstract Using C-banded preparations of Mus dunni it is possible to study the behavior of constitutive heterochromatin in early stages of meiotic prophase. The X and the Y chromosomes, both of which contain a large amount of heterochromatin, lie apart in leptotene but move toward each other during zygotene. They then form the sex vesicle at late zygotene. In autosomes zygotene pairing appears to start from the telomeric ends. The centromere of the Y chromosome associates end-to-end with the terminal end of the long arm of the X chromosome. The autosomal heterochromatic short arms show forked morphology in certain bivalents at pachytene, suggesting probable incomplete synapsis.

    Notes: Abstract Silver staining of mammalian spermatocytes revealed, in light microscopy, synaptonemal complex and structures within the sex vesicle. It is feasible to follow the chromosome pairing phenomenon from zygotene to pachytene by examining the behavior of synaptonemal complexes. Nucleolus organizer regions take heavy silver stain in pachytene but are no longer detectable in later stages of meiosis.
