    Notes: Summary The present study is based on the results of planimetric measurements of temporal bone pneumatization in 60 children. Radiological and clinical follow-up studies show that a myringotomy is sufficient therapy for cases of serous effusions. In contrast, mucous effusions require long-term middle ear ventilation with tympanostomy tubes to get approximately the same increase in temporal bone pneumaticzation.

    Notes: Abstract The present study is based on the results of a planimetric investigation of temporal bone pneumatization in 43 children. Radiological and clinical follow-up studies 10 years after therapy for secretory otitis media indicate an adaptation in the size of pneumatization to an almost normal area. The increase shows that a myringotomy is sufficient in cases of serous effusion, whereas a mucous fluid requires long-term middle ear ventilation.

    Description / Table of Contents: Summary This present study is based on the results of a planimetric investigation of temporal bone pneumatization in 73 children (with and without middle ear pathology). Radiologic and clinical follow-up studies indicate a definite inhibition of pneumatization due to chronic secretory or recurrent suppurative otitis media. This process is partially reversible if the underlying pathologic ventilation of the middle ear spaces is cured by an adequate treatment, such as direct adenoidectomy and/or long-term middle ear ventilation (6 months indispensible).
    Notes: Zusammenfassung In der vorliegenden Studie wurde die Ausdehnung der Felsenbeinpneumatisation ohrgesunder und ohrkranker Kinder planimetrisch berechnet. Wir stellten dabei fest, daß bei rezidivierenden Otitiden oder Seromukotympanon mittels einer Langzeit-Paukendrainage eine deutlich stärkere Zunahme der bisher gehemmten Pneumatisation erfolgt als nach alleiniger Adenotomie. Die radiologischen und klinischen Befunde weisen darauf hin, daß rezidivierende Mittelohrinfekte in früher Kindheit zu einer gehemmten Pneumatisation führen, diese aber nach adäquater Behandlung mit Paukendrainage und/oder Adenoidektomie wiederum fortschreitet.

    Description / Table of Contents: Zusammenfassung Der Einbau14-C-markierter Nucleotide und Aminosäuren in die Nucleinsäuren und in das celluläre Protein PHA-stimulierter menschlicher Lymphocyten wurde durch steigende Adriamycin-, Bleomycin- bzw. Na2HAsO4-Dosen unterschiedlich gehemmt. Die mit Hilfe des Flüssigkeits-Szintillationszählers und teilweise durch14C-Thymidin markierte Autoradiographien gewonnenen Befunde machen deutlich, daß Adriamycin die stärkste Einbauhemmung verursacht. Im Gegensatz dazu ist die Bleomycin- und Arsenatbedingte Inhibition wesentlich geringer ausgeprägt. Durch cytophotometrische Untersuchungen konnte gezeigt werden, daß Adriamycin in der Lage ist die Zellen in der G2-Phase und in der S-Phase zu arretieren, ähnlich reagieren die Zellen auf Bleomycin und Arsenat. Die unterschiedlich ausgeprägte Inhibition des Nucleinsäure- und Proteinstoffwechsels durch Adriamycin, Bleomycin und Arsenat ist mit einer verschiedenen Ansprechbarkeit von Lymphocyten auf die cytostatisch wirksamen Substanzen verbunden.
    Notes: Summary The incorporation of14C-marked nucleotides and amino acids into the nucleic acids and into the cellular protein of PHA-stimulated human lymphocytes was inhibited variably by increasing doses of Adriamycin, Bleomycin and Na2HAsO4. The findings, which were obtained with the aid of the liquid scintillation counter and partly by means of14C-thymidine-marked autoradiographies shows clearly that Adriamycin causes the strongest incorporation inhibition. In contrast to this, the inhibition caused by Bleomycin and arsenate is considerably less destructive. It was shown by means of cytophoto metrical examinations that Adriamycin is capable of fixing cells in the G2 phase and in the S phase. Cells react similarly to Bleomycin and arsenate. The differential inhibition of nucleic acid and protein metabolism induced by Adriamycin, Bleomycin and arsenate is correlated with a varying responsiveness of lymphocytes to the cytostatically effective substances.

    Description / Table of Contents: Zusammenfassung Der Einbau von14C-Thymidin in die DNA kultivierter menschlicher Lymphocyten wird durch Fumarsäuremonoäthylester dosisabhängig gehemmt. Szintillationsmessungen der Radioaktivität und Markierungsindex in Autoradiogrammen laufen parallel und zeigen damit, daß die Zahl der DNA-synthetisierenden Zellen und nicht die von der einzelnen Zelle eingebaute Radioaktivität abnimmt. Eine spezifische Wirkung auf bestimmte Zellzyklusphasen, vor allem ein von anderen cytostatisch wirkenden Substanzen bekannter G2-Block konnte nicht beobachtet werden. Im Tierexperiment führte die intraperitoneale Gabe von durchschnittlich 6,88 mg FSME pro g Maus zum Tode der Versuchstiere. Pathologisch-anatomisch standen neben Veränderungen an Lungen, Leber und Nieren diffuse Herzmuskelzellnekrosen im Vordergrund, die auch als Todesursache angesehen werden. Bei niedrigeren Dosierungen konnten histologisch weniger ausgeprägte aber qualitativ gleichartige Befunde erhoben werden.
    Notes: Summary The incorporation of14C-Thymidin into DNA of cultured human lymphocytes is depressed by added fumaric acid monoethylester (FSME) depending on the dosage of FSME. The decreased radioactivity in DNA as measured by scintillation counting is paralleled by a concomitant decrease in the labelling index in autoradiograms. Decreasing radioactivity is therefore due to a lower number of DNA synthesizing cells. No selective inhibition of proliferation during one of the cell cycle phases was observed. Especially a G2-block known from other cytostaties is absent. A mean dosis of 6.88 mg FSME per g body weight administered intraperitoneally is lethal to mice. The animal die from diffuse necroses of heart muscle cell. Alterations of other organs are less prominent. At lower doses of FSME the morphology of the organs investigated is altered to a smaller degree.
