    Notes: The allergenic proteins of mould, Rhizopus nigricans extract (RNE) were identified and characterized by crossed immunoelectrophoresis (CIE), thin-layer isoelectrofocusing (TLIEF) and RAST inhibition. CIE revealed that the extract contained at least 31 distinct antigens. On TLIEF the extract resolved into 22 bands in pi 3.5–6.8. Two important allergens, Rhiz IIIb and VIb were purified by a combination of ammonium sulfate precipitation, anion exchange chromatography on DEAE-Sephadex column and gel filtration. Twenty and 12 μg of Rhiz IIIb and Rhiz VIb were sufficient to give 50% RAST inhibition as against 43 μg of crude RNE. Rhiz IIIb and Rhiz VIb were found to be glycoproteins with molecular weights of 12,400 daltons and 14,200 daltons, respectively. Rhiz IIIb was found to be homogeneous on polyacrylamide gel electrophoresis (PAGE) and TLIEF with a pI of 4.8, while Rhiz VIb gave a single band on PAGE and resolved into two Coomassie blue stained bands with pI 3.6 and 3.8. It was possible to separate the components of RNE on fast protein liquid chromatography (FPLC) using an anion exchanger Mono Q column. The identification and characterization of the antigenic and allergenic proteins in the extract will be useful in standardization of RNE and in preparation of an in-house reference standard.

Der gelbe Cu-PTT-Komplex wird mit Chloroform extrahiert und ist für 8 Tage beständig.λ max liegt bei 360 nm. Das Beersche Gesetz gilt zwischen 0,04 und 0,36μg Cu/ml. Der Einfluß verschiedener Fremdionen wurde untersucht. Mit Hilfe der angegebenen Methoden wurde der Kupfergehalt in Legierungen bestimmt.
    Notes: Summary 1-Phenyl-3-thiobenzoyl thiocarbamide (PTT) is used for the extraction and spectrophotometric determination of copper at trace level. The yellow coloured Cu-PTT complex is extracted in chloroform, showsλ max at 360 nm and is stable for 8 days. Beer's law is followed in the range of 0.04 to 0.36μg of copper per ml. The effect of the presence of various ions is studied. Copper in alloys has been determined using this method.

Der gelb-rot gefärbte Fe-PTT-Komplex bildet sich bei Zimmertemperatur und erreicht sein Farbmaximum innerhalb 60 min. Er läßt sich mit Chloroform extrahieren und hat ein Absorptionsmaximum bei 410 nm. Im Bereich 2–20 ng Fe/ml ist das Beersche Gesetz erfüllt. Die Empfindlichkeit der Bestimmung beträgt 0,0266 ng/cm2, die molare Absorption 2,094·106l·mol−1·cm−1. Viele Ionen stören auch in großer Menge nicht. Das Verfahren wurde zur Eisenbestimmung in Drogen, in Wasser und in Erzen verwendet.
    Notes: Summary A highly sensitive reagent, 1-phenyl-3-thiobenzoyl thiocarbamide (PTT) is used for the extraction and spectrophotometric determination of iron at trace level. The yellowish-red coloured Fe-PTT complex is formed at room temperature and maximum colour intensity is attained in 60 min. It is extractable into chloroform and shows maximum absorption at 410 nm. Beer's law is followed in the range of 2–20 ng Fe/ml. The sensitivity of determination (Sandell's) is 0.0266 ng/cm2 and molar absorptivity is 2.094×106l·mol−1·cm−1. Many common ions are tolerated to a large ratio. The method has been utilized to determine iron in drugs, water and ores.

    Description / Table of Contents: Zusammenfassung Tetramethylthiuramdisulfid (TMTD) wurde zur Extraktion und spektrophotometrischen Bestimmung von Mikrogrammengen Osmium verwendet. 15 ml einer 1,052.10−3-molaren Lösung von TMTD in Toluol dienen zur Extraktion von Os aus 7-m Salzsäure. Der entstandene rote Komplex erreicht innerhalb 2 Stunden maximale Farbintensität und zeigt zwei Absorptionsmaxima bei 445 nm und 504 nm. Letzteres ist signifikanter. Zwischen 1,7 und 26,7μg Os/ml gilt das Beersche Gesetz. Nach der Ringbom-Gleichung liegt die geeignetste Konzentration zwischen 9,42 und 23,65μg Os/ml. Nach 30 sec stellt sich das Gleichgewicht ein und bleibt bis zu 5 Stunden konstant. Die extrahierte Substanz entspricht der Zusammensetzung 1 ∶ 1 (Os ∶ TMTD). Os läßt sich auch in Gegenwart anderer Platinmetalle so extrahieren und bestimmen.
    Notes: Summary Tetramethylthiuram disulphide (TMTD) is used for the extraction and spectrophotometric determination ofμg quantities of osmium. 15 ml of 1.052×10−3 M TMTD in toluene quantitatively extracts osmium from7 M HCl. The complex attains maximum colour intensity in 2 hours. It is red in colour and exhibits two λ max. One at 445 nm and the other at 504 nm. The maximum at 504 nm is more significant. Beer's law was found to hold good over the concentration range of 1.7 to 26.7μg of osmium per ml. The Ringbom plot shows the optimum range of 9.42 to 23.65μg of osmium per ml. The optimum period of equilibration is 30 seconds and the complex is stable up to 5 hours. The extracted species shows the composition to be 1 ∶ 1 (Os ∶ TMTD). It is possible to extract and determine osmium in the presence of other platinum metals with composition corresponding to the ore osmiridium.

    Notes: Abstract Completely damaged wheat grains, unfit for human consumption, were fermented to H2 by Bacillus licheniformis strain JK1. Batch-culture fermentation of wheat slurries [6% (w/v) total solids and 5.8% (w/v) organic solids (OS)] evolved 225, 205 and 203 l of biogas-H, a mixture of H2, CO2 and H2S, per kg OS at pH 6, 7 and 8, respectively. H2 constituted 25% to 41% of the total biogas-H evolved. In single-stage continuous culture, H2 generated/kg OS reduced was 70 l at pH 6 and 74 l at pH 7 and 8.

    Notes: Abstract Viable cells of H2-producers (Bacillus licheniformis and a mixed microbial culture) were immobilized on brick dust and in calcium alginate beads. In batch culture, cells of the mixed culture in the free state yielded 8.2 l H2/mol glucose utilized, whereasB. licheniformis evolved 13.1 l H2. Immobilized cells, however, gave 4-fold more H2 than the free bacteria. Highest yields were from the cells immobilized on brick dust. High H2-production rates continued over two rounds of re-use of the immobilized cells.

Als Arbeitselektrode dient ein hängender Quecksilbertropfen. Die Anreicherung erfolgt bei −0,55 V (Bezugselektrode: Ag/AgCl/KCl ges.) aus 1 N HCl in Gegenwart von CuCl2. Das Elektrolyseprodukt ist vermutlich eine intermetallische Verbindung der Zusammensetzung Cu3As, die auf der Oberfläche des Quecksilbertropfens abgeschieden wird. Die Auflösung durch »cathodic-stripping« auf Grundlage der Weiterreduktion des Arsens zu negativ 3 wertigem Arsen führt zu einem bei −0,77 V bis −0,79 V liegenden Peak, dessen Höhe von 0,2 bis 20 ppb As linear zunimmt. Auf Störeinflüsse wird hingewiesen.
    Notes: Summary A method for the determination of traces of As(III) by differential pulse cathodic stripping voltammetry (DPCSV) is described. The hanging mercury drop electrode (HMDE) is used as working electrode. Preelectrolysis is carried out in 1 N HCl in the presence of CuCl2 at a potential of −0.55 V (reference electrode: Ag/AgCl/KCl sat.). The product formed in the electrolysis is probably an intermetallic compound of the composition Cu3As. It is deposited on the mercury surface. Its redissolution by cathodic stripping on the basis of a further reduction of arsenic to As3− leads to a current signal at a potential between −0.77 V and −0.79 V. The dependence of peak height on concentration of arsenic shows linearity in the range of 0.2 to 20 ppb As. Interfering influences are also reported.
