    Notes: In order to clarify the role of Ti addition in improving the coercivity of Nd-TM-B magnets, the microstructure was studied extensively using an analytical transmission electron microscope. Electron micrographs revealed several phases in the Ti-doped Nd15(Fe0.585Co0.3Ni0.1Ti0.015)77B8 magnet, namely Nd2(Fe0.6Co0.3Ni0.1)14B, Nd(Fe0.8−xCoxNi0.2)4B (x=0.25–0.4), Nd-TM, and TiB2 phases. Some of the TiB2 precipitates are formed in the Nd-TM phases during the solidification of the magnet alloy, and the others precipitate during the sintering process. Fine TiB2 precipitates restrain grain growth of the magnetic phase during sintering, which leads to a fine-grained structure. It is suggested that the coercivity of Ti-doped Nd-TM-B magnets increases due to the presence of TiB2 precipitates.

    Notes: Aim: To examine the efficacy and safety of human embryonic stem (ES) cell-based therapies, allogeneic transplantation of monkey ES cells would be useful. We transplanted genetically marked monkey ES cells into the allogeneic fetus. Methods and Results: Cynomolgus ES cells were transduced once using a simian immunodeficiency virus-based lentivirus vector encoding the GFP gene driven by the CMV promoter at 1, 10 and 100 transducing units per cell. Five days posttransduction, 60, 80 and 90% of the cells expressed GFP, respectively, and the expression levels were stable for 5 months. GFP expression was still observed after embryoid-body formation. The gene-marked ES cells were transplanted into the cynomolgus fetus in the abdominal cavity (n = 2) or liver (n = 1) after the first trimester. The fetuses were delivered 1 month posttransplantation. Transplanted cell progeny were detected (∼1%) in multiple tissues by quantitative PCR and in situ PCR of the GFP sequence. No teratoma was found in the tissues. Conclusions: Cynomolgus ES cells can be engrafted in the allogeneic fetus. We are now trying to transplant cynomolgus ES cells differentiated to neural or hematopoietic lineage.

    Notes: A high-speed rotor which is characterized by a mechanical precession eliminator and by a magnetic suspension has been constructed. In this instrument, a cylindrical rotor 2 cm (0.02 m) in diameter and 20 cm (0.2 m) long was made of aluminum. The moment of inertia of this rotor was approximately 86 g cm2 (8.6×10−6 kg m2) about the long axis. A stable rotation of 120 000 rpm was obtained in air as well as in vacuum. The rotor was suspended vertically by a set of suspension magnets at the top of the rotor and a set at the bottom. In addition, the bottom of the rotor is flexibly supported by a ruby bearing which moves smoothly on a plate but is constrained by friction. When precession of the rotor begins, the frictional force eliminates the precession intrinsically. Measurements of slowing rate for the revolution were made at several different air pressures. There is good agreement with the theory.

    Notes: The fabric of high-Tc Nb3Ge superconducting material synthesized by coevaporation in the vacuum has been investigated using 200-kV high-resolution electron microscopy. The grain texture deduced from the high-resolution structure images, which consist of groups of small coaxial Nb3Ge grains (A15 structure) which have one cubic axis [001] in common, is observed and analyzed using the high-resolution structure images. The coaxial grains are related by a rotation of approximately ±30° around the [001] axis. Crystallites of Nb5Ge3 composition having either a D88 structure or D8m structure are distributed at random in the fabric and are coherent with the adjacent Nb3Ge grains. The statistical distribution of grain sizes of each compound is reported. The average grain size of Nb5Ge3 compounds is smaller than half the average size of the Nb3Ge grains.

    Notes: Abstract It has recently become clear that cyclin-dependent kinase (cdk) complex regulates the cell cycle by phosphorylating Rb protein, a tumor suppressor protein. It is likely that this complex is a target of various growth factors and anti-growth factors (UV TGF-β etc.) in keratinocyte (KC). It has also been suggested that abnormalities in the cell cycle regulating mechanism such as increased activity of cyclin-cdk due to mutation of p53, a tumor suppressor gene, and overexpression of cyclin D may be concerned with carcinogenesis of KC. Thus, recent studies indicate that the cyclin-cdk complex is a common target of proliferation and carcinogenesis in KC.

    Notes: Summary Wild-type p53 accumulation induced by DNA damaging agents such as ultraviolet (UV) radiation. γ-irradiation and drugs, may arrest the cell cycle until DNA damage is repaired. p21Waf1/Cip1 is a cyclin-dependent kinase (CDK) inhibitor induced by wild-type p53. CDK is activated by cyclin and progresses the cell cycle. On the other hand. CDK inhibitors inhibit CDK activity to arrest the cell cycle. Thus, p21Waf1/Cip1 is thought to mediate the signal of p53 induced by DNA damaging agents to arrest the cell cycle. p21Waf1/Cip1 is induced by wild-type, but not mutant p53. To investigate p21Waf1/Cip1 regulation by p53 in epidermis in vivo, immunohistochemical staining of p21Waf1/Cip1 and p53 were conducted in chronically sun-exposed normal epidermis and in neoplastic epidermis. p21Waf1/Cip1 expression was found to be coincident with the p53-positive regions or not coincident with the p53-positive regions in chronically sun-exposed normal epidermis, whereas there was only low or undetectable p21Waf1/Cip1 expression in any regions including the p53-positive regions of solar keratosis and squamous cell carcinoma of the skin. This suggests that wild-type p53 and p21Waf1/Cip1 may play a part in chronically sun-exposed normal epidermis response to UV exposure, whereas p21Waf1/Cip1 cannot be induced by mutated p53 in solar keratosis and squamous cell carcinoma of the skin.

    Notes: We report a case with rapidly growing subcutaneous and intra-muscular tumours that were identical histopathologically to a pre-existing left atrial myxoma. No invasion of visceral organs or skeleton was found. This is a very rare case of metastasizing myxoma with wide dissemination and rapid metastastic growth, despite the benign appearance of the original tumour.

    Notes: The distribution of cytoplasmic microtubules in cultured guinea-pig keratinocytes was investigated using immunofluorescence (IF) microscopy with monospecific anti-tubulin antibodies and electron microscopy (EM). In culture, adherent cells displayed networks of thin fluorescent fibres, while a homogeneous and/or granular cytoplasmic IF was shown in the cells of upper layers as well as in trypsinized cells. By EM many microtubules were shown in adherent cells but there were fewer or none in the upper layers. An increase in calcium ion (Ca2+) concentration and the addition of an ionophore (X537A) to the culture medium caused disassembly of microtubules. This effect was cancelled by a calmodulin inhibitor. Cryostat sections of normal human and guinea-pig epidermis stained with anti-tubulin antibodies showed a homogeneous and/or granular cytoplasmic IF from basal to granular layers but no detectable IF was seen in the horny layer. These results suggest that keratinocytes contain a cellular pool of tubulin in various states of polymerization and that microtubule disassembly may occur during differentiation, probably being regulated by Ca2+-calmodulin complexes.

    Notes: Dispase is a bacterial neutral protease which is obtained from the culture filtrate of Bacillus polymyxa, After 24-h treatment of the human skin with 500 and 1000 U/ml dispase, the epidermal sheet was easily peeled from the dermis, and its undersurface retained rete ridges. Electron microscopic observation showed that the basal surface was composed of cells with numerous slender villi and cytoplasmic projections. Although the intercellular spaces of the spinous as well as the basal layers were wide, all desmosomes were intact with their accompanying tonofilaments. An epidermal cell suspension was obtained by incubating the dispase-separated epidermal sheet in trypsin solution for 15 min and the viability of the cells was more than 95% as measured by the trypan blue dye exclusion test. A culture of keratinocytes was established by seeding the dissociated cells in the usual way.
