    Notes: Abstract 99mTc-organocation complexes were screened for heart affinity using the isolated perfused rat heart. The 99mTc-I compounds are better extracted by the heart and retained in it than the 99mTc-III compounds. 99mTcCl2(DMPE) 2 + shows low uptake (15%) and fast washout, but no impairment of uptake after incubation with human serum albumin(HSA), with a modified preparation, initial uptake is not much better (18%) but washout is clearly diminished. 99mTc(DMPE) 3 + is taken up to a greater extent (33%) if administered solely, but when given in conjunction with HSA, uptake decreases to about 4% because of strong affinity of the complex to the protein, but this compound persists well in the tissue. Detergents like tween 80 and dioctylsulphosuccinate are able to cause higher uptake of 99mTc(DMPE) 3 + even in the presence of HSA, thus indicating a diminished binding of the compound to the protein. 99mTc(CN-t-butyl) 6 + shows very high uptake (80%) by the heart, which is only moderately diminished by addition of HSA (t0 38%), and persists excellently in the myocardium. Basic features of the compounds such as accumulation in and elimination from the myocardium as well as plasma protein binding and their mutual relationships are well reflected by the isolated heart model.

    Notes: Summary Experimental results obtained with the neutral sucrose gradient sedimentation techniques of analysing mammalian DNA after irradiation in vivo (Chinese hamster cells V79) are evaluated theoretically in order to verify and extend a model by Ormerod and Lehmann that describes the gradual release of free DNA from the DNA-membrane complex. The model is based on the idea of chromatin organization in the form of membrane attached superstructure units (MASSUs) defined by consecutive attachment points in intervals ofM 0 (DNA molecular weight of a MASSU). DNA sedimentation after cell lysis with sarkosyl as detergent allows good separation of the released free DNA from that remaining in the complex. The dose-dependence of both the percentage of DNA released and pertinent molecular weight parameters as measured with X-rays and derived from the model confirms it and yields the MASSU sizeM 0 = 2.4 × 109 g/mol. Furthermore, the model is evaluated with respect to high-LET radiations by a mathematical theory based strictly on microdosimetry. This approach, which could be of general interest as a contribution to theoretical microdosimetry, reduces the measurable quantities explicitly to the single event distribution of lineal energy for the gross volume,V, occupied by a MASSU in the interphase nucleus. Experimental results obtained withα-particles agree with this extended version of the model, yielding 0.2 – 0.3 µm for the diameter ofV. In summary, the following conclusions are drawn: The entire chromatin, at least the greater part of it, must be organized in regular superstructure units with no detectable variance of their lengthsM 0, which is suggested to be a general principle whose functional implications remain to be elucidated.

Zur Bestimmung der Wirksamkeit alkoholischer Händedesinfektionsmittel gegenüber 3 MRSA-Stämmen im Vergleich zu 3 Methicillin-empfindlichen Staphylococcus aureus -Stämmen (MSSA) wurden 1-Propanol (60 %) sowie Sterillium® und Spitaderm® in verschiedenen Verdünnungen und bei verschiedenen Einwirkzeiten (15 s, 30 s und 60 s) im quantitativen Suspensionsversuch untersucht. In der Anwendungskonzentration war nach 30 s für alle Präparate eine Keimreduktion von mehr als 6 log10-Stufen gegenüber MRSA und MSSA nachweisbar. In Verdünnung (50 %) und bei kürzerer Einwirkzeit (15 s) wiesen alle Präparate signifikant niedrigere Reduktionsfaktoren gegenüber MRSA auf (p 〈 0,05). Verdünntes Sterillium® (50 %) erreichte weder gegenüber MRSA noch gegenüber MSSA nach 30 s eine Keimreduktion von mehr als 5 log10-Stufen. Die Bedeutung einer sachgerechten Durchführung der Händedesinfektion zur Unterbrechung von Infektketten mit MRSA wird deutlich.
    Notes: Summary. In order to determine the efficacy of hand disinfectants based on alcohol against three MRSA strains and 3 methicillin-susceptible S. aureus strains (MSSA), 1-propanol (60 %) as well as Sterillium and Spitaderm were investigated in the quantitative suspension test at various dilutions and reactions times (15, 30 and 60 s). All undiluted disinfectants revealed reduction factors 〉 6 against MRSA and MSSA after 30 s. Diluted disinfectants (50 %) were significantly less effective against MRSA at short reaction times (15 s) (p 〈 0.05). Sterillium in a dilution of 50 % did not reach 5 reduction factors against either MRSA or MSSA after 30 s. The impact of an appropriate use of hand disinfectants in order to break chains of infections with MRSA is obvious.

    Notes: Abstract Screening methods to identify methicillin-resistantStaphylococcus aureus (MRSA) were compared using 96 isolates representing 17 distinct clones. The sensitivity of four commercial agglutination tests was determined in comparison to the tube coagulation test, and the results related to the presence of the coagulase gene. The broth screening test, agar dilution test and disc diffusion test were carried out, and the results related to the presence of themecA gene. Mannitol salt agar and Iso-Sensitest agar with varying salt supplements were used. All agglutination tests had high rates of detection ofStaphylococcus aureus (95.8–99.0%). Resistance in mecA gene-positiveStaphylococcus aureus isolates was correctly detected by the oxacillin broth test, the agar dilution test and the disc diffusion test on mannitol salt agar, whereas on Iso-Sensitest agar detection rates were lower (between 68.5% and 94.4%, depending on the salt supplement). Incubation of the Iso-Sensitest plates for 48 hours significantly improved the rate of detection of resistance, but increased the major error rate up to 71.4%.MecA genepositiveStaphylococcus aureus isolates not detected by the disc diffusion test on Iso-Sensitest agar had significantly lower oxacillin minimal inhibitory concentration values and were significantly less resistant to a variety of antibiotics. Thus, mannitol salt agar might be a suitable medium for use in the disc diffusion and agar dilution test to detect resistance to oxacillin inStaphylococcus aureus.

    Notes: Abstract Zeolites ZnY with various overall zinc contents were prepared from mixtures of zeolite NH4Y and crystalline zinc chloride by solid-state ion exchange. The obtained materials were investigated with carbon monoxide, xenon, and nitrogen adsorption as well as with 129Xe NMR and XRF spectroscopy. From the results of these measurements, the zinc cation distributions between the different types of cages of the faujasite framework as well as between the crystallographic positions SIII and SII within the large voids (supercages) were quantitatively determined. The concentrations of zinc cations in the supercages of the presently prepared zeolites are considerably higher than in materials obtained from NaY by conventional wet ion exchange using aqueous zinc salt solutions. Experimental evidence is provided for salt inclusion under certain conditions of preparation.

    Notes: Abstract Trivalent radiometal tracers have been used for tumour imaging and metastatic pain palliation. For better understanding their tumour accumulation, basic model studies of uptake of different169Yb complexes into cultured normal and tumour cells were performed. Whereas the uptake of169Yb citrate is strongly dependent on the metabolic activity and is not tumour-cell specific, the uptake of169Yb complexed with aminocarbonic acids (NTA, EDTA, DTPA) does not correlate to the metabolic activities. These complexes are taken up to a greater amount by the tumour cells (by a factor of about 2). Uptake of both complex types leads to a stable association to cellular compounds,169Yb is not releasable by the strong complexing agent DTPA. Protein binding of the169Yb complexes shows great influence on their cellular uptake. The bound proportion is no more available for cellular uptake. The results indicate that i) uptake of169Yb citrate is an active cellular transport process which is not tumour-specific, ii) the169Yb aminocarbonic acid complexes show a weak favouring by the tumour cells, iii) different from earlier acceptions the Yb complexes studied are not taken up by the cells in protein-bound form. The structure of the Yb complex is decisive for its protein binding and cellular uptake.
