    Notes: Summary Previously, surgical reconstruction of the transected dog spinal cord by the delayed nerve graft technique has been shown to result in reinnervation of the nerve graft by axons. In the present study, we compared the results of surgical reconstruction of the severely contused cat spinal cord by the delayed nerve graft technique alone to those after reconstruction with a similar nerve graft plus cultured peripheral nonneuronal cells implanted between the grafted nerve and the spinal cord stumps. The spinal cord-nerve graft junction was examined by light and electron microscopy. The cultured cells were prelabelled with tritiated thymidine and their location after implantation determined by autoradiography. By 3 days after spinal cord reconstruction, the pelabelled cells were present at the junction and had migrated into the nerve graft and also into the spinal cord stumps where they were observed near axons. By 7 days, physical connections were observed bridging the junction between the spinal cord and nerve graft and axons ensheathed by Schwann cells had already penetrated at least 1 mm into the nerve graft. Wound healing took at least a week longer in animals repaired with a nerve graft alone. At one year or later after reconstructive surgery, in both groups of animals, the grafted nerve was reinnervated with myelinated and unmyelinated axons. Thus, the severely contused cat spinal cord could be reconstructed with the delayed nerve graft technique alone but the use of the cultured cells appeared to enhance wound healing and decrease the time required for axon elongation into the nerve graft.

    Notes: Summary We have previously reported that cultured peripheral non-neuronal cells could be used as an adjunct to spinal cord reconstruction with the delayed nerve graft technique. The cultured cells appeared to enhance axonal regeneration and with their use the time it took for axons from the spinal cord stumps to reach the nerve graft was reduced. To gain insight into the possible mechanisms through which peripheral nonneuronal cells can foster CNS regeneration, we have now investigated the behaviour of the peripheral nonneuronal cells after implantation into the spinal cord. Autologous mixed non-neuronal cell cultures were prepared from cat sciatic nerve biopsies and labeled in culture with tritiated thymidine. The labeled cells were implanted so as to completely fill the gap in the spinal cord produced by a narrow “slit transection”. Light-and electron-microscopic autoradiography was used to identify the cells 3 and 7 days after implantation and to determine their proximity to, and possible interaction with, axons in the spinal cord stumps. The implanted peripheral cells were frequently found near spinal cord axons and axon terminals. Some of the labeled cells ensheathed axons in which case they displayed morphological characteristics of Schwann cells. Other labeled cells had characteristics of fibroblasts and were surrounded by an extracellular matrix rich in collagen fibrils. Many of the labeled cells contained phagocytosed myelin debris. These observations are consistent with the implanted cells acting to enhance regeneration in the spinal cord either by direct interaction with axons (ensheathment) or indirectly via the production of soluble neuronotrophic factors or a favorable extracellular matrix. The ability of the implanted cells to rapidly move into the spinal cord stumps and attain positions close to spinal cord axons would be an important factor for any of these mechanisms.

    Notes: Polarization dependent extended x-ray absorption fine structure (PD-EXAFS) and magnetic circular dichroism (MCD) measurements of CoCrTa and CoCrPt films, sputter deposited at varying substrate temperatures, were performed to investigate the average local structure and chemistry about the Ta, Pt, and Co atoms and the average magnetic moment of the Co and Cr atoms within these films. Results from the MCD measurements indicate the average net magnetic moment of the Cr atoms is opposite in direction and five percent in amplitude relative to the Co moments. Inspection of the Fourier transforms of the XAFS data from these samples shows an increase in structural disorder around the Ta and Pt atoms with increasing substrate deposition temperature. A further comparison between the Ta and Pt edge EXAFS results show that the temperature-dependent increase in structural disorder is greater around the Ta atoms in the CoCrTa system than it is around the Pt atoms in the CoCrPt system. © 1996 American Institute of Physics.

    Notes: Spin resolved resonant Raman scattering measurements in Gd metal made by exciting x-ray resonant Raman scattering with circularly polarized x rays near the LIII edge of Gd are presented. The incident photon energy was fixed at the peak of the 2p→4f quadrupolar transition, and the scattered photon was energy analyzed around the 3d→2p fluorescent energy. Asymmetry ratios in the scattered intensity much larger than that of the LIII XMCD effect (x-ray magnetic circular dichroism) were observed upon reversal of the magnetization of the sample or the helicity of the photon. A detailed comparison of these results with XMCD results from both the MIV,V edges and LIII edge of Gd metal will be discussed. © 1996 American Institute of Physics.

    Notes: The reflectivity of circularly polarized (CP) soft x rays is a technique which combines the power of x-ray scattering and magnetic circular dichroism and allows the structural and magnetic characterization of heteromagnetic multilayers. In this technique, the energy and angular dependence of the differential reflectivity of left- and right-circularly polarized soft x rays at the relevant absorption edges of the constituent elements of the multilayers are measured. As examples the reflectivity curves for single films of Fe, Co, ordered and disordered Fe50Co50 alloys, and for a trilayer consisting of two single crystal Fe30Co70 films separated by an 8.7 A(ring) Mn spacer layer media utilizing complex dielectric tensors with no free parameters show that the rich reflectivity curves can be used to determine the multilayer magnetic ordering, film thicknesses, interface quality, as well as differentiating between ordered and disordered alloys. © 1996 American Institute of Physics.

    Notes: Recently, there has been a great deal of interest in x-ray magnetic circular dichroism (MCD) and its applications to the study of magnetic thin films and multilayers due to the dramatically enhanced dichroic effect and its element specific nature. In this work, the soft x-ray magneto-optical Kerr effect, an effect closely related to MCD, is investigated. Detailed comparison between MCD and the Kerr effect is discussed. As an example, recent results from a thin bcc Co film will be presented. In the Kerr effect measurement, specular reflectivity as a function of incident angle and photon energy was measured using both linearly polarized and circularly polarized x rays. To give the largest effect, in the case of linearly polarized light, specular reflectivity was measured in the transverse configuration, while in the case of circularly polarized light, the measurement was carried out in the longitudinal configuration. In both cases very large changes in reflectivity, up to 40% in the linearly polarized case and 75% in the circularly polarized case, were observed near the Co LII and LIII absorption edges upon reversal of the direction of the magnetic field. These results agree very well with a macroscopic model calculation.

    Notes: The tunability and the polarization of synchrotron radiation open up new possibilities for the study of magnetism. Studies on magnetic materials performed at the National Synchrotron Light Source are reviewed, and they fall into four areas: structure, evolution of magnetic order, separation of L and S, and resonance effects. In the vicinity of atomic absorption edges, the Faraday effect, magnetic circular dichroism, and resonant magnetic scattering are all related resonance effects which measure the spin-polarized density of states. The production and analysis of polarized beams are discussed in the context of the study of magnetism with synchrotron radiation.

    Notes: It is very difficult to characterize the polarization of a synchrotron radiation source in the soft and/or intermediate x-ray energy region particularly from 1 to 2 keV. Conventional multilayer mirror or single-crystal polarimeters do not work over this energy region because their throughput (the reflectivities combined with the phase shift) becomes insignificant. In this paper, we present a new ellipsometer scheme that is able to fully characterize the polarization of synchrotron radiation sources in this energy region. It is based on the dichroic x-ray resonant ferromagnetic scattering that yields information on both the polarization of the x-ray and the material (element specific) dielectric-constant tensor [C.-C. Kao et al., Phys. Rev. B 50, 9599 (1994)] due to the interband ferromagnetic Kerr effect [B.R. Cooper, Phys. Rev. A 139, 1504 (1965)]. © 1996 American Institute of Physics.
