A number of potential 14C- and 3H-labelled precursors were fed to growing cultures of Phoma spec. (Strain S 298), after the rate of formation of phomine (cytochalasin B; C29H37NO5) had been determined. By chemical degradation of [14C]-phomine and [3H, 14C]-phomine the distribution of the radioactivity originating from the incorporated precursors was determined. The results demonstrate that the building blocks of phomine are 1 unit of phenylalanine, 9 units of acetate or malonate respectively, and 2 units of methionine. Acetate acts as a starter and malonate as a propagator of a C16- or C18-chain. A possible combination of these elements leading to phomine and to 5-dehydrophomine is suggested.
