    Notes: Functional dendritic cells (DC) are professional antigen-presenting cells (APC) and can be generated in vitro from healthy as well as from leukaemic cells from AML patients giving rise to APC of leukaemic origin presenting leukaemic antigens. In a comparative methodological analysis of 50 AML samples, we could already show that leukaemia-derived DC can regularly be generated under serum-free culture conditions. In this study, we describe the generation and characterization of DC from different mononuclear cell (MNC) fractions from 24 myelodysplastic syndrome (MDS) patients under those different serum-free culture conditions, determine the optimal culture conditions and compare the results with that from 23 healthy donors. In parallel cultures, we compared DC harvests after 7- or 14-day culture, with total or adherent MNC or T-cell-depleted MNC or PB or BM–MNC, thawn or fresh MNC, in Xvivo or CellGro serum-free media, ±10% autologous plasma or ±FL. In detail, we could show that MDS–DC harvests compared to healthy DC were higher after 10- to 14-day culture; total or adherent PB or BM–MNC fractions yield comparable DC counts; however, from MACS-depleted MNC fractions or thawn MNC lower DC counts can be generated. Whereas the addition of FL increases the DC harvest, the addition of autologous plasma in many cases has inhibitory influence on DC maturation, CellGro and Xvivo media yield comparable DC counts. Optimal harvest of vital and mature DC from MDS samples was obtained with a GM-CSF, IL-4, FL and TNF-α containing serum-free Xvivo medium after 10–14 days of culture (18/26% DC; 54/64% vital DC; 59/51% mature DC were generated from MDS/healthy MNC samples). Surface marker profiles (e.g. costimulatory antigen expression) of DC obtained from MDS samples were comparable with that of healthy DC. The leukaemic derivation of MDS–DC was demonstrated by the persistence of the clonal cytogenetic aberration in the DC or by coexpression of leukaemic antigens on DC. Autologous T-cell activation of leukaemia-derived DC was demonstrated in cases with MDS. Autologous T cells proliferate and upregulate DC-contact-relevant antigens. We are the first who demonstrate that the generation of leukaemia-derived DC is feasible not only in AML but also in MDS under serum-free culture conditions giving rise to DC with comparable characteristics as healthy DC and offering an antileukaemia-directed immunotherapeutical vaccination strategy in AML and MDS.

Optimal harvest of vital and mature DC from AML samples was obtained with a granulocyte/macrophage-colony stimulating factor, interleukin-4, FL and tumour necrosis factor-α-containing serum-free Xvivo medium after 10–14 days of culture (36/26% DC; 38/64% vital DC; 46/51% mature DC were generated from AML/healthy MNC samples). Surface marker profiles (e.g. costimulatory antigen expressing) of DC obtained from AML samples were comparable with that of healthy DC. The leukaemic derivation of AML–DC was demonstrated by the persistence of the clonal cytogenetic aberration in the DC or by coexpression of leukaemic antigens on DC. Autologous T-cell activation of leukaemia-derived DC was demonstrated in cases with AML. Autologous T cells proliferate and upregulate DC-contact-relevant antigens.     Notes: Abstract. Following the commissioning of the PRISMA large-acceptance spectrometer, installed at the Laboratori Nazionali di Legnaro (LNL), an international nuclear-structure collaboration has started to develop a large $\gamma$ -ray setup to be installed in the target position of the spectrometer. The array is based on the EUROBALL composite CLOVER detectors. In this contribution the CLOVER detector array is described and its expected performance figures discussed. This new setup, by using the high-intensity heavy-ion beams provided by the LNL ALPI linac, will push the study of nuclear structure towards moderately neutron-rich nuclei by means of quasi-elastic and deep inelastic reactions.

    Notes: Abstract. The highly sensitive GASP array at the INFN Legnaro Laboratory was used to study the $\gamma$ -ray de-excitation of neutron-rich nuclei produced in the deep-inelastic processes which occur when 230 MeV 36S ions interact with a target of 176Yb. Yrast decay schemes were identified in over forty target-like fragments and in over twenty projectile-like fragments. Analysis of the data has resulted in extensions to the yrast decay sequences of the target-like species, 176Hf, 166Er, 172Yb, and 152Sm. New transitions have also been observed in a number of projectile-like species including 34P and 41Cl. Experimental results are compared with the results of shell model calculations.

    Notes: Abstract. The neutron transfer in a very heavy asymmetric nuclear system, in 206 Pb + 118 Sn has been measured using particle- $\gamma$ coincidence techniques with position-sensitive detectors, 5 EUROBALL-Cluster detectors (EB) and the Heidelberg-Darmstadt NaI-CRYSTAL BALL (CB). The fragments are identified via the known $\gamma$ -decays of the lowest excited states using the high resolution of EB. Using the unique feature of the set-up with the CB, transfer to well-defined final channels with known quantum numbers is selected using the high-efficiency multiplicity filter of the CB with no second $\gamma$ -ray, i.e. without feeding. The enhancement in the two-neutron transfer is deduced, for population of the low-lying super-fluid 2 + states in 120 Sn and 116 Sn, while the 2n-transition remains in the ground state for the recoiling 20X Pb-nuclei. Large enhancements up to EF $\simeq 10^3$ are observed. This is the first observation of neutron pair-transfer enhancement for a heavy nuclear binary system with superfluid properties with experimentally separated levels.

    Notes: Abstract We report on a measurement of the lifetime of the 1414 keV state in232Th by the Doppler-shift recoil-distance method. This 4+ state shows all characteristics of a nearly-harmonic two-phononγ-vibrational excitation. The result for the B(E2) value obtained from the lifetime,τ=3.2 ± 0.7 ps, is in agreement with an earlier estimate of the collectivity from Coulomb-excitation yields and supports the interpretation of the 1414 keV state as a two-phononγ-vibrational excitation.

    Notes: Abstract: One and two neutron transfer has been measured in the heaviest asymmetric nuclear system with semi magic nuclei showing superfluid properties, in 206Pb+118Sn collisions at an energy well below the Coulomb barrier with scattering orbits covering the largest angles. Particle-γ coincidence techniques using 5 Euroball-Cluster detectors (EB) combined in a set-up with the Heidelberg-Darmstadt NaI-Crystal Ball (CB) have been used. Transfer channels are identified with EB via their known γ-decays of the lowest excited states. Using the unique feature of the set-up with the CB, transfer to well defined final states with known quantum numbers (without feeding) are selected using the high efficiency multiplicity filter of the CB (no second γ-ray). The data are analysed using the semiclassical approach and transfer probabilities are obtained. The enhancement for the two-neutron transfer populating the low lying superfluid 2+ state in 120Sn (and 116Sn), while the Pb-branch is in the groundstate is deduced by comparison with the strongest single neutron transfer transition. Large enhancements (EF ≃ 103) are observed. This is the first direct measurement of enhancement for a heavy nuclear binary system with experimentally separated levels suggesting a strong contribution from superfluid pair transfer.
