    Notes: Induced spawning in perch, Perca fluviatilis L., was studied using follicle stimulating hormone and lutenizing hormone (FSH + LH) with the addition of pimozide or metoclopramide. The doses administered to fish were: 75 IU kg−1 of FSH + LH with 5 or 10mgkg−1 of pimozide (P) or metoclopramide (M) in females, and 25 IU kg−1 of FSH + LH with 2.5 or 5 mg kg−1 of P or M in males. Hormonal injections in males (except the injection of FSH + LH with 10 mg kg−1 of M) did not influence the quantity of milt obtained. All females treated in this experiment had oocytes at the same stage of nucleus migration, so the time of ovulation was very synchronous, i.e. 16-24 h after injection. The relationship between the quality of the eggs, expressed as a survival to eyed-egg stage, and latency was inverse. Spawning success, expressed as a spawning effectiveness coefficient (Se), was highest in the fish that had been treated with FSH + LH with a higher dose of M.

    Notes: Induced spawning in bream, Abramis brama (L), was studied using acetone-dried common carp pituitary (CP) and bream pituitary (BP) with or without the addition of human chorionic gonadotrophin (hCG). The total dose administered to fish was of 5.0 mg kg−1 BP or 4.0 mg kg−1 CP with or without 2000-2200 IU hCG kg−1 for females and 2.5 mg kg−1 BP or 2.0 mg kg−1 CP with or without 1000–1100 IU HCG kg−1 for males.In all male treated groups 100% of spermiation was observed: in females the most effective method was a triple injection with hCG and carp pituitary, resulting in 79% of females ovulated (over 68% of eyed eggs). Biological quality of eggs, expressed as a percentage of eyed eggs, was negatively correlated with time elapsing between resolving (final) injection and ovulation. Spawning success, expressed as a value of Se (spawning effectiveness coefficient), was higher in fish treated with triple injection.

    Notes: Induced spawning in Eurasian perch, Perca fluviatilis L., was studied using human chorionic gonadotrophs (hCG) or acetone-dried common carp pituitary with or without hCG. The dose administered to fish was 5700 IU hCG kg-1 or 4.0 mg kg-1 carp pituitary with or without 500-700 IU hCG kg-1 for females and 2850 IU hCG kg-1 or 2.0 mg kg-1 carp pituitary with or without 250-350 IU hCG kg-1 for males. There were no statistically significant differences in quantity of milt in treated and control groups, although the best result was observed when males were treated with a triple injection of hCG and carp pituitary extract. Male spermiating success, expressed as a quantity of milt, was negatively correlated with fish weight. All females treated in this experiment had oocytes at the same division, so time of ovulation was highly synchronous. Spawning success, expressed as a spawning effectivity coefficient (Sc), was highest in fish treated with the triple injection. Spawning methods described in this paper were successful, even though hormones from another order of fish were used.

    Notes: Concentration and motility of spermatozoa, total protein content and its electrophoretic profile, glucose content, activity of aspartate aminotransferase (AspAT) and acid phosphatase (AcP) were assessed in 18 samples of semen from common bream Abramis brama L. males, which were hormonally stimulated to spermiation. Also, milt pooled from four donors was cryopreserved as pellets in vapours of liquid nitrogen (−80 °C) using four extenders (each with or without the addition of hen egg yolk). Mean spermatozoa concentration was 11.68 × 109 mL−1, and mean spermatozoa motility was about 60%. Protein content in seminal plasma was 2.08 mg mL−1; both PAGE and SDS–PAGE showed considerable heterogeneity of protein fractions. Mean glucose content was over 11 mg%. AspAT and AcP activities were detected in both seminal plasma and spermatozoa extracts. As calculated to 1 × 109 spermatozoa, AcP and AspAT activities were almost sixfold and 46-fold higher in spermatozoa than in seminal plasma respectively. In the best variant, cryopreservation attempts resulted in 66.6% of eyed embryos (compared with control fertilization) obtained after fertilization of eggs with cryopreserved semen.

    Notes: Seven published extenders, three thawing media and two thawing temperatures were tested in order to determine their suitability for cryopreservation of northern pike (Esox lucius L.) sperm. Sperm motility during successive steps of cryopreservation was evaluated. Erdahl and Graham's extender with the addition of egg yolk proved to be the most efficient (maximum hatching rate of 74.5%) when semen was thawed in 120 mm NaCl solution warmed up to 30° C. No correlations between motility of sperm (diluted in extenders or diluted in extenders and then activated with thawing solution) and the subsequent hatching success were observed. The relationship between motility of thawed sperm and its fertilization ability was considerable, but correlation was not significant. Spermatozoa frozen in some extenders were frequently motile after thawing but they were not able to fertilize the eggs, this resulted in a poor hatching rate. Depending on the extender, the addition of yolk induced either positive or detrimental effects on fertilization success.

    Notes: Anti-proteinase activity has been found in seminal plasma of eight teleost fish species: brown trout, rainbow trout, brook trout, lake whitefish, bream, northern pike, Danube salmon and burbot. This activity correlated with seminal plasma protein and sperm concentrations. Using a mammalian (bovine) trypsin for detecting proteinase inhibitors it was found for the first time that there are species-specific electrophoretic profiles of anti-proteinase activity. One to three bands could be identified by this method. However, additional proteinase inhibitors could be identified by using fish (cod) trypsin. These inhibitors were detected in seminal plasma of salmonids and coregonids and have a slow migration rate. Fast-migrating proteinase inhibitors were present in rainbow, brown and brook trout, northern pike, whitefish and burbot. These inhibitors could be detected in brook and brown trout by using either trypsins. However, they were detected only with bovine trypsin in rainbow trout, northern pike, whitefish and burbot. These results suggest that multiple forms of serine proteinase inhibitors exist in seminal plasma of teleost fish and they differ in their affinity toward serine proteinases. Seminal plasma serine proteinase inhibitors of rainbow trout migrated during electrophoresis similarly to blood plasma proteinase inhibitors, and suggests that the two inhibitors may be similar or the same. Anti-proteinase specific activity was similar in blood and seminal plasma. Proteinase inhibitors of fish seminal plasma seem to be an important part of sperm physiology, possibly related to protection of spermatozoa. Staining for detection of serine proteinase inhibitors also allowed detection of presence of nonspecific esterase in seminal plasma of most species.

    Notes: Using gelatin-SDS-PAGE, proteolytic activity was found in the seminal plasma of 10 teleosts: common carp Cyprinus carpio, bream Abramis brama, ide Leuciscus idus, chub Leuciscus cephalus, rainbow trout Oncorhynchus mykiss, grayling Thymallus thymallus, perch Perca fluviatilis, pike Esox lucius, goldfish Carassius carassius and pikeperch Stizostendion lucioperca. This activity was also measured, using azoalbumin as a substrate, in the seminal plasma of these species, with exception of pikeperch and goldfish. When azoalbumin-hydrolysing activity was expressed per volume, it was highest in common carp. Otherwise, as expressed per g of protein, the activity was highest in pike. The lowest proteolytic activity (expressed per g and volume) was observed in perch seminal plasma. Using gelatin containing polyacrylamide gels for detecting gelatinolytic activity, species-specific electrophoretic profiles were found. For all cyprinids two similar bands with a molecular mass of 68 and 74 kDa were found. The seminal plasma of grayling and rainbow trout showed similarities in the 41 kDa band. Perch and pikeperch had one similar main band with a molecular mass of 61 kDa. Proteolytic enzymes of seminal plasma from pike showed high individual variability. These results suggest that multiple forms of proteolytic enzymes exist in seminal plasma of teleosts and they differ among fish families and species.
