    Notes: Abstract Purpose: To assess the effectiveness and safety of self-expanding metallic stents as a primary palliative treatment for inoperable malignant colorectal strictures. Methods: Under radiological guidance 20 self-expanding metallic Wallstents were implanted in 16 consecutive patients with colorectal stenoses caused by malignant neoplasms, when surgical treatment of the condition had been ruled out. The patients were followed up clinically for 1–44 months, until death or termination of this study. Results: The stents were successfully implanted in all cases and resolved the clinical obstruction in all the patients except one, who underwent subsequent colostomy. During follow-up of the remaining 15 patients, clinical complications arising from the procedure were pain (two patients), minor rectal bleeding (one patient), and severe rectal bleeding (one patient) (26%). There were three cases of stent migration and three cases of stent occlusion, and reintervention by us was necessary in 20% of cases (3/15). The mean life span following the procedure was 130 days, and none of the patients exhibited clinical symptoms of obstruction at the time of death (12 patients) or termination of the study (3 patients). Conclusion: Deployment of metallic stents under radiologic guidance is an effective alternative as a primary palliative measure in malignant colorectal obstruction, though the possible clinical complications and need for repeat intervention during follow-up should be taken into account.

    Notes: Abstract To further elucidate the role of the constitutive heat shock protein-70 (HSC70) as a chaperone for the synthesis of myelin basic protein (MBP), HSC70 content was decreased in oligodendrocyte precursor cells prior to MBP expression either by transfection with an antisense oligonucleotide specific for HSC70, or by exposure to low levels of quercetin, a bioflavonoid known to decrease synthesis of HSC70. As these cells underwent differentiation in vitro, antisense treatment decreased HSC70 levels to 66% of controls. At the same time, a sharp induction resulted in the stress-inducible heat shock protein-70 (HSP70). Levels of two other stress proteins increased as well, namely, the 25-kDa heat shock protein (HSP25) and the 78-kDa glucose regulated protein (GRP78). MBP synthesis proceeded over a normal time course, but at only 50% of control values. As HSC70 content returned to normal, MBP synthesis was also restored to normal levels. Quercetin reduced the expression of HSC70 to an even greater extent than transfection, and prevented the induction of HSP70. In contrast to antisense-treated cells, MBP synthesis was essentially blocked in quercetin-treated cells even though levels of HSP25 and GRP78 increased. Taken together, these observations (a) indicate that HSP70 partially compensates for decreased chaperoning of nascent MBP by HSC70 (HSC70 and HSP70 are closely related and perform similar functions); (b) preclude the involvement of HSP25 and GRP78 in MBP synthesis; and (c) emphasize the requirement of HSC70 for optimal synthesis of MBP.

    Notes: Abstract Purpose. To investigate the potential of chitosan nanoparticles as a system for improving the systemic absorption of insulin following nasal instillation. Methods. Insulin-loaded chitosan nanoparticles were prepared by ionotropic gelation of chitosan with tripolyphosphate anions. They were characterized for their size and zeta potential by photon correlation spectroscopy and laser Doppler anemometry, respectively. Insulin loading and release was determined by the microBCA protein assay. The ability of chitosan nanoparticles to enhance the nasal absorption of insulin was investigated in a conscious rabbit model by monitoring the plasma glucose levels. Results. Chitosan nanoparticles had a size in the range of 300−400 nm, a positive surface charge and their insulin loading can be modulated reaching values up to 55% [insulin/nanoparticles (w/w): 55/100]. Insulin association was found to be highly mediated by an ionic interaction mechanism and its release in vitro occurred rapidly in sink conditions. Chitosan nanoparticles enhanced the nasal absorption of insulin to a greater extent than an aqueous solution of chitosan. The amount and molecular weight of chitosan did not have a significant effect on insulin response. Conclusions. Chitosan nanoparticles are efficient vehicles for the transport of insulin through the nasal mucosa.

    Notes: Abstract Purpose. The aim of this study was to investigate the interaction between the components of novel chitosan (CS) and CS/ethylene oxide-propylene oxide block copolymer (PEO-PPO) nanoparticles and to evaluate their potential for the association and controlled release of proteins and vaccines. Methods. The presence of PEO-PPO on the surface of the nanoparticles and its interaction with the CS was identified by X-ray photoelectron spectroscopy (XPS). The mechanism of protein association was elucidated using several proteins, bovine serum albumin (BSA), and tetanus and diphtheria toxoids, and varying the formulation conditions (different pH values and concentrations of PEO-PPO), and the stage of protein incorporation into the nanoparticles formation medium. Results. BSA and tetanus and diphtheria toxoids were highly associated with CS nanoparticles partly due to electrostatic interactions between the carboxyl groups of the protein and the amine groups of CS. PEO-PPO also interacted electrostatically with CS, thus competing with the proteins for association with CS nanoparticles. A visible amount of PEO-PPO was projected towards the outer phase of the nanoparticles. Proteins were released from the nanoparticles at an almost constant rate, the intensity of which was closely related to the protein loading. Furthermore, the tetanus vaccine was released in the active form for at least 15 days. Conclusions. CS and CS/PEO-PPO nanoparticles prepared by a very mild ionic crosslinking technique are novel and suitable systems for the entrapment and controlled release of proteins and vaccines.

    Notes: Abstract The transformation of α-pinene was studied in both gas and liquid phase over a series of faujasite‐like zeolites having different framework composition. A comparison with an amorphous silicoaluminate having 13 wt% of Al2O3 (SA) was made. Camphene + limonene were the major products observed in liquid phase at 393 K at conversion levels lower than 90%. At 473 K, where the reaction in gas phase took place, limonene suffered an ulterior transformation mainly into terpinenes and terpinolenes and a faster deactivation of the zeolite catalysts as compared to the one in liquid phase was observed. This behaviour contrasts with the high stability shown by SA in gas phase. Over the wider pore system of SA undesired heavy products were formed in liquid phase at the expenses of the terpene isomers. However, in gas phase this ulterior transformation of monoterpenes was minimised. A maximum activity for the zeolite catalysts was observed when the number of AlIV per unit cell reached about 17, a value where the population of Si(0Al) sites has been shown to reach the maximum.

    Notes: A chromium-substituted silicoaluminophosphate molecular sieve (CrAPSO-11) and a Cr-supported SAPO-11 molecular sieve (Cr/SAPO-11) were studied by XPS and reduction-oxidation (redox) cycles. The distribution of oxidation states as well as the average oxidation state for chromium were determined for both preparations. The XPS results showed that two chromium 2p doublets (Cr(VI) and Cr(III)) were necessary to curve fit the experimen-tal data. Comparison of curve-fitting results for the two oxidic samples (O2, 773 K) revealed a significantly higher percentage of Cr(III) species in the Cr/SAPO-11 catalyst. The difference between the two samples remained following reduction (H2, 773 K). For the CrAPSO-11 sample, Cr(VI) showed a higher stability towards reduction, compared to chromium supported on conventional supports. This result suggests the incorporation of Cr(VI) into the silicoaluminophosphate framework, as mentioned in the literature. For the Cr/SAPO-11 solid, Cr(III) was extremely stable towards oxidation at 773 K. Due to the redox characteristics and the XPS results, the formation of chromia species on this catalyst is suggested.

    Notes: Friedel-Crafts acetylation of phenanthrene (1a) in sym-tetrachloroethane yields mixtures of 2-, 3- and 9-acetylphenanthrenes (2a, 3a, 4). The distribution of isomers is found to depend strongly upon the method of mixing the reagents. Acetylation of 9-chlorophenanthrene (1b), performed by a variety of methods and solvents, led mainly to 3-acetyl-9-chlorophenanthrene (3b) (≥85%). Previously unreported 2-acetyl-9-chlorophenanthrene (2b) was found to form up to a maximum 11% in nitrobenzene.
