    Notizen: A novel two-stage bioreactor has been designed for a combined submerged (SF) and solid substrate fermentation (SSF) of wheat straw. The straw was pretreated with steam, and cellulases from the culture fluid of Trichoderma reesei were adsorbed on it for increased bioconvertibility. SSF was conducted in the top part of the bioreactor by inoculating the straw with a 36-h mycelial culture of T. reesei, or Coriolus versicolor. In the bottom part of the fermenter, Endomycopsis fibuliger was grown in SF. The SF liquor was recirculated through the SSF stage at 24 h intervals to remove glucose and other metabolites that may inhibit growth, and to maintain optimum moisture level and temperature. The removed glucose and other metabolites provided nutrients for the yeast in the SF stage. The combined fermentation resulted in overall higher biomass yield, increased bioconversion, increased cellulase production, and increased digestibility compared with single SSF or SF.

    Notizen: The present paper deals with almost wasteless technologies of pretreatment and obtaining of sugars and biomass from straw and potato tops. Several variants ensure a 1 t of straw per hour productivity using original auger reactors 290 and 150 mm in diameter. The authors have studied the submerged and solid state cultivation of various microorganisms applying the obtained substrates.

    Notizen: L-lysine synthesis pathway enzyme activities: β-aspartate kinase (EC.2.7.2.4), diaminopimelate decarboxylase (EC.4.1.1.20) for two L-lysine producing strains Brevibacterium flavum 22LD and RC-115 were studied. It has been found that β-aspartate kinase and diaminopimelate decarboxylase in the Br. flavum RC-115 are less sensitive to feed-back inhibition by lysine and threonine. It is supposed that desensitized β-aspartate kinase in the Br. flavum RC-115 can be determined by genetical changes of the regulatory properties of the β-aspartate kinase.Auxotrophity in the locus of homoserine dehydrogenase was tested and no homoserine dehydrogenase (EC.1.1.1.3) activity was found in either strain.The combination of these both types of mutation supplemented by the lack of catabolic repression in the RC-115 strain makes it an active lysine producer in the medium with high carbohydrates content.

    Notizen: The growth energetic efficiency (η) of two mycelial forms of Thielavia terrestris (pellets and diffused form) was studied by different methods. η Values determined by the pulse method are similar for the two forms, but the values determined by C balance for pellets were lower than those for diffused mycelium. These balance data prove that pellets yield more extracellular products than the diffused mycelium form, which is also confirmed by experimental data for different amounts of carbon in the culture fluid.Growth efficiency can be determined by various methods based on the principles of mass and energy balance. The estimates most frequently used are the biomass and substrate balances. However, growth efficiency determination according to oxygen balance (particularly by the pulse method) is simpler and more accurate; as it makes possible the immediate fixation of changes in the physiological condition of microorganisms and the determination of complex substrate utilization efficiency [1]. Earlier the possible use of this method for evaluating the growth efficiency of heterotrophic bacteria [2], hyphalic and yeast forms of microscopic fungi [3] was shown. The aim of the present study is the comparative investigation of the growth efficiency of two mycelial structures (hyphalic and pellets) by different methods as well as by pulse additions.

    Notizen: The review characterizes the submerged and solid state fermentation processes (SF and SSF) for obtaining of microbial biomass protein (MBP) on cellulose- and lignin-containing (CL) agricultural wastes (straw, wine, branches of fruit trees). There are discussed other alternatives as well. The main technological parametres are presented (protein (biomass) yield, degree of available substrate utilization, process efficiency, etc.). The use of a special stirring system of the authors' design for mycelial cultures results in an increase of CL substrate concentration in the initial nutrient medium from 2% to 8%of dry matter (DM) and in a threefold increase of the specific productivity (from 0.07 g biomass/l · h to 0.21 g biomass/l · h). The technological parametres can be increased also in the processes with substrate addition. The obtained preparations are analyzed as to their usefulness for the feeds of ruminants.
