    Notes: Most studies on the biochemistry and structure of the corpus luteum have focused on elucidating the processes of progesterone synthesis and release. In the present work, the histochemical composition of the corpus luteum of the rat was evaluated using lectinhistochemistry on rats at the end of pregnancy (days 18–23). We also analysed the morphology of the luteal cells, to characterize the changes attributable to regression in this organ. Seven biotinylated lectins were used (CON-A, WGA, DBA, SBA, PNA, RCA and UEA-I) following pre-set protocols (ABC method). The average diameter and area of the cells and their nuclei were measured. High reactivity of the luteal cells was observed with CON-A and a lower reactivity with WGA. The capillary endothelium gave positive reactivity with WGA and to a certain extent with SBA, PNA and RCA. Vesicular structures were intensely stained with DBA, and were more abundant in sections from animals with more advanced pregnancy, which could be attributable to cellular debris, on the basis of their morphologic characteristics. There were no significant differences among the cytometric variables analysed in comparisons of the values corresponding to the different days of gestation. These observations, together with previous research, suggest that, on the day of delivery, the corpus luteum of the rat is in the very early stages of structural regression, with no changes at the morphological level, but with changes at the molecular level.

    Notes: The aims of this study were to detect the expression of intermediate filaments and to verify the existence of marker substances for neuronal and neuroendocrine cells within the interstitial Leydig cells of laboratory rodent's testes, such as it has been described in other species. Adult male rats, mice, gerbils, Syrian hamsters and guinea-pigs were used and the localization of the different markers was achieved by the streptoavidin–peroxidase immunohistochemical method. The present study demonstrates in all rodents studied a similar pattern of localization in Leydig cells of intermediate filaments (vimentin, cytokeratin, neurofilament 200 kD and glial fibrillary acidic protein) and other marker substances (S-100, CgA, substance P and neurone-specific enolase), which are typical of neuroendocrine (APUD cells or paraneurones) and glial cells. The expression of these substances, related to neurotransmitters or neurohomones and other proteins characteristic of neuroendocrine cells, could suggest that it is a neural crest derived cell. Although this study provides more evidences about the immunoexpression of neuronal and glial markers in Leydig cells, this fact cannot be related directly to their embryological origin, because the current data support the hypothesis of a mesenchymal origin of the Leydig cells.

    Notes: Abstract Fove whole bronchoalveolar lavages were performed in 2 patients with pulmonary alveolar proteinosis with continuous monitoring of mixed venous and arterial oxygen saturation. Hemodynamic parameters and gas-exchange status were measured during the different phases of the lavage. In the phase of filled lung, a significant increase of arterial partial pressure (PaO2) and arterial saturation of oxygen were observed, secondary to a decrease in the intrapulmonary shunt. The mean pulmonary arterial pressure, pulmonary vascular resistances and cardiac index were higher during the filling of the lung as compared to the controls. During the empty lung phase, although PaO2 decreased (without reaching statistical significance), due to an increase in the intrapulmonary shunt, the increase in cardiac output during this phase left the oxygen delivery (DO2) unchanged.

    Description / Table of Contents: Zusammenfassung Wir haben die Variation der spezifischen Aktivität des wichtigsten lysosomalen Enzyms untersucht: Cathepsin D. Diese Untersuchung ist in Schnittwunden in Schweinehaut entwickelt worden im Hinblick auf die Anwendung der Methode zur Differentialdiagnose zwischen vitalen und postmortalen Wunden. Gleichzeitig wurden die Histamin- und Serotonin-Konzentrationen bestimmt, um einen Vergleich mit den Cathepsin-Ergebnissen zu ermöglichen. Unsere Ergebnisse zeigen die Brauchbarkeit dieses neuen Markers, um die Vitalität von Wunden zu bestimmen, auch bei sehr kurzen Überlebenszeiten. Trotzdem besteht keine Korrelation zwischen den Cathepsin-D-Ergebnissen und den Bestimmungen der Histaminund Serotonin-Gehalte. Das ist ein Befund, der uns daran denken läßt, daß dieser neue Marker nicht zur Wundaltersbestimmung geeignet ist.
    Notes: Summary We have studied the variation of specific activity of the most important lysosomal enzyme: Cathepsin D. This study has been developed in pig skin incision wounds for its application to the differential diagnosis between vital and postmortem wounds. At the same time, we have determined histamine and serotonine levels to make a comparative study with Cathepsin results. Our results show the utility of this new marker to determine the vitality of wounds, even when the time elapsed until death has been very short. Nevertheless, there is no correlation between Cathepsin D results and those of histamine or serotonin, a fact that induces us to think that this new marker is not useful to determine wound age.

    Notes: Abstract Since 1992 the Spanish and Portuguese Working Group (GEP) of the International Society for Forensic Haemogenetics (ISFH) has been organizing collaborative exercises on DNA profiling with the aim of making progress on standardization and discussing technical and statistical problems in DNA analysis. A total of four exercises (GEP-92 to GEP-95) have been carried out until now. A consequence of these exercises was the creation of a quality control programme in Spain and Portugal in 1995 which was carried out simultaneously with the GEP-95 exercise. The number of participating laboratories increased from 10 in the first exercise (GEP-92) to 19 in the last exercise (GEP-95). Despite this increasing number of participating laboratories, results remained satisfactory. In the last exercises, all the laboratories used PCR-based DNA polymorphisms with an increasing number of markers obtaining good results. SLPs were used by only 30% of laboratories in the last two exercises but the results indicated a good level of expertise in most of these laboratories. The reasons for these successful results are the common use of the EDNAP protocol for SLP analysis and commercially available kits or common sequenced allelic ladders for PCR-based DNA polymorphisms.

Wir haben 53 Proben mit Hilfe enzymatischer und histologischer Techniken untersucht. Die Ergebnisse zeigen die Fähigkeit von Cathepsin D, den vitalen Ursprung von Wunden zu etablieren, selbst wenn sie 5 Minuten oder geringere Zeit vor dem Tod gesetzt wurden. In den letzteren erreichte die spezifische Aktivität von Cathepsin D 0,055 Einheiten an den Wundränden und 0,01 Einheiten in den entsprechenden Kontrollen (P 〈 0,001). Wie früher in experimentellen Untersuchungen gezeigt wurde, scheint Cathepsin D ein sehr nützlicher Marker von hoher forensischer Bedeutung besonders in schwierigen Fällen zu sein. Weitere Studien sind in der Bearbeitung, um den Einfluß verschiedener Faktoren, wie Medikamenteneinnahme und klinischer Zustand, auf die Cathepsin D-Aktivität zu überprüfen.
    Notes: Summary This paper shows the results obtained by studying the lysosomal enzyme Cathepsin D as a potential marker for the vitality of wounds in human specimens. We have analyzed 53 samples using enzymological and histological techniques. Our results show the ability of Cathepsin D to establish the vital origin of wounds inflicted 5 minutes or less before death, where the specific activity of cathepsin D reached 0.055 units at the wound edge and 0.01 units in their respective controls (P 〈 0.001). As previously demonstrated in an experimental series, Cathepsin D seems to be a very useful marker of high forensic interest in especially difficult cases. Further studies are in progress to check the influence of different factors such as drugs intake and clinical conditions on Cathepsin D activity.
