    Notes: The gene cluster (rfb region) which determines the biosynthesis of the Shigella flexneri O-antigen of the Y serotype specificity was cloned from a S. flexneri serotype 2a strain. Two plasmids, pPM2212 and pPM2213, which conferred O-antigen biosynthesis were generated from separate cosmid clones by deletion with Cla l. These plasmids expressed O-antigen in Escherichia coli K12 like that of the parental strain, as assessed by reactions to antisera in colony and Western immunoblots, sensitivity to bacteriophage Sf6, and by silver staining of lipopolysaccharides separated by sodium dodecyl sulphate/polyacryl-amide gel electrophoresis. These plasmids also mediated O-antigen expression in an E. coli K12 rfb-delete background, indicating that all the necessary genes have been cloned. A detailed restriction map of the region has been constructed and analysis of various subclones has allowed the limits of the coding region for O-antigen biosynthesis to be defined to a maximum of 11kb. Expression of these plasmids demonstrates a novel phenotype associated with control of lipopolysaccharide chain length. The gene(s) responsible maps adjacent to, but separate from, those associated with the biosynthesis of the O-antigen unit. Analysis of plasmid-encoded proteins in minicells and maxicells has facilitated the construction of a physical map. Finally, plasmid pPM-2212 was used to probe a collection of S. flexneri serotypes by Southern hybridization. With the exception of serotype 6, which appears to be unrelated, a similar pattern was found in all serotypes.

    Notes: Abstract Park wheat (Triticum aestivum cv. Park) is susceptible to stem melanosis when grown on Cu-deficient soils. Three field experiments were conducted to evaluate the effectiveness of various Cu sources, using different methods and rates of application for reducing the incidence of stem melanosis and increasing the grain yield of Park wheat on a Cu-deficient Black Chernozemic soil in central Alberta. A fourth experiment determined the residual effect of Cu over 4 years. In the first experiment, Park wheat had less disease and higher grain yield when Cu-chelate solution at 2 and 4 kg Cu ha−1 or Cu-sulfate solution at 10 and 20 kg Cu ha−1 was applied to the soil surface and incorporated to a depth of 8 cm or was applied at these rates as a foliar spray. The foliar application, however, was phytotoxic and delayed maturity by approximately two weeks. Sidebanding Cu (4 cm to the side and 4 cm below the seed row) was least effective. In the second experiment, Cu-sulfate solution incorporated into the soil was more effective than the soil incorporated granular Cu-sulfate in reducing disease incidence and increasing grain yield in the year of application. In the following year the granular Cu was as effective as solution Cu. The third experiment showed that Cu seed dressings did not have any effect on disease incidence or grain yield of Park wheat. In the fourth experiment, the residual effect of Cu-chelate was evident four years after application. The grain yield in the fourth year was about four times that of the control.

    Description / Table of Contents: Sommaire Cet article décrit l'étude, la réalisation et le mode de fonctionnement d'un ensemble de micrométrie par télévision réalisé tout particulièrement pour la mesure intra-buccale, en exploitation clinique, de la déterioration marginale d'une obturation dentaire. Schématiquement l'ensemble de micrométrie intra-buccale par télévision est constitué d'un microscope relié à une système de télévision en circuit-fermé à double caméra exploité par des circuits de mesure. Une zone circulaire de 1 mm de diamètre de la périphérie du plombage peut être projetée en une serie d'images magnifiées 250 fois avec une erreur de linéarité mesurée, inférieure à 2μ. L'effet des mouvements du patient est limité, en raison de cette amplification, par l'utilisation d'un tube à mémoire qui conserve l'image pendant la mesure.
    Abstract: Zusammenfassung Die vorliegende Arbeit beschreibt Entwicklung, Konstruktion und Arbeitsweise eines Fernseh-Mikroskopinstruments, das speziell zur intraoralen und unter klinischen Bedingungen durchgeführten Messung der Adaptation im vom Rande ausgehenden Zersetzungsprozess in Zahnplomben der Milchzähne konstruiert wurde. Das Prinzip des intraoralen Fernsehmeßinstruments besteht in der Verbindung eines petrographischen Mikroskops mit einem geschlossenen Fernsehsystem aus zwei Kameras. Die Schaltung erfolgt durch Scan-Linienmessung. Ein kreisförmiges Gebiet von 1-mm Durchmesser in der cavumfüllenden Randzone kann serienmäßig in 250facher Vergrößerung betrachtet werden. Dabei können lineare Meßunterschiede von 2μ erfaßt werden. Die Bewegung des Patienten wird bei diesen Vergrößerungen mit einer Permachonröhre (elektronisches Gedächtnis) begrenzt; die Röhre speichert das Bild während des Meßvorgangs.
    Notes: Abstract The paper describes the development, design and operation of a television microscopic instrumentation which was specifically constructed to measure intra-orally, under clinical conditions, the adaptation of the cavo-filling margin deterioration of restorations in primary teeth. Basically, the intra-oral television micromeasurement instrumentation is achieved by coupling a petrographic microscope to a two-camera closed-circuit television system through scan line measurement circuitry. A 1-mm dia. circular area of the cavo-filling margin can be serially viewed at 250 × magnification with linear measurement change as small as 2μ noted. Movement of the patient, at these magnifications, is limited by the use of a permachon (electronic memory) tube, which retains the image during measurement.

    Description / Table of Contents: Sommaire L'article décrit un ensemble d'instrumentation mis au point pour l'exploitation d'un équipement de microscopie intra-buccale par télévision et d'un micro et macro densitomètre par télévision. Il peut également s'appliquer à tout autre équipement dont l'exploitation se fait sur écran de télévision. Cet appareil est susceptible de recueillir une information issué d'une ligne donnée et/ou d'un point donné d'une ligne d'une image télévisée. Il est décrit dans l'optique de sa réalisation et de sa mode de fonctionnement. La possibilité qu'il offre de référer des zones de mesure par marquage lumineux se superposant à l'image rend l'appareil apte à calibrer l'ensemble de la chaine de mesure indépendamment de l'aggrandissement apporté par l'image.
    Abstract: Zusammenfassung Ein Meßsystem wird dargestellt, das zur Verwendung beim intraoralen Fernsehmikroskop und bei dem Fernseh- Mikro- und Makrodichteinstrument entwickelt wurde. Es kann auch zu Fernseh-abbildungen bei anderen Eingangsquellen benutzt werden. Das Gerät, das Meßdaten von einer bestimmten Scan-Linie und/oder jedem Punkt innerhalb der Scan-Linie des Fernsehbildes aufnehmen kann, wird hinsichtlich Konstruktion und Arbeitsweise diskutiert. Das Instrument kann unabhängig von der Vergrößerung des Fernsehbildes geeicht werden; das geschieht mit Hilfe der Identifizierung der Meßfläche und mit erleuchteten Markierungspunkten innerhalb der gemessenen Scan-Linie.
    Notes: Abstract Measurement instrumentation is presented that has been designed for use with the Intra-oral Television Microscope and Television Micro and Macrodensity Instrumentation. It can also be applied to any television display from other input sources. The equipment, which is capable of collecting measurement data from a particular scan line and/or any point within a scan line of the television image, is discussed from the standpoint of design and operation. Identification of the area measured, along with illuminated reference marker dots within the scan line being measured, make it possible to calibrate the instrumentation, regardless of the magnification of the televised image.
