Notes: The effect of intracerebroventricular (i.c.v.) injection or infusion of various doses of corticotropin-releasing hormone (CRH) on the LH and FSH surge was studied in pro-oestrous rats supplied with a jugular vein and an i.c.v. cannula. Additionally, we investigated if arginine vasopressin (AVP) was involved in the CRH-induced alterations to the surge of gonadotropins. I.c.v. injection of 10 μg CRH given 5 min before the presumed onset of the LH surge caused a strong inhibition of the LH surge and a slight inhibition of the FSH surge. Three to four h after CRH injection, its inhibitory effect diminished. A 6 h i.c.v. infusion of CRH started 1 h before the presumed onset of the LH surge, caused a dose-related inhibition of the LH and FSH surge. Infusion of 1 μg/h CRH did not suppress the surge of both hormones while infusion of 5 or 10 μg/h CRH inhibited the LH surge. Infusion of 10 μg/h CRH caused a strong suppression of plasma LH during the first 3 h of the LH surge. Despite continuation of CRH infusion, the inhibitory effect disappeared and plasma LH increased to similar levels as in controls at corresponding points of time of the LH surge. The FSH surge was also suppressed by infusion of 10 μg/h CRH. The surge of LH and FSH was not affected by a 9-h infusion of 10 μg/h CRH started 4 h before the presumed onset of the LH surge. This observation also indicates that the inhibitory effect of CRH may last for only 3–4 h. The surge of LH and FSH was not affected by i.c.v. injections of AVP-antiserum. However, pretreatment with AVP-antiserum prolonged the inhibitory effect of CRH on the LH surge. In conclusion, CRH can inhibit the pro-oestrous LH and to a lesser extent the FSH rise for only 3–4 h after the beginning of CRH administration. AVP may play a role in limiting the inhibitory effect of CRH on LH to 3–4 h.

Notes: Summary Adult Cichlasoma biocellatum were kept in deionized water, 25% sea-water and tap-water. The mucous cells in the epithelium of the gills and the buccal floor were not affected by the osmotic experiments. In animals kept in deionized water the mucous cells in the skin remained unaffected, but the ionocytes (chloride cells) in the gills were strongly stimulated. Keeping the animals in salt water led to a strong regression of the epidermis including the mucous cells, and of the ionocytes. The regression could be counteracted by prolactin injections. The changes in the skin and the ionocytes could be correlated with the activity of the prolactin producing cells in the adenohypophysis. It is suggested that the epidermis and its mucous cells as well as the ionocytes are under prolactin control.

Notes: Summary In the adenohypophysis of Cichlasoma biocellatum, which may be divided into a pro-, meso-, and meta-adenohypophysis, seven cell types can be distinguished. Treatment with deionized- and salt water effects three of these cell types. The rostral acidophils, which were proved to produce prolactin by means of an immunochemical method, are distinctly stimulated in deionized water and markedly inactivated in 25% sea-water. The putative ACTH cells, bordering neurohypophysial branches in the pro-adenohypophysis, are moderately stimulated by both treatments. The presumptive TSH cells, which in control animals appear to be very active, do not show marked changes in deionized water; they are strikingly inactivated, however, in salt water specimens.

Notes: Summary In order to identify the thyrotropic cells in the pituitary gland of Cichlasoma biocellatum, adult animals were treated with thyroxine. Periodic acid Schiff (PAS)- and aldehyde fuchsin (AF)-positive, and Alcian blue (AB)-negative basophils in the meso-adenohypophysis as well as the thyroid were strongly inactivated by such treatment. Since other cell types in the adenohypophysis were not affected, it is concluded that the PAS-positive and AB-negative basophils in the meso-adenohypophysis are the thyrotrops. In normal animals the thyrotrops and the thyroid are very active. Propylthiouracil did not cause a further activation of TSH cells. Keeping the animals in 25% sea-water caused a strong inactivation of the prolactin cells, the thyrotrops and the thyroid. In deionized water the prolactin cells were stimulated, but the thyroid was less active than in controls. Injections of ovine prolactin did not enhance the activity of the thyroid in salt water animals. It is suggested that in Cichlasoma biocellatum prolactin does not have a direct stimulatory effect on the thyroid.

Notes: Summary The development and activity of the basophils in the meso-adenohypophysis of Anopthichthys jordani were studied in relation to the maturation of the gonads. In addition, pituitaries and ovaries of post-spawning females were studied during a period 0–11 days subsequent to spawning. The large and mainly PAS-positive basophilic cells in the meso-adenohypophysis were the only cell type that showed a correlation with the development and maturation of the ovaries and testes. These cells were subjected to a partial or complete degranulation in females following oviposition, when yolk formation in the ovary was at a maximum. It is concluded that the large PAS-positive basophils in the mesoadenohypophysis have a gonadotropic function and that only one gonadotropic cell type is present in the adenohypophysis of Anoptichthys jordani.

Notes: Summary In order to identify the TSH cells in the pituitary gland of Anoptichthys jordani, the development of the adenohypophysial cell types was studied in relation with the differentiation of the thyroids. In addition experiments were performed in young and adult animals with propylthiouracil (PTU) and thyroxine. At the time of the first thyroid development two meso-adenohypophysial cell types in the pituitary gland of Anoptichthys jordani are distinctly differentiated: orangeophilic cells and Alcian Blue (AB)-positive basophils. Because thyroxine and PTU could evoke changes in activity of the latter but had no effect on the orangeophils, it is concluded that the angular AB-positive basophils in the meso-adenohypophysis of Anoptichthys are responsible for the TSH production. PTU and thyroxine were unable to affect TSH cell activity in adult animals.

Notes: Summary In the pituitary gland of the Stenohaline teleost Anoptichthys jordani the pro-adenohypophysial acidophilic cells undergo a marked hypertrophy and hyperplasia in deionized water, whereas their activity is suppressed in 20% sea-water. Consequently, it is concluded that these cells are involved in osmoregulation. Correlated changes occur in the mucous cells of the epithelium in the oral cavity; these cells are stimulated in deionized water and are degenerating in salt water. The mucous cells in the epithelium of the gills show an inhibition in salt water, whereas the mucous cells in the epidermis are unaffected by either treatment. Since ovine prolactin may also induce a stimulation of the mucous cells in the buccal cavity, it is supposed that the osmoregulatory factor, produced in the adenohypophysis, is a prolactin-like hormone.

Notes: Summary Staining sagittal sections of the pituitary of the Blind Mexican Cave Fish, Anoptichthys jordani, with a number of methods including Herlant's tetrachrome, Herlant's Alcian Blue (AB)-periodic acid Schiff (PAS)-orange G, Gabe's aldehyde fuchsin and lead hematoxylin according to Mac Conaill, made it possible to distinguish seven tinctorial cell types in the adenohypophysis: in the pro-adenohypophysis erythrosin-positive acidophils, and lead hematoxylin-positive amphiphils, lining branches of the neural lobe; in the meso-adenohypophysis orange G-positive acidophils, large, weakly AB- and AF-positive, strongly PAS- positive basophils, and smaller strongly AB- and AF-positive basophils; in the metaadenohypophysis lead hematoxylin-positive and -negative cells. These cell types are compared with those in other teleosts.

Notes: Summary Administration of the anti-adrenal drug metopirone (SU 4885) allows for the identification of the ACTH cells in the adenohypophysis of the teleost Anoptichthys jordani. The drug causes a correlated stimulation of the interrenal cells and one cell type in the pituitary gland, i.e. the amphiphilic pro-2 cells, situated in a palisade like layer between the acidophils of the pro-adenohypophysis and the neurohypophysis.
