    Notes: Abstract. Nitric oxide (NO) is synthesized in neurons and is a potent relaxor of vascular and nonvascular smooth muscle. The uterus contains abundant NO-synthesizing nerves which could be autonomic and/or sensory. This study was undertaken to determine: 1) the source(s) of NO-synthesizing nerves in the rat uterus and 2) what other neuropeptides or transmitter markers might coexist with NO in these nerves. Retrograde axonal tracing, utilizing Fluorogold injected into the uterine cervix, was employed for identifying sources of uterine-projecting neurons. NO-synthesizing nerves were visualized by staining for nicotinamide adenine dinucleotide phosphate (reduced)-diaphorase (NADPH-d) and immunostaining with an antibody against neuronal/type I NO synthase (NOS). NADPH-d-positive perikarya and terminal fibers were NOS-immunoreactive (-I). Some NOS-I/NADPH-d-positive nerves in the uterus are parasympathetic and originate from neurons in the pelvic paracervical ganglia (PG) and some are sensory and originate from neurons in thoracic, lumbar, and sacral dorsal root ganglia. No evidence for NOS-I/NADPH-d-positive sympathetic nerves in the uterus was obtained. Furthermore, double immunostaining revealed that in parasympathetic neurons, NOS-I/NADPH-d-reactivity coexists with vasoactive intestinal polypeptide, neuropeptide Y, and acetylcholinesterase and in sensory nerves, NOS-I/NADPH-d-reactivity coexists with calcitonin gene- related peptide and substance P. In addition, tyrosine hydroxylase(TH)-I neurons of the PG do not contain NOS-I/NADPH-d-reactivity, but some TH-I neurons are apposed by NOS-I varicosities. These results suggest NO-synthesizing nerves in the uterus are autonomic and sensory, and could play significant roles, possibly in conjunction with other putative transmitter agents, in the control of uterine myometrium and vasculature.

    Notes: After injection of true blue (TB) into the pericardial sac and nuclear yellow (NY) into the left ulnar nerve of rats, most perikarya in the left eighth cervical and first thoracic dorsal root ganglia are single-labeled with either TB or NY. However, 7.1-14.7% of the sensory perikarya from the cardiac area labeled with TB are simultaneously labeled with NY. This finding indicates that some primary sensory perikarya receive processes from both the left arm and the heart. In addition to other mechanisms that may be operative in the referral of pain of cardiac origin to the medial left arm, these results suggest that sensory neurons with dichotomizing somatic and visceral peripheral processes may also contribute to this phenomenon.

    Notes: The endocrine pancreas of the channel catfish is segregated into a large primary islet and numerous smaller secondary islets. In view of cell distribution differences in mammalian islets of ventral and dorsal primordia, we have determined the percentage volumes of insulin-, glucagon-, and somatostatin-containing cells in primary and secondary catfish islets to ascertain if these islets correlated with those derived from ventral and dorsal primordia in mammals. Islets were immunocytochemically stained using antisera to anglerfish insulin, porcine glucagon, and synthetic somatostatin and volume densities were quantified on light micrographs by point-counting procedures. In both primary and secondary islets the insulin-, glucagon-, and somatostatin-containing cells comprised approximately 32%, 23%, and 38% of the endocrine cell volumes, respectively. Therefore, the cell populations did not reflect any embryological differences between the two groups of islets. In this study, the volume densities of insulin-reactive cells in the primary islet were less than previously reported, and the overall insulin staining was about one-half of that seen in mammals. The volume density of somatostatin-reactive cells in primary islets was greater than previously reported. Based on these data, primary and secondary islets of the catfish do not appear to have a similar development to the ventral and dorsal islets of the mammalian system.

    Notes: Afferent perikarya in dorsal root ganglia (DRG) at the T13 and L1 segmental levels projecting to the rat ovary were identified by utilizing the fluorescent retrograde tracer true blue (TB). Subsequently, TB-labeled ovarian afferent perikarya in DRG were examined for vasoactive intestinal polypeptide (VIP), substance P (SP), cholecystokinin-8 (CCK-8), neuropeptide Y (NPY), and somatostatin (SOM) immunoreactivity and for the presence of fluoride-resistant acid phosphatase (FRAP) enzyme activity. Of the ovarian afferent perikarya at the T13 and L1 segmental levels, 20.5% displayed VIP immunoreactivity, 23.8% displayed SP immunoreactivity, and 43.1% were immunoreactive for CCK-8. No ovarian afferent perikarya contained SOM or NYP immunoreactivity or FRAP activity. It is suggested that different neuropeptides may participate in modulation of specific ovarian functions.
