Summary In supravital staining with toluidine blue, globular doubly refracting structures occur in the protoplasma of the granulated leukocytes, of the lymphocytes, and of the monocytes of the peripheral blood. According to the intensity and strength of the anisotropy, they can be classified into small, medium, and large structures. The number and distribution of these doubly refracting structures may vary from cell to cell and from cell type to cell type. In healthy humans small and medium-sized structures of this type can be demonstrated in most neutrophilic leukocytes; large structures occur only in isolated cases. Monocytes are distinguished by a relatively large number of small and medium-sized doubly refracting structures in the individual cell, whereas lymphocytes reveal relatively few, mostly small structures of this type. According to the polarization-optical behaviour of these doubly refracting structures, their fine structure consists of tightly superposed globular coats, into which toluidine blue is selectively deposited. The longer axis of the particles of the stain are in a vertical position to the radius of the globules. The entire anisotropy of these formations consists of the double refraction of the stain particles selectively arranged at a plane and the particular double refraction caused by the superposition in layers of the individual globular coats (form double refraction).

Zusammenfassung Bei Supravitalfärbung mit Toluidinblau treten im Protoplasma der granulierten Leukozyten, der Lymphozyten und der Monozyten des peripheren Blutes kugelige, doppelbrechende Strukturen auf. Diese können nach ihrer Größe und der Stärke der Anisotropie in kleine, mittlere und große unterteilt werden. Die Anzahl und Verteilung dieser doppelbrechenden Strukturen kann von Zelle zu Zelle und von Zellart zu Zellart unterschiedlich sein. Beim gesunden Menschen sind in den meisten neutrophilen Leukozyten kleine und mittlere derartige Strukturen nachzuweisen, große nur vereinzelt. Die Monozyten zeichnen sich durch eine verhältnismäßig große Anzahl kleiner und mittlerer doppelbrechender Strukturen in der Einzelzelle aus, die Lymphozyten dagegen durch verhältnismäßig wenige, meist kleine derartiger Strukturen. Aus dem polarisationsoptischen Verhalten dieser doppelbrechenden Strukturen können wir schließen, daß sie feinstrukturell aus eng aufeinanderliegenden Kugelschalen aufgebaut sind, in welche Toluidinblau gerichtet eingelagert wird. Die Farbstoffteilchen liegen dabei mit ihrer längeren Achse senkrecht zum Radius der Kugeln. Die gesamte Anisotropie dieser Gebilde setzt sich zusammen aus der Doppelbrechung der in einer Ebene gerichtet angeordneten Farbstoffteilchen und jener Doppelbrechung, welche durch die schichtweise Übereinanderlagerung der einzelnen Kugelschalen (Formdoppelbrechung) hervorgerufen wird.
