    Notes: Summary The early part of the life cycle of Eimeria maxima has been reexamined. The occurrence of a schizont in the deep glands 48 h after oocyst inoculation has been confirmed and the schizont illustrated for the first time. Further observations indicate that three generations of schizonts occur before gametogony. At least three generations are necessary to correlate with the reproduction of this parasite judged by oocyst production.

    Notes: A simple gas chromatographic mass spectrometric determination of the impurity 3-quinuclidinyl benzilate in the anticholinergic agent clidinium bromide (3-hydroxy-1-methylquinuclidinium bromide benzilate) is not feasible because clidinium bromide decomposes to 3-quinuclidinyl benzilate in the injection port of the gas chromatograph; the trimethylsilyl derivative of clidinium bromide also decomposes to the trimethylsilyl derivative of 3-quinuclidinyl benzilate. Therefore, a quantitative thin-layer chromatographic gas chromatographic mass spectrometric method has been developed for determining 3-quinuclidinyl benzilate in clidinium bromide, using β-piperidylethyl benzilate as an internal standard. A band containing only 3-quinuclidinyl benzilate and β-piperidylethyl benzilate can be eluted from the thin-layer chromatographic plate. The samples are analyzed by single ion monitoring of m/z 255 produced by the trimethylsilyl derivatives of 3-quinuclidinyl benzilate and β-piperidylethyl benzilate which are separated by gas chromatography. A level of 0.03% of 3-quinuclidinyl benzilate can be determined with a precision of about 6%; the lower limit of detection of 3-quinuclidinyl benzilate alone is about 1 ng on the thin-layer chromatographic plate. Analysis of a reference standard of clidinium bromide by the thin-layer chromatographic gas chromatographic mass spectrometric procedure showed that it contained no detectable 3-quinuclidinyl benzilate. One commercial bulk sample of clidinium bromide contained about 0.018% of 3-quinuclidinyl benzilate, while a formulated sample of clidinium bromide contained no detectable 3-quinuclidinyl benzilate. Commercial samples of clidinium bromide containing chlordiazepoxide can be analyzed for 3-quinuclidinyl benzilate only with difficulty using this method.

    Notes: The mass spectra of twenty-six commercially available medicinal phenothiazines have been determined at high resolution. In each compound, the side chain attached to the 10-position of the phenothiazine ring contains nitrogen and fission of the C—C bond α to this nitrogen gives in most cases the base peak. Some of the compounds are also substituted at the 2-position and this substituent generally remains intact, though some substituents such as acetyl, propionyl, methoxyl and methylthio can undergo fragmentation. Fragmentations fall into three groups; those which give ions representing a part of the side chain; those which give ions representing the intact phenothiazine ring with part of the side chain attached; those which give ions representing a partially fragmented ring system.

    Notes: The main fragmentation sequences of glycollide and its homologues are initiated by fission of a CO—O bond, leading to the formation of fragment ions of low, m/e, such as [R1CO]+ and [CR1R2CCO]+. When a hydrogen atom is present on a ring carbon atom, 1,3 hydrogen migration occurs to produce [CHR2OH]+. In case where a ring carbon atom carries an alkylchain ≥ C2H5, a McLafferty rearrangement occurs with the adjacent carbonyl group. When both ring carbon atoms are dimethyl substituted, a 1,4 hydrogen migration must be invoked to account for the observed fragmentation sequence.

    Notes: Although the behaviour of 1H, 2H-2,1,3-benzothiadiazoline 2,2-dioxide is similar to that of sulphoxides in the mass spectrometer in losing a molecule of SO2, the 1,3-dimethyl derivative loses the radical SO2H. In addition the radical CH3SO2 is lost in a one step process that must involve a methyl migration. The radical SO2H is also lost from 2,1,3-benzothiadiazine 2,2-dioxides methylated in the 3-position and the hydrogen involved is shown to originate from this methyl group by deuterium labelling. The mass spectra of other 2,1,3-benzothiadiazine 2,2-dioxides are also discussed.

    Notes: The mass spectra of nineteen substituted benzothiazoles have been recorded and the identity of the various ions in the mass spectra has been established by high resulution (accurate) mass measurement. Deuterium labelling has been used to elucidate the fragmentation processes of these compounds. The parent compound of the series, benzothiazole, exhibits the loss of hydrogen cyanide and carbon monosulphide from the parent ion as the most important decomposition pathways. The hydrogen atom concerned in the loss of hydrogen cyanide is shown to originate from the 2-position of benzothiazole, while in 2-substituted benzothiazoles, different mechanisms are apparent for the loss of hydrogen cyanide, and these are clarified by deuterium labelling. Some substituted benzothiazoles can lose sulphur from their molecular ions, a process which does not occur in benzothiazole itself. The substituted benzothiazoles undergo many other types of fragmentations, in some cases retaining the substituent, and in other cases losing it prior to collapse of the thiazole ring.

    Notes: The mass spectra of some δ-lactones substituted in various positions by methyl groups have been studied both by accurate mass measurements at high resulution and by deuterium labelling. The loss of carbon dioxide has been shown to be important only for a mono-substituted lactone. A fragmentation mode common to all the lactones studied is the elimination of the ring oxygen atom, plus the adjacent carbon atom with its substituents, as a neutral carbonyl molecule such as formaldehyde, acetaldehyde or acetone. Deuterium labelling has uncovered the existence of a rearrangement involving hydrogen transfer in an even-electron ion, and a mechanism involving a six-membered transition state is proposed for this. The bond fissions, common to all the lactones, by which the principal ions in the mass spectra arise, are summarised.
