    Notes: Background.  Both various virulence factors of Helicobacter pylori and host factors influence the clinical outcome of H. pylori infection. In animal experiments with Helicobacter felis, large variations in the severity of disease have been observed between different mouse strains infected with a single isolate of H. felis. C57BL/6 J mouse strain that lacks the expression of group IIA phospholipase A2 has been shown to develop more severe gastric inflammation than other mouse strains. Thus, group IIA phospholipase A2 has been suggested to play a role in regulating inflammation in gastric mucosa. The aim of this study was to examine the possible role of group IIA phospholipase A2 in experimental Helicobacter infection.Materials and Methods.  Transgenic mice expressing human group IIA phospholipase A2 and their group IIA phospholipase A2 deficient nontransgenic C57BL/6 J littermates were infected with H. felis. The mice were killed 3, 8, and 19 weeks after inoculation of bacteria to determine the histopathological changes in gastric mucosa.Results.  The infected mice developed chronic inflammation in gastric mucosa. We found no differences in the colonization of bacteria between transgenic and nontransgenic mice. At 3 and 8 weeks, no difference was found in the severity of inflammation between the two groups. Nineteen weeks after the administration of bacteria the inflammation was more marked in nontransgenic than transgenic mice. Group IIA phospholipase A2 was expressed by in situ hybridization in the neck cells of the glandular stomach in transgenic mice.Conclusions.  The results of the present study suggest that the endogenous expression of group IIA phospholipase A2 diminishes chronic inflammation in gastric mucosa in experimental H. felis infection in mice.

    Notes: Summary The development of left ventricular contracture and myocardial perfusion defect was studied in isolated rat hearts during global ischaemia of 90 min duration. The left ventricular pressure was measured by a balloon catheter inserted into the ventricle and filled with water. The pressure reached the maximum at 16 min of ischaemia. The left ventricular volume and compliance (passive distensibility) were measured by the same balloon, the former by connecting the balloon to an open catheter and the latter by applying a constant additional volume (0.020 ml) into the balloon. The left ventricular volume and compliance both decreased progressively for 20 min of ischaemia after which they remained low for the rest of the observation period (90 min). The myocardial perfusability was tested by infusing 0.1 per cent sodium fluorescein in isotonic saline into the cannulated aortic root of the isolated heart preparation. The percentage perfused with the fluorescent tracer in horizontal frozen myocardial sections was estimated by point counting from colour photographs taken under ultraviolet light. The proportion of the perfused area decreased gradually from 100% at 0 min of ischaemia to 93, 67, 43 and 37% at 15, 30, 60 and 90 min of ischaemia, respectively. It was concluded that ischaemic contracture of the left ventricle is followed by the development of a myocardial perfusion defect in isolated ischaemic rat heart.

    Notes: Summary Samples of gastric leiomyosarcomas from two male patients (69 years and 57 years of age) were studied by electron microscopy. The tumour cells contained abundant 50–90 å thick microfilaments and microtubules. Another cell type, regarded as immature leiomyosarcoma cell, contained numerous profiles of granular endoplasmic reticulum, free ribosomes and a well developed Golgi apparatus. There were intercellular junctions of zonula adherens-type between adjacent tumour cells. It was concluded that electron microscopy offers a valuable aid in the diagnosis of gastric neoplasms of smooth muscle origin.

Das spärliche Cytoplasma enthält normale Zellorganellen. Freie Ribosomen sind zahlreich und der Golgi-Apparat ist gut entwickelt. Die Kerne sind groß und unregelmäßig, die Nukleolen prominent. Einige Kerne enthalten “nuclear pockets” mit Material, das der cytoplasmatischen Grundsubstanz ähnlich ist. Reste degenerierender Zelle werden ebenfalls gefunden.
    Notes: Summary A case of primary lymphosarcoma of testis was studied by light and electron microscopy. The fine structure of the lymphosarcoma cells was simple and monotonous. The cytoplasm was scanty and contained normal-looking organelles. Free ribosomes were abundant and the Golgi apparatus was well developed. The nuclei were large and irregular in shape with prominent nucleoli. “Nuclear pockets” enclosing material similar to the cytoplasmic ground substance were found infrequently. Remnants of disintegrating cells were also encountered.

    Notes: Abstract Inbred BALB/c mice were given vinblastine sulfate (VelbeR) and/or pilocarpine hydrochloride (20 mg/kg) intraperitoneally and amylase activity was determined in the pancreas homogenate. When vinblastine was given 5 hours before measuring the pancreatic amylase content it inhibited the pilocarpine stimulated (2 hours) pancreatic secretion in a dosis-dependent fashion at doses 4, 40 and 400 mg/kg. Vinblastine (200 mg/kg) also delayed amylase accumulation during 3 hours in mice given vinblastine 2 hours after pilocarpine stimualtion. The results support the hypothesis that faultless function and integrity of microtubules might be a prerequisite of pancreatic acinar cell secretionin vivo. Other effects of vinblastine, such as interference with amino acid metabolism, anticholinergic action, and cell injury must also be considered when interpreting the present findings form the point of view of the secretory mechanism of the pancreatic acinar cells.

    Notes: Abstract Objectives. Atherosclerotic lesions result from inflammatory-proliferative responses of the endothelium and smooth muscle of the arterial wall. Poor prognosis of acute myocardial infarction (AMI) patients has been associated with elevated levels of acute phase proteins including C-reactive protein. We investigated the significance of circulating phospholipase A2 in the long-term prognosis of suspected AMI patients. Methods. The concentration of phospholipase A2 was measured by an immunoassay in sera of 100 suspected AMI patients. Admission phospholipase A2 95% fractile outliers were excluded to eliminate the effect of acute infectious diseases. The total and atherosclerotic mortalities were followed for a 4-year period. Results. The most powerful prognostic limit for both admission (p=0.02, RR=2.6 and 95% CI=1.2 to 5.6) and maximal (p=0.06, RR=2.4 and 95% CI=0.96 to 5.9) phospholipase A2 groups was ≥ 8 μg/l. The admission phospholipase A2 level had an independent prognostic significance for atherosclerotic mortaligy (p=0.04, RR=2.4 and CI=1.02 to 5.8) in multivariate analysis with CK-MB and age. Conclusions. The eleated serum phospholipase A2 level at admission is an independent predictor of long-term atherosclerotic mortality in patients with suspected AMI. The prognostic significance of phospholipase A2 weakens during hospitalisation concomitant to the onset of the acute inflammatory response to myocardial injury.

    Notes: Summary Thein vitro secretory response of mouse pancreas to pilocarpine stimulation was studied. A sigmoid-shaped concentration-response relationship was found for amylase release and concentrations of pilocarpine hydrochloride from 10−8 to 10−2 M. Higher concentrations failed to stimulate secretion. The minimal effective concentration was 10−6 M and the maximum response—approximately 3 times that of the control level during 60 min incubation—was obtained at 10−4 to 10−2 M. The secretory rate was linear during this period. The secretory response to pilocarpine was abolished by atropine and was greatly reduced under hypoxic conditions.

    Notes: Abstract The catalytic activity of phospholipase A2 (cat-PLA2) and the concentration of pancreatic (group I) phospholipase A2 (pan-PLA2) and synovial-type (group II) phospholipase A, (syn-PLA2) were studied in 19 human tissues in order to find potential sources of circulating phospholipase A2. Five specimens of each tissue were collected at autopsies or from normal deliveries (placentas and amnionic membranes). The concentrations of pan-PLA2 and syn-PLA2 were measured by specific timeresolved fluoroimmuncassays. The cat-PLA2 was measured by radioactive dipalmitoyl phosphatidylcholine as substrate. The concentration of pan-PLA2 was negligible in all tissue homogenales except the pancreas. Immunoreactive syn-PLA2 was found in the homogenates of the digestive tract, cartilage, and prostatic and parotid glands. By immunohistochemistry, syn-PLA2 was localized in Paneth cell secretory granules, chondrocytes, cartilage: matrix, and glandular cells of prostate.

    Notes: Abstract Immunohistochemical detection of prostate–specific antigen (PSA) is an aid in determining the prostatic origin of metastatic cells. However, small amounts of PSA have also been found in non–prostatic tissues and tumors, for example in some breast carcinomas, by highly sensitive immunofluorometric methods, but also by immunohistochemistry. Our aim was to evaluate the prevalence and prognostic value of histologically confirmed PSA immunoreactivity in breast carcinoma. Sections of formalin–fixed, paraffin–embedded samples from 171 breast carcinomas were immunostained for PSA. The staining results were compared with the mitotic activity, tumor size, histological grade, steroid receptors and follow–up data. For analysis the material was divided into subgroups according to the patients' age (pre- and postmenopausal). PSA was found by immunohistochemistry in 54 (32) breast carcinomas. In survival analysis of the whole patient material PSA positivity did not show prognostic value. Among premenopausal patients concomitant estrogen receptor and PSA–negativity proved to be associated with high risk of breast cancer death (RR 6.2), also after adjustment for tumor size, histological grade, and axillary lymph node status. Among postmenopausal patients PSA positivity was associated with progesterone receptor positivity and high differentiation but not with age, nodal status, or mitotic activity. PSA can be detected by immunohistochemistry in a considerable number of breast carcinomas. PSA immunoreactivity alone does not seem to have any value as general prognosticator of breast carcinoma patients. However, concomitant absence of PSA and estrogen receptors was an indicator of unfavourable prognosis among premenopausal patients.

    Notes: Calcium-deprivation of three weeks duration causes marked degenerative changes in the shell gland mucosa of the laying hen. The height of the columnar epithelium decreases, but the secretory granules in the columnar epithelial cells and the cilia on the apical cells of the columnar epithelium are preserved. The tubular glands decrease in size and number. The microvilli surrounding the tubular gland lumen disappear. The cytoplasm of the tubular gland cell contains autophagic vacuoles (cytolysomes) and large pigment accumulations, presumably lipofuscin. The changes are due to dietary calcium deficiency, apparently mediated by decreased sex steroid secretion and decreased stimulation of the oviduct. These changes are totally reversible, since the fine structure of the shell gland mucosa of calcium-deprived hens returned to a normal laying diet does not show any signs of degeneration and is identical with the fine structure of the shell gland mucosa of the laying control birds.
