    Notes: To evaluate the usefulness of polylactic acid (PLA), a bioabsorbable and plastic polymer, as a carrier of bone morphogenetic protein (BMP), a preliminary study has been carried out to investigate any negative effect of PLA on osteoinduction of demineralized bone (DB). PLA (10,600mol. wt) was mixed with DB particles (1– 1.5 mm square) prepared from rat femurs and laid subcutaneously on the intercostal muscle of 4-week-old Wistar rats. The PLA/DB pellets were harvested at 2, 4, 8 and 24 weeks after the operation, and prepared for light microscopic examination. Histological examination revealed cartilage formation at 2 weeks and new bone formation at 4 weeks. Extensive bone and marrow formation were observed at 24 weeks. PLA was gradually absorbed and completely disappeared at 24 weeks to be replaced by connective tissue. These results demonstrate that PLA does not have a negative influence on the osteoinductive activity and that PLA could well be a promising bioabsorbable carrier of BMP.

    Notes: In order to assess the tissue compatibility of the hybrid materials for the dental implant (hydroxyapatite, titanium oxide and titanium nitride coated titanium alloys), tissue response to these materials implanted in the rat subcutaneous tissue was histologically examined. Initial inflammatory response was less evident in titanium oxide coated and non-coated titanium alloys. All materials were encapsulated by thin fibrous connective tissues. The membrane thickness of hydroxyapatite coated titanium alloy was significantly higher than that of titanium nitride coated one. These results suggest that all materials possess favourable tissue compatibility and may encourage clinical use as the dental implant.

    Notes: To elucidate the Ultrastructure of peri-implant junctional epithelium (IJE) on singlecrystal sapphire dental implants connected to the adjacent teeth by a metal superstructure, the peri-implant gingivae of ten monkeys were examined using a transmission electron microscope (TEM) at 3, 6 and 12 months after implant insertion. At the time of examination, the ultrastructural features of the IJE were almost identical to those of junctional epithelium (JE) attached to natural teeth, i.e. developed Golgi complexes, rough-surfaced endoplasmic reticulum, numerous free ribosomes and mitochondria. The innermost cells of IJE were attached to the implant surface by means of basal lamina-like structures (500–1000 Å in thickness) and hemidesmosomes, but lacked a dental cuticle as seen on teeth. Such epithelial attachment of the IJE was often indistinct or absent at the apical portion of the IJE which terminated at the level of alveolar crest. A continuous basal lamina-like structure was also present along basal cells at the interface between the IJE and gingival connective tissue except at its apical portion.

    Notes: Peri-implant tissues of the single-crystal sapphire implant connected with neighbouring teeth by a metal bridge-work were examined clinically, radiographically, and histologically in ten monkeys. Professional tooth cleaning was performed during the study. At 3–12 months after insertion, most of the implants were firmly connected to the surrounding tissues and peri-implant gingiva was regarded as healthy, based on various periodontal parameter scores. Destructive changes of the peri-implant bone were not found radiographically. Histologically peri-implant gingiva was revealed to show similar structure to that of the gingiva around natural teeth. Direct bone-implant interface was observed at 3 months after insertion, while a thin loose fibrous connective tissue layer was present between bone and implant at 6 and 12 months. Such different interrelationship between bone and implant might be attributable to the difference in distribution of functional stress.

    Notes: The aim of this study was to investigate whether a new connective tissue attachment can occur on a surface of hydroxyapatite (HA) when peri-odontal ligament (PDL)-derived cells with the ability to form new connective tissue attachment are allowed to populate the surface of HA. Six maxillary canines from 3 cats were used for this experiment. Under anesthesia, fenestration of the buccal alveolar bone was performed over the canine root and a cavity was prepared on the root surface. A synthetic HA block was inlaid and cemented into the root cavity. A Teflon membrane was then placed to cover the access opening of the alveolar bone to guide PDL-derived cell proliferation into the surface of the HA block. Finally, the gingival tissue flap was replaced and sutured. Six months after the operation, histologic evaluation was done by serial sections. In 4 of the 6 canines, PDL-derived cells successfully populated the HA surfaces and a layer of cementum-like tissue was formed on the HA blocks. A PDL-like connective tissue layer was seen between the cementum-like layer covering the exposed HA surface and the newly formed alveolar bone sealing the bone fenestration. Collagen fibers in the PDL-like tissue inserted their ends into the newly formed bone and the cementum-like tissue. The present findings demonstrated that PDL-derived cells can 1 form a new connective tissue attachment on HA.

    Notes: A rare case of calcifying epithelial odontogenic tumor (CEOT) devoid of calcification is reported with histochemical, immunohistochemical and electron microscopic studies. The tumor occurred intraosseously in the left maxillary canine and premolar region of a 58-year-old man. The tumor chiefly consisted of scattered small islands of epithelial cells in an abundant fibromyxoid connective tissue stroma. Among the nests, there were many spherical bodies of eosinophilic substance for which non-AA amyloid and non-keratin or basal lamina-like natures were demonstrated histochemically and immunohistochemically. In some nests, there were a few, occasionally several, cells positive for S-100 protein. Ultrastructurally, Langerhans cells with indented nuclei and Birbeck's granules were seen among tumor cells. The prognostic significance of the paucity of calcification and the presence of Langerhans cells in CEOT of which this is only the second description is discussed.

    Notes: Florid cemento-osseous dysplasia (FCOD) with concomitant simple bone cysts is not common. We report a case of FCOD involving three quadrants of the jaws and associated with two large cystic lesions of the mandible in a 40-year-old Japanese woman. Microscopic examination revealed unencapsulated fibroblastic proliferation with formation of bone and cementum, showing different developmental stages, and cystic lesions resembling a simple bone cyst.

    Notes: In 2 parotid adenolymphomas, the composition of lymphoid stroma was analysed, using Stereologic point-counting methods at the electron microscopic level. In the 2 lesions, the stroma was almost identical with respect to organization and composition. Most areas of the stroma showed accumulation of mixed cell populations. On average, the latter were comprised of 5.1 % fibroblastic/relicular cells, 2.7% macro-phages, 68.9% small and 18.8% medium/large lymphocytes, 0.5% blastforming T-lymphocytes, 2.3% blastforming 13-lymphocylcs, and 1.6% plasma cells. Polymorphonuclear leukocytes and mast cells were extremely rare. Areas corresponding to the mantle zone of lymphoid follicles were found to contain 83.0% small and 15.8% medium/large lymphocytes. The present data seem to be in accordance with the speculation that the lymphoid stroma of adenolymphoma represents an immune reaction.

    Notes: Topographically, the AOT occurs in peripheral and central variants, the latter further in follicular (with embedded tooth) and extrafollicular (no embedded tooth) types. The AOT is slow growing with few or no symptoms. Tumor growth may cause displacement of teeth rather than root resorption. The follicular AOT mimics a follicular cyst, the extrafollicular a residual or “globulo-maxillary” cyst and the peripheral a gingival fibroma. All variants of AOT show identical histologic features. The central variants account for 97.2%, 73.0% of which are follicular. The follicular variant (M:F ratio 1 to 1.9) is three times as frequent as the extrafollicular. The follicular variant is diagnosed earlier in life (mean age 17 yr) than the extrafollicular (mean age 24 yr). 53.1% of all variants occur within the teens (13–19 yr). Follicular AOT is associated with one embedded tooth in 93.2%. Maxillary permanent canines account for 41.7% and all four canines for 60.1% of AOT-associated embedded teeth. Ranking four among the odontogenic tumors the AOT is not a particularly rare tumor. Conservative surgical excision is the treatment of choice. Documented recurrences have not been reported.

    Notes: Abstract  Desmoplastic ameloblastoma (DA) is an unusual subtype of ameloblastoma histologically characterized by the pronounced collagenized stroma. In the present study, the immunolocalization of transforming growth factor beta (TGF-β), one of the most potent local factors for modulating extracellular matrix formation, was observed in DA in order to study its participation in the stromal desmoplasia. Seven cases of DA, including a ”hybrid” lesion, were studied together with ten cases of ordinary follicular and plexiform ameloblastomas as the control. In contrast to ordinary ameloblastomas, marked immunoexpression was observed in all DAs but one. In the ”hybrid” lesion, TGF-β was not expressed in the area of follicular ameloblastoma but in that of DA. These results show that TGF-β produced by tumor cells of DA plays a part in the desmoplastic matrix formation.
