    Notes: Summary In vivo treatment of the jejunal mucosa with glycosidic enzymes seems to remove the enteric surface coat of the enterocyte. As a consequence, the mucosa-to-serosa unidirectional flux of acetamide increases remarkably. The glycocalyx probably represents a barrier to the diffusion of small hydrosoluble solutes.

    Notes: Abstract A Cl/HCO3 exchanger mediates HCO3 extrusion across rat jejunal basolateral membrane. Previous studies demonstrated that anion antiport activity is positively affected by Na, but evidence was given that this cation is not translocated by the carrier protein. Basolateral membranes isolated from rat jejunum were used to give more insight on Na effect. Uptake studies, performed together with vesicle sidedness determinations, indicated that the greatest stimulation of Cl-dependent HCO3 uptake occurs when Na is present at both vesicle surfaces. The kinetic dependence of Cl/HCO3 exchange on equal intra- and extravesicular Na concentration showed a hyperbolic relationship, and the calculated kinetic parameters were V max=0.153 ± 0.006 nmol mg protein-1 sec-1, K m =23.0 Mm. Ion replacement studies indicated that Na can be partially substituted only by Li and not by other monovalent cations. Results of this study suggest that Na could act as a nonessential activator of the Cl/HCO3 exchanger. A possible role of the Na-sensitive modifier site in the physiology of jejunal enterocyte is suggested.

    Notes: Summary The fine structure of the plastids in the leaf's basal meristem and in the leaf tissues 2 cm immediately above has been studied in maize leaves of different ages. In the young leaves the proplastids of the basal meristem differentiate, in the tissues within 2 cm above the meristem, into chloroplasts containing two or more prolamellar bodies, indipendently whether the tissues have been fixed 11 hours after a period of illumination or of darkness. In the oldest leaves, in the tissues immediately above the basal meristem, no prolamellar body is present in the plastids, and the proplastids differentiate directly into chloroplasts, without passing through an “etio-chloroplast” stage.

    Notes: Abstract The self-orienting Percoll-gradient centrifugation was used to separate basolateral membranes (BLMs) from rat jejunal enterocytes. Bicarbonate uptake into osmotically active BLM vesicles was studied by a rapid filtration technique. The time course of [14C]-labelled bicarbonate uptake was followed for 30 min at 18° C and at pH 8.2. Bicarbonate uptake was fast, not saturable and stimulated by countertransport. This work demonstrates that it is possible to determine bicarbonate flux across BLM vesicles at pH and temperature values close to usual experimental conditions. Together with simple diffusion, bicarbonate would cross the BLM of the enterocyte via a carrier-mediated diffusion process.

    Notes: Abstract The mechanisms of HCO 3 − and Cl− transport across basolateral membranes from rat ileum were investigated in isolated vesicles by means of uptake experiments. Neither Cl−/HCO 3 − exchanger nor Na+−(HCO 3 − )n cotransport seem to be present in ileal basolateral membranes. Moreover Cl− uptake is unaffected bycis Na+ and/or K+ gradients, indicating the absence of Na+−Cl−, K+−Cl− and Na+−K+−2Cl− symport activity. An electrically conductive pathway seems to be responsible for both HCO 3 − and Cl− fluxes. Evidence is also given for the presence of a Na+/H+ exchanger at the basolateral pole of ileal enterocytes.

    Notes: Abstract Enterocytes isolated from rat jejunum were tested for the existence of a Cl−/HCO 3 − exchange, previously evidenced in basolateral membrane vesicles but not in brush border. Cells were found to retain functional integrity and transport capabilities long enough to allow Cl− fluxes to be measured. Both efflux and uptake experiments indicate that a Cl−/HCO 3 − antiport, inhibited by 4,4′-diisothiocyanostilbene-2-2′-disulfonic acid (DIDS), is functional under resting conditions.

    Notes: Summary Chloroplast differentiation in relation to increasing leaf age has been investigated in maize plants exposed to continuous illumination. In the young leaves the proplastids differentiate into chloroplasts containing well organized grana as well as prolamellar bodies. In the older leaves, while plastids differentiate, the prolamellar bodies are no longer detectable. Chloroplast ability to build up prolamellar bodies does not seems so much a light dependent process as it is affected by cell differentiation rate.

    Notes: Summary Ultrastructural surveys, carried out into an olive necrotic maize mutant (Neuffer E 283 B) grown under a 16 hours photoperiod, have shown remarkable morphological alterations in the plastids. Such alterations, affecting both the mesophyll and the bundle sheath plastids, appear to be photodependent. Quite normal etioplasts are present in dark-grown mutant seedlings. Moreover, light appears also to inhibit the overall growth of mutant plants. 12-day-old mutant plants grown under illumination are 4 cm in comparison with the 10 cm of the wild type, while corresponding dark-grown seedlings of both types are 12 cm high.
