    Notes: The study was undertaken to develop a suitable design to accommodate liquefying hygroscopic samples in the Proximity Equilibrium Cell (PEC) for equilibration to desired water activity. The system suggested has overcome the limitation of the earlier reported methods using PEC and is found to be rapid compared to other gravimetric methods used for hygroscopic materials.

    Notes: Methylobacterium sp. ZP24 is able to produce poly-3-hydroxybutyrate (PHB) from sucrose and lactose. As the production of PHB is growth-associated, a strategy of intermittent feeding of sugars and other nutrients was assessed for obtaining high yields of the polymer. Higher PHB synthesis was obtained at increased sugar feed rates. Cellular PHB contents of 63% and 71%, with productivities of up to 0.354 and 0.645 g PHB/l h were obtained from sucrose and lactose, respectively. A short-duration semicontinuous production level of up to 2.4 g PHB/l h was achieved in the lactose fermentation. Journal of Industrial Microbiology & Biotechnology (2000) 25, 276–279.

    Notes: Abstract Penicillin acylase from Bacillus megaterium was purified 6.22 fold using ultrafiltration followed by ammonium sulphate precipitation and gel filtration chromatography using Sephadex G-100. The final specific activity was 2.62 U/mg protein and was free from interfering proteases.

    Notes: Abstract Butyl acetate, isoamyl acetate and isoamyl valerate were prepared by Mucor miehei lipase catalyzed esterification of free acids and alcohols carried out in non-aqueous systems using heptane and silica gel which removes water formed in the reaction. For butyl and isoamyl acetate 1:3 and for isoamyl acetate 1:2 molar proportions of acid to alcohol were found to be optimal. Heptane(5 ml) and 0.01g silica gel per 0.1M acid were found to improve the yields. Under optimum conditions using 60°C, within 48 hours 40% butyl acetate, 53% isoamyl acetate and 61% isoamyl valerate conversions were observed.

    Notes: Summary Escherichia coli cells with penicillin acylase activity were permeabilized with aqueous solutions of the cationic detergent N-cetyl-N,N,N-trimethylammonium bromide (CTAB), at pH 8.0 and the activity was found to have almost doubled. The concentration of CTAB, the time and temperature of treatment were optimised for maximum enzyme activity and were found to be 0.2%, 20 min and 5°C respectively. Subsequently, the cell bound activity was retained for a longer period by chemical cross-linking with 0.1% glutaraldehyde.

    Notes: Summary Optimisation of reaction conditions for the esterification of geraniol of palmarosa oil with n-butyric acid using immobilized lipase from Mucor miehei in non-aqueous system was carried out. Palmarosa oil could be easily esterified upto 95% w/w at 40°C in 24 h. Effect of moisture content was studied using Na2SO4 and recycling of the immobilized enzyme.
