    Notes: Despite several studies showing that the rat supraoptic (SON) and paraventricular (PVN) nuclei are innervated by noradrenergic afferents, the respective contribution of these inputs to the oxytocinergic and vasopressinergic neuronal populations remains to be clearly defined. In the present study, we used the unbiased disector method to estimate the numerical density of noradrenergic varicosities on identified oxytocinergic and vasopressinergic somata in the rat SON and PVN. The analysis was carried out on semithin (1 μm) plastic sections cut from vibratome slices (50 μm) of the SON and PVN which had been double-labelled for noradrenaline (NA) and oxytocin- or vasopressin-related neurophysin. These preparations displayed many noradrenergic varicosities which electron microscopy showed to represent, in the main, synaptic boutons. Our quantitative analysis revealed that noradrenergic varicosities contacted oxytocinergic and vasopressinergic somata to a similar extent in male and female rats, under basal conditions of hormone secretion. The incidence of these axo-somatic contacts was similar in the SON and PVN. In contrast, in lactating rats, in which oxytocin secretion is enhanced, there was a significant increase in the density of noradrenergic varicosities apposed to oxytocinergic somata, in both nuclei. Our observations indicate that, in male and female rats under normal conditions, noradrenergic afferents innervate each type of neurosecretory somata, in both magnocellular nuclei, in a similar fashion. They reveal, moreover, that noradrenergic afferents participate in lactation-induced structural plasticity of synapses impinging on oxytocinergic somata.

    Notes: Abstract Light microscopic analysis of the rat midbrain periaqueductal grey (PAG) showed vasoactive intestinal polypeptide immunoreactive (VIP-ir) neurons localized at the lateral and ventral walls of the aqueduct. Some varicose VIP-ir elements were detected closely associated with the ependyma. While several VIP-ir elements were encountered immediately under the ependyma, in a few cases, VIP-ir cell bodies were seen on the luminal surface of the ependymal cells lining the aqueduct. Electron microscopy revealed that most of these cells possessed the characteristics of a local circuit neuron. All VIP-ir cells had indented nuclei. Two types were distinguished: one with rounded cell body receiving numerous axo-somatic synapses established by VIP-negative axons. The other cell type was fusiform and its surface was almost fully isolated from axonal contacts by a glial sheath. The VIP-ir processes were interconnected with other periaqueductal cells by a variety of synaptic contacts. VIP-ir axon terminals formed asymmetric synapses with immunonegative dendritic shafts often in glomerulus-like assemblies. The postsynaptic immunonegative dendrites were of the aspinous, beaded type. We suggest that VIP-ir cells and processes in the midbrain PAG establish connections between the longitudinal functional columns of this region. On the basis of their morphology, VIP-ir cells in the PAG appear to be excitatory, terminating on inhibitory interneurons. Thus, a VIP-stimulated inhibition may be instrumental in the coordination of responses evoked by the stimulation of PAG columns.

    Description / Table of Contents: Rearrangements of 2,3-Diazabicyclo[2.2.2]octane DerivativesThe ketones 4a - e have been synthesized and their reactions with methylmagnesium iodide were studied. A mechanism is given for the rearrangement of some magnesium alkoxides to 6,7-diazabicyclo[3.2.1]octane derivatives. - Dehydration of the alcohols 12d, 14a; 17a, b, g, and h with trichlorophosphane oxide is accompanied by migration of the group antiperiplanar to the hydroxy function, and thus 6,7-diazabicyclo[3.2.1]octane derivatives are formed. - Ketones 4a, c and e give on sensitized irradiation the tricyclooctane derivatives 40a, b and d, respectively, while direct irradiation leads to the bicycloheptane derivatives 42a, b and d, respectively.
    Notes: Die Darstellung der Ketone 4a - e sowie deren Umsetzung mit Methylmagnesiumiodid werden beschrieben. Für die bei einigen dieser Grignard-Additionen auftretende Umlagerung zum 6,7-Diazabicyclo[3.2.1]octan-System wird ein Mechanismus aufgestellt. - Die Dehydratisierung der Alkohole 12d, 14a, 17a, b, g und h mit Trichlorphosphanoxid erfolgt unter stereokontrollierter Umlagerung zum Bicyclo[3.2.1]octan-System mit Wanderung der zur Hydroxyfunktion antiperiplanaren Gruppierung. - Je nach Belichtungsbedingungen erhält man bei der Photoisomerisierung von 4a, c und e die Tricyclooctan-Derivate 40a, b bzw. d oder die Bicycloheptan-Derivate 42a, b bzw. d.
