    Notes: Abstract SC-41930, 7-[3-(4-acetyl-3-methoxy-2-propylphenoxy)propoxy]-3,4-dihydro-8-propyl-2H-1-benzopyran-2-carboxylic acid, is a selective, orally active, LTB4 receptor antagonist currently in clinical trials for psoriasis and ulcerative colitis. Exhaustive SAR studies found a potential backup compound, SC-50605, which was 7–16 times more potent than SC-41930 in LTB4 receptor binding, chemotaxis and degranulation assays. SC-50605 also inhibited LTB4-induced intradermal chemotaxis in cavine skin at an oral dose of 0.10 mg/kg and displayed good activity in animal models of colitis and epidermal inflammation both orally and topically.

    Notes: Summary In this immunocytochemical study, the brains of nine squirrel monkeys (Saimiri sciureus), ranging from 8 to 27 years of age, were examined for the presence and distribution of β/A4 amyloid, a 4-kilodalton peptide. In aged squirrel monkeys, amyloid is associated primarily with intracerebral and meningeal capillaries and arterioles and occurs to a lesser degree as small and/or diffuse deposits in the neural parenchyma and in the dense cores of senile plaques. Cerebrovascular amyloid is found primarily in neocortex, amygdala, and septum verum and is rare or nonexistent in other subcortical gray structures, white matter, cerebellum, and spinal cord; this pattern of localization is comparable to that in humans with cerebral amyloid angiopathy. There is a significant correlation between cerebrovascular and parenchymal deposits of amyloid. However, cerebrovascular amyloid is always the most abundant form in squirrel monkeys, even in cases of severe cerebral amyloidosis. In contrast to squirrel monkeys, aged rhesus monkeys (Macaca mulatta) develop, mostly parenchymal deposits of amyloid and have relatively less vascular amyloid. This species difference in the histological distribution of amyloid suggests that separate mechanisms may influence the accumulation of amyloid in cerebral blood vessels and in the neural parenchyma. These data also indicate that the squirrel monkey can serve as a model for investigations of cerebrovascular amyloidosis.

    Notes: Summary Patients with Alzheimer's disease (AD) and some aged controls may have diminished functions of the pineal gland. In this immunocytochemical study, we stained pineal glands from cases of AD and young and aged controls for cytoskeletal elements and amyloid. We found no evidence of neurofibrillary tangles (NFT) or the accumulation of neurofilaments, tau, A68, or β/A4 amyloid deposition in pinealocytes or associated structures in cases of AD or controls. In both AD and controls, we observed dense immunoreactivity for phosphorylated neurofilaments in marginal plexuses associated with processes of pinealocytes, boutons, and knob-like endings. The accumulation of phosphorylated neurofilaments in the processes of pinealocytes appears to be a normal morphological characteristic of the pineal gland and may not represent a pathological change.

    Notes: Abstract The photosynthetic fluorescence ratio Fv:Fm, in vivo absorption spectra and ion leakage were evaluated as biomarkers of ambient and elevated UV-B (280 to 320 nm) exposure of the intertidal alga Enteromorpha intestinalis (Chlorophyta) and the sublittoral alga Palmaria palmata (Rhodophyta). Measurements of thallus growth were also used to assess adverse biological effects. Ambient and elevated UV-B significantly inhibited photosynthesis in both species. It was shown that the Fv:Fm ratio is a sensitive, non-specific general biomarker of UV-B exposure in both species. Moreover, the in vivo absorption of what was tentatively identified as chlorophylls a and b as well as phycoerythrin and/or carotenoids, phycoerythrobilin and phycocyanin decreased in a dose-response dependent manner and was associated with a decrease in growth rate in P. palmata. The intertidal alga E. intestinalis showed a greater degree of tolerance to UV-B exposure. These results indicate that changes in the Fv:Fm ratio together with reductions in in vivo pigment absorption could provide an early quantitative warning of the detrimental effects of UV-B in marine macroalgae.

    Notes: Summary Injections of horseradish peroxidase conjugated with wheat germ agglutinin (WGA-HRP) were made into the cortex of kittens. When cytochrome oxidase histochemistry was performed on sections taken through the injection sites staining of the WGA-HRP deposits occurred, demonstrating that WGA-HRP can cross-react during processing for cytochrome oxidase.

    Notes: Summary The laminar pattern of cytochrome oxidase activity was studied in visual cortical areas 17,18 and 19 in adult cats and kittens, following electrophysiological determination of the boundaries of these areas in all but the youngest animals. The patterns of cytochrome oxidase staining and the cytoarchitectonic appearances of areas 17, 18 and 19 were compared. At all ages activity was especially high in the region of layers IV and VI in areas 17 and 18, and was low in all laminae in area 19. The results suggest that the degree of cytochrome oxidase activity in these regions of the visual cortex may be related to the strength and type of projection that they receive from the lateral geniculate nucleus. The cytochrome oxidase technique is a useful means of defining the 18/19 border, and may help locate the boundary between areas 17 and 18, in both adult cats and kittens.

    Notes: Summary We used quantitative electrophysiological techniques to study the spatial properties of single units recorded extracellularly in the lateral geniculate and perigeniculate nuclei of the adult pigmented ferret. All neurones examined had approximately circular receptive fields, whose central regions gave responses antagonistic to those elicited from the surrounds. We presented sinusoidally modulated grating patterns, either drifting or counterphased, to obtain spatial frequency tuning curves, contrast sensitivity functions and to assess linearity or non-linearity of each neurone's response. In the ferret, as in other species, two types of lateral geniculate neurone could be distinguished, and we have termed these X-cells and Y-cells; both groups responded briskly to visual stimulation but X-cells gave sustained and linear responses whereas Y-cells responded transiently and non-linearly. Perigeniculate cells gave linear responses. For neurones in the lateral geniculate and perigeniculate nuclei, both the limit of spatial resolution (‘acuity’) and optimum spatial frequency were inversely related to receptive field eccentricity and the diameter of the receptive field centre. We recorded geniculate neurones in the ferret with acuities up to 3 cycles deg-1 and contrast sensitivities up to 114, values that are lower than those found previously for many geniculate cells in the cat.

    Notes: Abstract Nineteen (12 male, 7 female) children, who have received craniospinal irradiation for the treatment of a brain tumour distant from the hypothalamic-pituitary axis, resulting in growth hormone (GH) deficiency (CS-PRGHD), have been treated with GH. Eight have completed growth. Comparison has been made with the growth of seven untreated children, whose heights and growth rates at presentation were normal despite GH deficiency secondary to irradiation. GH produced a significant increase in growth velocity over the first 3 years' treatment in CS-PRGHD patients with a mean first year increment of 3 cm/year. Patients, treated to completion of growth, showed a significant increase in leg length standard deviation (SD) score (ΔSDS+0.2) compared to that of the untreated (ΔSDS−0.9) (P〈0.05). Stitting height SD scores decreased irrespective of GH therapy (by -1.7 for the treated and -2.2 for the untreated). The onset of puberty in the irradiated patients occurred at a mean bone age of 10.7 years in males and 9.9 years in females. This limited the time available for GH therapy. These factors resulted in a decrease in standing height SDS of 0.9 at completion of GH therapy in CS-PRGHD, but a decrease of 1.7 in those not treated with GH. Thus GH therapy failed to induce “catch-up” growth in irradiated patients, but it did prevent further loss of adult stature, with a mean final height SD score of -3.4 in CS-PRGHD patients.
