    Description / Table of Contents: Summary Electron microscopic studies of particles of the human fundic mucosa aspirated by suction biopsy suggest a functional cycle, especially in the cymogenic cells. When gastric secretion has been stimulated by pentagastrin an accumulation of lysosomes, i.e. granules, and their expulsion into the lumen of the gastric glands has been observed, as well as a disappearance of the endoplasmatic reticulum. The secretory phase has been followed by a phase of regeneration of the endoplasmatic reticular structures. 5 1/2 hours after a secretory stimulus the ultrastructure of the fundic mucosa has been identical with the state of secretory rest observed at the beginning of the experiment. The establishment of phasic functional changes in the ultrastructure of the gastric mucosa with a definite time course may be important for further studies of gastritis and of the mode of action of anti-inflammatory drugs.
    Notes: Zusammenfassung Elektronenmikroskopische Untersuchungen an bioptisch gewonnenen Partikeln der Korpusschleimhaut lassen einen Funktionscyclus vor allem an den Hauptzellen erkennen. Nach Stimulation der Magensekretion mit Pentagastrin kommt es zur Häufung von Lysosomen und ihrer Ausstoßung in das Lumen sowie zu einem Verlust des endoplasmatischen Reticulums. Dieser Sekretionsphase folgt eine Regenerationsphase mit Wiederaufbau des endoplasmatischen Reticulums. Nach 5 1/2 Std ist der Zustand der sekretorischen Ruhephase wieder erreicht. Die ultrastrukturellen Veränderungen der Magenschleimhaut in der Sekretionsphase ähneln dem nach Betazol-Injektion beschriebenen Bild, sind jedoch schwächer ausgeprägt. Die Festlegung des zeitlichen Ablaufs der Ultrastrukturveränderungen ist für weitere Untersuchungen über die Wirkung entzündungshemmender Pharmaka und auch für Funktionsstudien bei der Gastritis bedeutsam.

    Notes: Zusammenfassung Direktbeobachtungen der Papilla duodeni zeigen die enge funktionelle Kopplung der Darmperistaltik mit der Entleerungsmechanik der Papille. Druckmessungen des Sphincter Oddi im Durchströmmungsversuch lassen die Schwankungen im Schließungsdruck dieses Muskels erkennen. Insbesondere zeigt sich deutlich die Tonusabnahme nach Ätheroder Magnesiumsulfatauftropfung. Acetylcholingabe führt hingegen zu einem Spasmus im Papillengebiet. Ebenso kommt es am sensibilisierten Tier nach Wiederberührung mit dem Allergen zu einem Sphincterkrampf. Die Bedeutung dieser Feststellung für die Genese allergisch bedingter Gallenspasmen mit Koliken findet hierin ihre Erklärung.

    Notes: Zusammenfassung Durch direkte Bestimmung der in das Duodenum abgesonderten Fermentmenge bei einer jugendlichen und einer alten Personengruppe zwischen 10 und 25 bzw. 57 und 70 Jahren, ließ sich feststellen, daß erst im Doppelbelastungsversuch der Nachweis eines latenten Lipasemangels im Alter erbracht werden konnte. Auch für die Eiweißresorption deutet sich eine leichtere Erschöpfbarkeit der Trypsinausscheidung an. Die Diastaseabgabe jugendlicher und älterer Personen weist keine Unterschiede auf.

    Notes: Summary To detect structural alterations in human oocytes that may give rise to predisposition to aneuploidy, unfertilized human oocytes from an IVF programme were processed for indirect anti-tubulin immunofluorescence. The spindle of oocytes aged for 2 days is rather small, and bi- or multipolar. Chromosomes are no longer aligned at the spindle equator but are scattered all over the degenerating spindle. This implies that human oocytes aged for 2 days may no longer be able to develop into a chromosomally balanced, normal embryo. In oocytes aged for 3–4 days the chromosomes become more decondensed and form a restitution nucleus. Microtubules radiate out from the latter towards the cell periphery and form a network of fibres in the cytoplasm. A similar alignment of tubules is found in unfertilized, activated oocytes. Oocytes with an aberrant cytoskeleton and chromosomal array were predominantly obtained from aged females. They include two binucleated oocytes with two sets of chromosomes and two oocytes with displaced chromosomes one of which had a tripolar spindle.

    Notes: Abstract.  Consumption of trichlorfon-poisoned fish by women in a small Hungarian village has been associated with trisomy resulting from an error of meiosis II in oogenesis. We therefore examined mouse oocytes exposed for 3 h during fertilization to 50 µg/ml trichlorfon. Spindle morphology was not visibly altered by the pesticide. Chromosomes segregated normally at anaphase II with no induction of aneuploidy. However, formation of a spindle was disturbed in many oocytes resuming meiosis I in the presence of trichlorfon. In spite of the spindle aberrations and the failure of bivalents to align properly at the equator, oocytes did not become meiotically arrested but progressed to metaphase II. At this stage, spindles were highly abnormal, and chromosomes were often totally unaligned, unattached or dispersed on the elongated and disorganized spindle. By causing spindle aberrations and influencing chromosome congression, trichlorfon appears, therefore, to predispose mammalian oocytes to random chromosome segregation, especially when they undergo a first division and develop to metaphase II during exposure. This is the first case in which environmentally induced human trisomy can be correlated with spindle aberrations induced by chemical exposure. Our observations suggest that oocytes may not possess a checkpoint sensing displacement of chromosomes from the equator at meiosis I and may therefore be prone to nondisjunction.

    Description / Table of Contents: Zusammenfassung Anhand einer Gesamtstudie an 120 nach Billroth II-Resezierten wurden bei einer Gruppe von 45 Fällen in ca. der Hälfte (21) Störungen der Fettresorption, überprüft am Vitamin A-Stoffwechsel, gefunden. Erstaunlich war die Gewichtskonstanz, denn nur 12 von 75 in der Zeit von 1952–65 Operierte waren leicht (bis 4 kg = 9) oder stärker (bis zu 12 kg = 3) untergewichtig. Die Xyloseabsorption war jedoch reduziert. Mit diffizilen Methoden waren mehr oder weniger larvierte Resorptionsinsuffizienzen festzustellen.
    Notes: Summary Among 120 patients with Billroth II resections, half the number (21) of a group of 45 had disorders of fat absorption. This was checked in the vitamin A metabolism. The constancy of weight was surprising: only 12 out of 75 patients operated during 1952–65 were below weight, either slightly (up to 4 kg = 9) or to a greater extent (up to 12 kg = 3). Xylose absorption was, however, reduced. By the use of highly sensitive methods some masked failures of absorption were found.
