    Notes: Abstract. Each kringle of human plasminogen (HPg) except kringle 3 (K3) exhibits affinity for ω-aminocarboxylic acids. Assuming that the K3 domain contains a preformed but nonfunctional lysine binding site (LBS), Lys311 was altered by site-directed mutagenesis into Asp311 in accordance with the consensus sequence of the LBS. Cys297 involved in the interkringle disulfide bridge was mutated into Ser297 to minimize dimerization and aggregation. The mutated K3 TYQ[K3HPg/C297S/ K311D]DS (r-K3mut) was expressed in Escherichia coli, isolated on an Ni2+-nitrilotriacetic acid-agarose column, refolded and purified on a lysine Bio-Gel column. Fluorescence titration indicates affinity of r-K3mut for ω-aminocarboxylic acids with the following association constants (Kass, mM−1) 5-aminopentanoic acid 1.3; 6-aminohexanoic acid 4.2; 7-aminoheptanoic acid 0.5; trans-(aminomethyl)cyclohexanecarboxylic acid 12.7; p-benzylaminesulfonic acid 11.8. r-K3mut exhibits an affinity similar to native and mutated (R220G, E221D) K2. The results indicate the presence of a preformed but nonfunctional LBS in native K3 of HPg. We were able to demonstrate for the first time that an appropriate mutation in the LBS of a kringle produced a weak but distinct affinity for ω-aminocarboxylic acids.

    Description / Table of Contents: Summary A 58 year old patient suffering from psoriasis vulgaris had typical erythematous, scaly plaques for 20 years. In addition, an erosive, therapy-resistant plaque was found in the perianal region. A biopsy was taken to exclude malignancy; it showed intracytoplasmic Michaelis-Gutmann-bodies as a marker for malakoplakia, which rarely occurs in the skin. In further electron microscopic investigation, intracytoplasmatic bacteria could be demonstrated. An underlying malignancy or chronic renal insufficiency with immunosuppression were not found in this patient; both are often associated with malakaplakia.
    Notes: Zusammenfassung Es wird über einen 58jährigen Patienten berichtet, bei dem seit 20 Jahren rezidivierend eine Psoriasis vulgaris mit typischen erythemato-squamösen Plaques an den Prädilektionsstellen besteht. Desweiteren fand sich perianal ein seit ca. 5 Jahren bekannter, therapieresistenter und teils erodierter Plaque. Die zum Ausschluß eines malignen Prozesses durchgeführte Probebiopsie ergab die seltene kutane Manifestation einer Malakoplakie durch den Nachweis der typischen intrazytoplasmatischen Michaelis-Gutmann-Körperchen sowie durch elektronenmikroskopisch nachweisbare intrazytoplasmatische Bakterien. Ein Tumorleiden oder eine Niereninsuffizienz mit entsprechender Immunsuppression, als häufig beobachtete Grunderkrankung der ätiopathogenetisch bisher nicht geklärten Malakoplakie, konnten bei dem hier vorgestellten Fall ausgeschlossen werden.

    Notes: In a side-by-side comparison, the q dependence of the collective internal decay constant ∼(ΔΓ)∼qγ of linear polystyrene in good and theta solvents was determined using photon correlation spectroscopy. The results q2.9 and q3.9 for the theta and good solvents, respectively, correspond to relatively strong hydrodynamic interactions (nondraining) when excluded volume effects are minimal for the compact coil at theta and to relatively weak hydrodynamic interactions (partial draining) when excluded volume effects are manifest in the swollen coil in good solvent.

    Notes: Background Increased serum-IgE levels in patients with bullous pemphigoid (BP) are one of the well known immunological deviations in this autoimmune dermatosis.Aim and methods Since the mechanism that leads to an increase of IgE in BP is still unclear, we investigated serum-IgE levels and IgE regulating cytokines (soluble low affinity Fc II receptor for IgE, IL-4, IFN-g and soluble IL-2 receptor) in sera and blister fluids of 15 BP patients before and during treatment with immunosuppressants using an ELISA-technique.Results All 15 patients examined proved to have high IgE levels in blister fluids (〉 100 KU/l) and 13 of the 15 patients had elevated serum-IgE levels. Soluble low affinity Fc II receptor for IgE (sCD23R) was also increased in sera (7.9 ± 2.4 pg/ml) and ten fold higher in blister fluids (76.7 ± 15.3 pg/ml). Investigation of cytokines interfering with IgE production showed significantly increased levels of the soluble IL-2 receptor in sera whereas the concentrations of IL-4 and IFN-g revealed no significant differences compared to controls. In blister fluids we detected all the cytokines investigated. During treatment with immunosuppressants the elevated concentrations of sCD23R and soluble IL-2 receptor were reduced to normal values.Conclusions The results show a direct correlation between IgE levels, sCD23R and soluble IL-2-receptor concentrations and suggest a connection between increased IgE levels and T-cell activation in BP.
