    Notes: Summary 35 specimens of human parotid gland and 37 of submandibular gland were transplanted into athymic nude mice. At distinct time intervals, from 1 day to 8 months the transplants were collected and examined. The transplanted glands were studied by light microscopy, immunohistology and autoradiography. The following changes were detectable: 1. acute injury to the xenograft and inflammatory reaction (day 1–7), 2. regeneration of the transplant and the beginning of adaptation to the “mouse milieu” (day 8–30), 3. completion of adaptation (day 30 and later). The presence of the following substances was analysed: amylase, lactoferrin, secretory component, tissue polypeptide antigen (TPA). Amylase was only detected in the early transplants. Lactoferrin was seen only in the small duct system. TPA was present during all transplantation periods and was quantitatively correlated with the3H thymidine labeling index. From our observations we can say that the salivary glands show two different reacting compartments: a large and a small duct system. The histogenesis of the xenografts, and the relationships of the changes observed to human salivary gland diseases were discussed.

    Notes: Summary 10 pleomorphic adenomas of the human parotid gland were transplanted on several groups of nude mice. For comparative reasons, 10 other pleomorphic adenomas, a neurinoma and a chordoma and transplants of squamous cell carcinomas and of normal salivary gland tissue were also analysed. In the primary tumours and in the transplants, the presence of keratin, carcinoembryonic antigen, tissue polypeptide antigen, lactoferrin, lysozyme, immunoglobulins, secretory component, amylase, fibronectin and of several lectin-receptors (PNA, WGA, HPA, Ulex europaeus) was sought. The immunohistological observations show that many of the features of a pleomorphic adenoma are constant under the conditions of transplantation. In the transplanted tumour, the same heterogeneity as in the primary tumours can be observed. Autoradiographic studies show little labelling with 3-H thymidine, which is in good accordance with the biological behaviour of the tumour. The distribution of fibronectin shows an interesting association with myoepithelial-like cells. Our results support the hypothesis that the histogenetic origin of the pleomorphic adenoma is a cell pool of the terminal ductal segment. A differentiation towards ductal cells (with production of secretory substances) and towards myoepithelial cells (associated with large amounts of basal membrane like substances) is observed.

    Notes: Summary The migratory and proliferative capacity of oral epithelium was studied after induction of subepithelial suction blisters in palatal rat mucosa. FITC-coupled lectins and their affinities to epithelial cells at the wound margin were analyzed by immunfluorescence microscopy. Immunocytochemical distribution of 67 K-keratin polypeptides in different epithelial layers were studied using specific antisera against keratin proteins. By means of3H-thymidine-autoradiography the proliferation of epithelial basal cells and their migration at the wound edge was quantified by evaluation of the labelling index. Double labelling was performed, combining the autoradiographic and the keratin staining technique. It was found that the subepithelial suction blister showed rapid repair (epithelial regeneration after 3–4 days). Peanut Agglutinin (PNA) showed a selective affinity to the basal 2–3 layers of oral rat mucosa. No modifications of lectin affinities were found in epithelial wound repair. PNA-positive keratinocytes were demonstrated in the migratory epithelial tongue. The selective presence of 67 K-keratin (67 K) in the suprabasal epithelial cell layers was interpreted as an indication of the differentiation process in rat oral mucosa. The3H-thymidine-labelling index of basal cells increased significantly, beginning at the wound edge after 24 h. The peak of the labelling index curve was located in the vicinity of the wound after 96 h. Double labelling technique revealed 67 K-negative and3H-thymidine-positive basal cells as well as 67 K-positive and3H-thymidine-negative cells at the migration front. These observations support findings that migrating epithelial cells in oral wound healing derive both from undifferentiated (str. basale) and from differentiated (inferior str. spinosum) cell compartments.

    Description / Table of Contents: Zusammenfassung Resorbierbare Kunststoffnetze werden erfolgreich bei der Versorgung von Rupturen parenchymatöser Organe verwendet. Eine experimentelle Studie am Kolon der Ratte sollte klären, inwieweit eine Sicherung nahtinsuffizienz gefährdeter Anastomosen hiermit ebenfalls möglich sei. Bei 75 Wistar Ratten wurde das Querkolon durchtrennt und reanastomosiert, wobei bei der Hälfte der Tiere die Anastomose potentiell insuffizient angelegt wurde. In beiden Gruppen wurde bei einigen Tieren die Anostomose mit einem Polyglykolsäure-Filament-Netz (PGS-Netz) umhüllt. Die Ergebnisse zeigten, daß bei allen Versuchstieren deren Kolonanastomose mit PGS-Netz umhüllt war, postoperative Motilitätsstörungen sowie eine verzögerte Anastomosenheilung zum Teil mit spontanen Dehiszenzen der Anastomose und Ausbildung einer Peritonitis auftraten. Die Festigkeit der Anastomose wurde durch Messung der Berstungsdrucke am 2., 4., 8., 30., 60. und 90. postoperativen Tag bestimmt. Hierbei zeigte sich, daß der Berstungsdruck bei suffizient und insuffizienter Anastomosenanlage ohne PGS-Netz-Umhüllung deutlich über den Werten der Versuchstiere mit PGS-Netz lag. Hieraus folgt, daß eine Umhullung mit PGS-Netz keine zusätz-liche Anastomosensicherung darstellen kann. Vielmehr scheint der fehlende peritoneale und omentale Kontakt für eine verzögerte Heilung verantwortlich zu sein.
    Notes: Summary Resorbable polyglycohc-acid (PGA)-nets are used successfully for the treatment of traumatic ruptures of parenchymous organs. This experimental study was carried out to investigate whether PGA-nets can also reinforce jeopardized colon anastomoses. In 75 Wistar rats the transverse colon was transsected and reanastomosed. Animals were divided into 2 groups: in one group the anastomosis was applied sufficiently, in the other group the anastomosis was applied potentielly insufficient. Out of each group a PGA-net was placed around the anastomosis in one half of the animals. The results demonstrated that delayed healing of the anastomoses as well as spontaneous suture line disruptions followed by peritonitis only occurred in those animals in which a PGA-net had been used. The evaluation of the bursting strength could confirm this finding of delayed consolidation. The bursting pressure therefore was significantly decreased in all animals in which a PGA-net had been applied, compared to the control group operated without PGA-net application. We therefore conclude that PGA-net application in colon anastomoses inhibits the consolidation of the suture line, probably because of a lack of contact to the peritoneal and omental tissue.

    Notes: Summary The value of isolated hepatocyte transplantation as a temporary support in acute hepatic failure remains controversial regarding the functional capacities of freshly isolated and transplanted hepatocytes. To evaluate the survival rate of intrasplenically transplanted liver cells and their response to a proliferation stimulus like partial hepatectomy,3H-thymidine incorporation into hepatic and auxiliary transplanted hepatocyte DNA was determined. The survival rate of intrasplenically transplanted hepatocytes was evaluated by analyses of m-albumin-RNA within the splenic tissue and compared to the morphological findings. The histological results show a marked decrease (〉 × 100) of intrasplenically transplanted hepatocytes within 1 week after injection. The amount of surviving cells then remained constant for 3 months without any signs of proliferation. After partial hepatectomy a stimulation of hepatic regeneration was observed in the remaining liver tissue but not in auxillary transplanted hepatocytes. M-albumin-RNA determination of auxiliary transplanted hepatocytes revealed a decrease of m-albumin-RNA concentration of 〉 100 times within 24 h after transplantation indicating early cell necrosis of the transplanted cells. Since intrasplenically transplanted hepatocytes underwent an early cell necrosis without any evidence for a directly postoperatively inducible cell proliferation, it is concluded that a sufficient metabolic support in acute hepatic failure cannot be taken into consideration.
