    Description / Table of Contents: Summary The effect of prior hypertension on the in vitro incorporation of [1-14C]-oleate into phospholipid and cholesterol ester in aortae from cholesterolfed and normal fed rabbits was studied. Incorporation of [1-14C]-oleate into phospholipid was not increased in aortae from either hypertensive normal-fed or hypertensive cholesterol-fed rabbits when compared to the appropriate normotensive controls. In the normal-fed rabbits, incorporation of [1-14C]-oleate into cholesterol ester was increased by hypertension in all aortic regions. In cholesterol-fed rabbits cholesterol esterification was found to be proportional to the intimal cholesterol concentration, irrespective of the prior blood pressure or the particular aortic region studied. It is concluded that the increased lipid synthesis in atherosclerotic vessels from hypertensive rabbits is a consequence of the increased lipid accumulation produced by hypertension and not the result of hypertension directly stimulating arterial wall metabolism.
    Notes: Zusammenfassung Der Einfluß von früherem arteriellen Hochdruck auf den in-vitro Einbau (1-14C)-Oleat in Phospholipide und Cholesterin-Ester in Aorten von Cholesterin-gefütterten und normal-gefütterten Kaninchen wurde untersucht. Im Vergleich zu den respektiven normotensiven Kontroll-Tieren stieg der Einbau (1-14C)-Oleat in Phospholipide in Aorten von hypertensiv normal-gefütterten und hypertensiv Cholesterin-gefütterten Kaninchen nicht an. In normal-gefütterten Kaninchen wurde der Einbau der (1-14C)-Oleat in allen Gegenden der Aorta mit Hochdruck vermehrt. Bei Cholesterin-gefütterten Kaninchen wurde die Cholesterin-Ester-Synthese der jeweiligen intimalen Cholesterin-Konzentration angemessen, unabhängig von welcher Aorten-Gegend untersucht wurde oder von der Höhe des früheren Blutdrucks. Daraus folgert, daß die gesteigerte Lipid-Synthese in atherosklerotischen Gefäßen hypertensiver Kaninchen eine Folge gesteigerter Lipidanhäufung ist, und nicht auf einen vom Hochdruck direkt vermehrten Arterienwandstoffwechsel zurückzuführen ist.

    Notes: 1. The effects of acute unilateral renal denervation were examined in 17 anaesthetized rats. Renal haemodynamic changes were monitored using standard clearance techniques. Lithium clearance was used to assess fractional proximal sodium and water reabsorption.2. Denervation resulted in ipsilateral renal vasodilatation with marked natriuresis and diuresis, a small increase (15%, P 〈 0.05) in glomerular filtration rate (GFR) and a consequent reduction in filtration fraction. Fractional lithium reabsorption decreased (67.3 ± 2.9% to 54.5 ± 4.0%, P 〈 0.01) and absolute proximal reabsorption did not change, indicating impairment of proximal glomerulotubular balance (GTB). No similar changes in haemodynamic or transport parameters were observed in the contralateral, innervated kidney, although vascular resistance increased.3. In 9 experiments following denervation of the left kidney, systemic low dose infusion (10 ng/min) of atrial natriuretic factor (ANF) resulted in a fall in mean arterial blood pressure from 116 ± 3 mmHg to 107 + 3 mmHg (P 〈 0.05). In the denervated kidney ANF increased urine flow rate and sodium excretion to rates above those established following denervation alone. However, in the right kidney, despite the increased filtered load (35%, P 〈 0.01), the natriuretic and diuretic responses to ANF were abolished.4. In the denervated kidney, ANF further reduced the fractional reabsorption of lithium from 53.6 ± 2.3% to 45.6 ± 3.8% (P 〈 0.05). GFR increased by 32% (a total of 49% higher than during pretreatment) but absolute proximal reabsorption (APR) did not change. However, in the right, innervated kidney ANF infusion produced a 35% increase in GFR accompanied by a 53% rise in APR.5. It is concluded that the natriuresis induced by unilateral denervation is due predominantly to impaired proximal GTB. The natriuretic action of ANF was associated with further impairment of proximal GTB, not dependent upon decreasing activity of renal sympathetic nerves, but was abolished when filtration fraction and renal sympathetic tone were elevated.

The concentrations of aldosterone, corticosterone, cortisol and renin substrate in plasma and the plasma renin activity were determined in five non-pregnant and five pregnant rhesus monkeys.2. The mean aldosterone concentrations are 25.9 ng/100 ml in the non-pregnant and 31.4 ng/100 ml in the pregnant monkey: the difference is not significant (P〉0.5).3. The mean corticosterone concentrations are 1.11 and 1.84 μg/100 ml respectively: the difference is not significant (P〉0.1).4. The mean cortisol concentrations are 21.3 and 28.2 μg/100 ml respectively: the difference is significant (P〈0.05).5. Pregnancy does not result in increases in either plasma renin activity, mean values being 4.96 and 3.72 ng/ml per h, respectively, or renin substrate concentrations, the means being 1.09 and 0.82 μg/ml, respectively.6. The lack of elevation of these hormones is consistent with findings that concentrations of progesterone and oestrogen are only slightly increased in pregnancy in rhesus monkeys.

    Notes: An investigation has been made into the cellular source of renin in human chorion. Using cytogenetic techniques the cells producing renin in tissue cultures of chorion have been identified as fetal and not maternal in origin.Histochemical studies using the Bowie stain have shown the presence of cells containing Bowie-positive granules in the cytoplasm in the spongy layer of amnion and the reticular layer of chorion. These cells do not occur in the trophoblastic layer and morphologically have been shown to resemble the Hofbauer cell.Chorion cells producing renin during in vitro culture do not, however, display Bowie-positive cytoplasmic granules.

    Notes: Abstract This study investigated the relationships between resting whole blood viscosity (WBV), haemoglobin concentration (HGB), haematocrit (HCT), and performance in 25 highly-trained national squad rowers (11 women and 14 men). The WBV and HGB were measured at rest prior to a 2500 m simulated race on a Concept rowing ergometer when performance (P) was measured by average velocity. A group of 12 rowers were measured on just one occasion, another 11 were measured twice with an intervening 5 weeks of continued training and 2 were measured three times, the third test after another 4 weeks. Regression analyses making simultaneous use of both intra- and interindividual data indicated a significant inverse relationship between P and WBV (at both high and low shear rates), a relationship which was strengthened after statistically controlling for the effects of HGB, this effect being slightly more significant than HCT. A significant positive regression also emerged between P and HGB, but only after statistically controlling for the influence of WBV at high shear rate. Overall, stronger relationships were demonstrated in the male rowers compared with the female. These data, in the light of previous evidence that fitter people tend to have lower WBV, would indicate that blood rheology unrelated to HGB (or HCT) is related to performance in relatively homogeneous and already highly-trained athletes.
